Abstract

The invention relates to a halogenated salicylanilide selected from closantel, rafoxanide, oxyclozanide and niclosamide and derivatives thereof including salts, hydrates, esters and the like for use in the topical treatment or prevention of infections caused by Gram-positive bacteria such as Staphylococcus , in particular Staphylococcus aureus , and Streptococcus , in particular Streptococcus pyogenes . Gram positive bacteria treated with the halogenated salicylanilides exhibit a very low frequency of appearance of resistant mutants compared to commonly used topical antibiotics.

Claims (27)

1. A method of treating atopic dermatitis in a subject, the method comprising topically administering to the subject an effective amount of niclosamide or a pharmaceutically acceptable salt or hydrate thereof.

2. The method of claim 1 , wherein the treatment ameliorates symptoms of the atopic dermatitis caused by Gram-positive bacteria.

3. The method of claim 2 , wherein the Gram-positive bacteria are from the Staphylococcus genus or the Streptococcus genus.

4. The method of claim 2 , wherein the Gram-positive bacteria are selected from Staphylococcus aureus and Streptococcus pyogenes.

5. The method of claim 2 , wherein the Gram-positive bacteria are resistant to an antibiotic other than niclosamide.

6. The method of claim 2 , wherein the Gram-positive bacteria are resistant to an antibiotic selected from fusidic acid, mupirocin and retapamulin.

7. The method of claim 2 , wherein the Gram-positive bacteria are methicillin-resistant Staphylococcus aureus.

8. The method of claim 1 , wherein the niclosamide is administered topically to the subject in the form of a pharmaceutical formulation comprising the niclosamide, or a hydrate thereof.

9. The method of claim 1 , wherein the niclosamide is administered topically to the subject in the form of a pharmaceutical formulation comprising from 2 to 10% by weight of niclosamide.

10. The method of claim 1 , wherein the subject is a human.

11. The method of claim 1 , wherein the subject is not being treated for acne vulgaris with niclosamide.

12. A method of treating atopic dermatitis colonized by Gram-positive bacteria in a subject, the method comprising topically administering to the subject an effective amount of niclosamide or a pharmaceutically acceptable salt or hydrate thereof.

13. The method of claim 12 , wherein the Gram-positive bacteria are from the Staphylococcus genus or the Streptococcus genus.

14. The method of claim 12 , wherein the Gram-positive bacteria are selected from Staphylococcus aureus and Streptococcus pyogenes.

15. The method of claim 12 , wherein the Gram-positive bacteria are resistant to an antibiotic other than niclosamide.

16. The method of claim 12 , wherein the Gram-positive bacteria are resistant to an antibiotic selected from fusidic acid, mupirocin and retapamulin.

17. The method of claim 12 , wherein the Gram-positive bacteria are methicillin-resistant Staphylococcus aureus.

18. The method of claim 12 , wherein the niclosamide is administered topically to the subject in the form of a pharmaceutical formulation comprising the niclosamide in free form, or a hydrate thereof.

19. The method of claim 12 , wherein the niclosamide is administered topically to the subject in the form of a pharmaceutical formulation comprising from 2 to 10% by weight of niclosamide.

20. The method of claim 12 , wherein the subject is a human.

21. The method of claim 12 , wherein the subject is not being treated for acne vulgaris with niclosamide.

22. A method of treating or eliminating bacterial colonization by Gram positive bacteria in a subject affected by atopic dermatitis, the method comprising topically administering to the subject an effective amount of niclosamide or a pharmaceutically acceptable salt or hydrate thereof.

23. The method of claim 22 , wherein the Gram-positive bacteria are from the Staphylococcus genus or the Streptococcus genus.

24. The method of claim 22 , wherein the Gram-positive bacteria are resistant to an antibiotic other than niclosamide.

25. The method of claim 22 , wherein the Gram-positive bacteria are methicillin-resistant Staphylococcus aureus.

26. The method of claim 22 , wherein the subject is not being treated for acne vulgaris with niclosamide.

27. The method of claim 22 , wherein the Gram-positive bacteria are resistant to an antibiotic selected from fusidic acid, mupirocin and retapamulin.
