Methods of using chimeric receptors to identify autoimmune disease
The present invention provides methods and compositions useful in the diagnosis and management of autoimmune diseases. In particular, the present invention provides improved methods and compositions for the diagnosis and management of Graves' disease. The methods of the present invention not only avoids the need for radioactivity and are much simpler, economical, and rapid than methods traditionally used for the diagnosis of Graves' disease, but also improve upon the sensitivity and detection abilities of previous luciferase-based autoantibody detection assays.
1. A method, comprising:
i) preparing a recombinant plasmid vector, wherein the recombinant plasmid vector comprises:
a) a chimeric thyrotropin stimulating hormone (TSH) receptor gene, and
b) a reporter gene;
wherein, the chimeric TSH receptor gene encodes a polypeptide having an N-terminal portion encoded by nucleotides 45-2234 of SEQ ID NO: 3.
2. The method of claim 1 , wherein the recombinant plasmid vector further comprises:
c) a promoter, wherein the promoter facilitates an initiation of transcription of the chimeric TSH receptor gene and the reporter gene.
3. The method of claim 2 , wherein the reporter gene is selected from the group consisting of a bacterial β-galactosidase gene, a luciferase gene, and a β-glucuronidase gene.
4. The method of claim 2 , wherein the reporter gene is a firefly luciferase gene.
5. The method of claim 2 , wherein the reporter gene allows detection of cAMP.
6. The method of claim 2 , wherein the recombinant plasmid vector further comprises:
d) an antimicrobial resistance gene.
7. The method of claim 6 , wherein the antimicrobial resistance gene is selected from the group consisting of a chloramphenicol resistance gene, a neomycin resistance gene, a G418 resistance gene, a hygromycin resistance gene, and a mycophenolic acid resistance gene.
8. The method of claim 7 , wherein the antimicrobial resistance gene is a neomycin resistance gene.
9. The method of claim 6 , wherein the preparing comprises ligating the chimeric TSH receptor gene to the reporter gene, the promoter, and the antimicrobial resistance gene.
10. The method of claim 9 , wherein the promoter is a glycoprotein hormone alpha subunit promoter.
11. The method of claim 10 , further comprising:
ii) transfecting a one or more cells from a cell line with the recombinant plasmid vector to provide a recombinant cell line.
12. The method of claim 11 , wherein the cell line is a Chinese Hamster Ovary (CHO) line or a human Rhabdomyosarcoma (RD) line.
