Abstract

Engineered human tissue seed constructs are provided that are suitable for implantation in subjects. Methods of making and using the engineered tissue seed constructs are provided.

Claims (41)

1. An ex vivo engineered tissue seed suitable for implantation in a host, comprising

(a) a patterned biomaterial comprising one or more endothelial cell cords comprising primary human endothelial cells embedded in a biocompatible hydrogel scaffold; and

(b) hepatocyte aggregates formed by coculturing a first population of cells comprising primary human hepatocytes and a second population of cells comprising human fibroblast cells,

wherein the hepatocyte aggregates are combined with the patterned biomaterial, and wherein the hepatocyte aggregates are not in direct contact with the endothelial cell cords, thereby forming an engineered tissue seed suitable for implantation in a host.

2. The engineered tissue seed of claim 1 , which expands in response to a regeneration cue, following implantation in the host.

3. The engineered tissue seed of claim 1 , wherein liver parenchyma in the engineered tissue seed expands in response to regeneration cues, following implantation in the host.

4. The engineered tissue seed of claim 1 , wherein vasculature in the engineered tissue seed expands in response to regeneration cues, following implantation in the host.

5. The engineered tissue seed of claim 1 , which expands about 50-fold in situ, following implantation in the host.

6. The engineered tissue seed of claim 1 , which secretes a hepatocyte blood factor selected from the group consisting of albumin, transferrin, alpha-1-antitrypsin, and fibronectin, or a combination thereof, following implantation in the host.

7. The engineered tissue seed of claim 1 , which expands to form densely packed hepatocytes, form duct-like structures resembling bile ducts, form biliary epithelial-like cells, form blood vessels, or any combination thereof, following implantation in the host.

8. The engineered tissue seed of claim 1 , which expands in response to paracrine signaling between cells in the engineered tissue seed, following implantation in the host.

9. The engineered tissue seed of claim 1 , which expands in response to a regeneration cue from the host, wherein the regeneration cue is due to an injury, disease or infection in the host, or occurs during native development of the host.

10. The engineered tissue seed of claim 1 , wherein the regeneration cue is a growth factor or cytokine.

11. The engineered tissue seed of claim 1 , further comprising a small molecule or growth factor which stimulates or enhances expansion of the engineered tissue seed, following implantation in a host.

12. The engineered tissue seed of claim 1 , wherein the scaffold comprises one or more channels, wherein the channels are in parallel or non-parallel formation.

13. The engineered tissue seed of claim 12 , wherein the channels are organized with specific branch patterns selected from rectilinear grids, bifurcated trees, 2D and 3D organizations.

14. The engineered tissue seed of claim 1 , wherein the seed is about 10-30 mm in diameter.

15. The engineered tissue seed of claim 1 , wherein the endothelial cell cords consist of endothelial cells.

16. The engineered tissue seed of claim 1 , which expands in response to liver injury following implantation in the host.

17. An ex vivo engineered tissue seed suitable for implantation in a host, comprising

(a) a patterned biomaterial comprising a plurality of endothelial cell cords comprising primary human endothelial cells embedded in a biocompatible hydrogel scaffold, wherein the plurality of endothelial cell cords are organized in a repeating and parallel arrangement; and

(b) hepatocyte aggregates formed by coculturing a first population of cells comprising primary human hepatocytes and a second population of cells comprising human fibroblast cells,

wherein the hepatocyte aggregates are combined with the patterned biomaterial, thereby forming an engineered tissue seed suitable for implantation in a host.

18. The engineered tissue seed of claim 17 , which expands about 50-fold in situ, following implantation in the host.

19. The engineered tissue seed of claim 17 , which expands in response to liver injury following implantation in the host.

20. The engineered tissue seed of claim 17 , which expands to form densely packed hepatocytes, form duct-like structures resembling bile ducts, form biliary epithelial-like cells, form blood vessels, or any combination thereof, following implantation in the host.

21. The engineered tissue seed of claim 17 , wherein the endothelial cell cords consist of endothelial cells.

22. The engineered tissue seed of claim 17 , wherein the hepatocyte aggregates are not in direct contact with the endothelial cell cords.

23. The engineered tissue seed of claim 17 , wherein the seed is about 10-30 mm in diameter.

24. The engineered tissue seed of claim 17 , wherein the hepatocyte aggregates are micropatterned between the endothelial cell cords.

25. The engineered tissue seed of claim 24 , wherein the hepatocyte aggregates are not in direct contact with the endothelial cell cords.

26. An ex vivo engineered tissue seed suitable for implantation in a host, the engineered tissue seed comprising:

(a) one or more human endothelial cell cords; and

(b) one or more hepatocyte aggregates, wherein each hepatocyte aggregate comprises a first population of cells comprising primary human hepatocytes and a second population of cells comprising human fibroblast cells,

wherein the one or more human endothelial cell cords and the one or more hepatocyte aggregates are each in a biocompatible scaffold, and wherein the one or more hepatocyte aggregates are not in direct contact with the one or more human endothelial cell cords.

27. An ex vivo engineered tissue seed suitable for implantation in a host, the engineered tissue seed comprising:

(a) a plurality of human endothelial cell cords organized in a repeating and parallel arrangement; and

(b) one or more hepatocyte aggregates, wherein each hepatocyte aggregate comprises a first population of cells comprising primary human hepatocytes and a second population of cells comprising human fibroblast cells,

wherein the plurality of human endothelial cell cords and the one or more hepatocyte aggregates are each in a biocompatible scaffold.

28. The engineered tissue seed of claim 27 , wherein the hepatocyte aggregates are micropatterned between the endothelial cell cords.

29. The engineered tissue seed of claim 28 , wherein the hepatocyte aggregates are not in direct contact with the endothelial cell cords.
