Abstract

Provided herein are cyclic nucleotide phosphodiesterase inhibitors, and pharmaceutical compositions thereof, useful for the treatment of, for example, central nervous system and metabolic diseases and disorder.

Claims (63)

1. A compound of formula (I):

A-L-B   (I)

or a pharmaceutically acceptable salt or stereoisomer thereof,

wherein:

L is —C(R 6 ) 2 —O—;

A is

B is

A 1 -A 2 is N═CR 8 ;

A 3 is N;

A 4 is NR 5 , O, or S;

A 5 is CR 5 or N;

A 6 is N or CR 5 ;

R 1 and R 2 , together with the carbon atoms to which they are attached, form a 1,2-phenylene ring, optionally substituted with one or more R 11 , wherein R 11 is other than ═O;

R 3 is (i) hydrogen, cyano, or halogen; or (ii) (C 1 -C 10 )alkyl, (C 2 -C 10 )alkenyl, (C 1 -C 10 )alkoxy, (C 1 -C 10 )aminoalkyl, (C 1 -C 10 )heteroalkyl, (C 3 -C 10 )cycloalkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, (3 to 12 membered)heterocyclyl, hydroxy, amino, imino, amido, or thiol, each of which is optionally substituted with one or more R 11 ;

R 4 is (i) hydrogen, cyano, or halogen; or (ii) (C 1 -C 10 )alkyl, (C 2 -C 10 )alkenyl, (C 1 -C 10 )alkoxy, (C 1 -C 10 )aminoalkyl, (C 1 -C 10 )heteroalkyl, (C 3 -C 10 )cycloalkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, (3 to 12 membered)heterocyclyl, hydroxy, amino, imino, amido, or thiol, each of which is optionally substituted with one or more R 11 ;

R 5 is (i) hydrogen, cyano, or halogen; or (ii) (C 1 -C 10 )alkyl, (C 2 -C 10 )alkenyl, (C 1 -C 10 )alkoxy, (C 1 -C 10 )aminoalkyl, (C 1 -C 10 )heteroalkyl, (C 3 -C 10 )cycloalkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, (3 to 12 membered)heterocyclyl, hydroxy, amino, imino, amido, or thiol, each of which is optionally substituted with one or more R 11 ; or

R 3 and R 4 , together with the atoms to which they are attached, form a ring, optionally substituted with one or more R 11 ; or

R 4 and R 5 , together with the atoms to which they are attached, form a ring, optionally substituted with one or more R 11 ;

R 6 is hydrogen;

each occurrence of R 8 is independently (i) hydrogen, cyano, or halogen; or (ii) (C 1 -C 10 )alkyl, (C 2 -C 10 )alkenyl, (C 1 -C 10 )alkoxy, (C 1 -C 10 )aminoalkyl, (C 1 -C 10 )heteroalkyl, (C 3 -C 10 )cycloalkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, (3 to 12 membered) heterocyclyl, hydroxy, amino, imino, amido, or thiol, each of which is optionally substituted with one or more R 11 ;

each occurrence of R 11 is independently (i) halogen, cyano, ═O, OR 13 , NR 13 R 14 , N(R 13 )C(O)R 14 , C(O)R 13 , C(O)NR 13 R 14 , C(O)OR 13 , OC(O)R 13 , SR 13 , S(O)R 13 , S(O) 2 R 13 , or S(O) 2 NR 13 R 14 ; or (ii) (C 1 -C 10 )alkyl, (C 1 -C 10 )heteroalkyl, (C 3 -C 10 )cycloalkyl, (C 7 -C 12 )aralkyl, (C 3 -C 12 )heteroaralkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, or (3 to 12 membered)heterocyclyl, each of which is optionally substituted with one or more R 12 ;

each occurrence of R 12 is independently (i) halogen, cyano, ═O, OR 13 , NR 13 R 14 , NR 13 C(O)R 14 , C(O)R 13 , C(O)NR 13 R 14 , C(O)OR 13 , OC(O)R 13 , SR 13 , S(O)R 13 , S(O) 2 R 13 , or S(O) 2 NR 13 R 14 ; or (ii) (C 1 -C 6 )alkyl or (C 3 -C 6 )cycloalkyl, each of which is optionally substituted with one or more R 13 ;

each occurrence of R 13 and R 14 is independently hydrogen, (C 1 -C 6 )alkyl, (C 1 -C 6 )heteroalkyl, (C 3 -C 6 )cycloakyl, (C 7 -C 10 )aralkyl, (C 3 -C 12 )heteroaralkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, or (3 to 12 membered)heterocyclyl; or

R 13 and R 14 , together with the atom(s) to which they are attached, may form a 3 to 10 membered ring;

R 19 is (i) hydrogen, cyano, or halogen; or (ii) (C 1 -C 10 )alkyl, (C 2 -C 10 )alkenyl, (C 1 -C 10 )alkoxy, (C 1 -C 10 )aminoalkyl, (C 1 -C 10 )heteroalkyl, (C 3 -C 10 )cycloalkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, (3 to 12 membered)heterocyclyl, hydroxy, amino, imino, amido, or thiol, each of which is optionally substituted with one or more R 11 ; and

R 20 is (i) hydrogen; or (ii) (C 1 -C 10 )alkyl, (C 2 -C 10 )alkenyl, (C 1 -C 10 )heteroalkyl, (C 3 -C 10 )cycloalkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, or (3 to 12 membered)heterocyclyl, each of which is optionally substituted with one or more R 11 .

2. The compound of claim 1 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein:

each occurrence of R 15 is independently (i) halogen, cyano, ═O, OR 13 , NR 13 R 14 , N(R 13 )C(O)R 14 , C(O)R 13 , C(O)NR 13 R 14 , C(O)OR 13 , OC(O)R 13 , SR 13 , S(O)R 13 , S(O) 2 R 13 , or S(O) 2 NR 13 R 14 ; or (ii) (C 1 -C 10 )alkyl, (C 1 -C 10 )heteroalkyl, (C 3 -C 10 )cycloalkyl, (C 7 -C 12 )aralkyl, (C 3 -C 12 )heteroaralkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, or (3 to 12 membered)heterocyclyl, each optionally substituted with one or more R 12 ; and

m is 0, 1, 2, 3 or 4.

3. The compound of claim 2 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein R 8 is hydrogen or CH 3 .

4. The compound of claim 1 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein:

A 5 is CR 5 ; and

R 3 and R 4 , together with the atoms to which they are attached, form a ring, optionally substituted with one or more R 11 .

5. The compound of claim 4 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein R 5 is hydrogen or CH 3 .

6. The compound of claim 1 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein:

7. The compound of claim 1 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein the compound has formula (I-A):

8. The compound of claim 7 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein:

A 4 is NR 5 ; and

R 5 is (i) hydrogen; or (ii) (C 1 -C 10 )alkyl, (C 2 -C 10 )alkenyl, (C 1 -C 10 )heteroalkyl, (C 3 -C 10 )cycloalkyl, (6 to 10 membered)aryl, (5 to 10 membered)heteroaryl, or (3 to 12 membered)heterocyclyl, each of which is optionally substituted with one or more R 11 ; or

R 4 and R 5 , together with the atoms to which they are attached, form a ring, optionally substituted with one or more R 11 .

9. The compound of claim 8 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein R 5 is hydrogen or CH 3 .

10. The compound of claim 9 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein:

k is 0, 1, 2, 3, or 4;

J is O, S, NH, or CH 2 ; and

each occurrence of G is independently CH or N.

11. The compound of claim 10 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein:

12. The compound of claim 10 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein R 3 is (i) hydrogen; or (ii) (C 1 -C 10 )alkyl, (6 to 10 membered)aryl, or (5 to 10 membered)heteroaryl, each of which is optionally substituted with one or more R 11 .

13. The compound of claim 10 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein R 4 is (i) hydrogen; or (ii) (C 1 -C 10 )alkyl, (6 to 10 membered)aryl, or (5 to 10 membered)heteroaryl, each of which is optionally substituted with one or more R 11 .

14. The compound of claim 1 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein R 5 is (i) hydrogen; or (ii) (C 1 -C 10 )alkyl, (6 to 10 membered)aryl, or (5 to 10 membered)heteroaryl, each of which is optionally substituted with one or more R 11 .

15. The compound of claim 1 , or a pharmaceutically acceptable salt or stereoisomer thereof, wherein each occurrence of R 11 is independently (C 1 -C 10 )alkyl, (6 to 10 membered)aryl, or (5 to 10 membered)heteroaryl, each of which is optionally substituted with one or more R 12 .

16. The compound of claim 1 , wherein the compound is:

or a pharmaceutically acceptable salt or stereoisomer thereof.

17. A pharmaceutical composition comprising a compound of claim 1 , or a pharmaceutically acceptable salt or stereoisomer thereof, and at least one pharmaceutically acceptable excipient, diluent or carrier.

18. The pharmaceutical composition of claim 17 , wherein the pharmaceutical composition further comprises one or more additional active agents.

19. A method for inhibiting phosphodiesterase-10A activity in a subject, comprising administering to a subject in need thereof a therapeutically or prophylactically effective amount of a compound of claim 1 , or a pharmaceutically acceptable salt or stereoisomer thereof.

20. The method of claim 19 , wherein the subject suffers from a neurological disorder, a schizophrenia-related disorder, a disease having a psychosis component, a psychotic disorder, a behavior disorder, a neurodegenerative disease, a mood disorder, a manic disorder, a movement disorder, a sleep disorder, an addiction or an eating disorder.

21. The method of claim 19 , wherein the subject suffers from a central nervous system disorder.

22. The method of claim 19 , wherein the subject suffers from a metabolic disorder.

23. The method of claim 19 , wherein the subject suffers from schizophrenia, schizophrenia spectrum disorder, schizoaffoctive disorder, schizophreniform disorder, paraphrenia, paranoid personality disorder, schizoid personality disorder, schizotypal personality disorder, delusional disorder, psychosis, psychoaffective disorder, aggression, delirium, Tourette's syndrome, convulsion, seizure, agitation, posttraumatic stress disorder, Huntington's disease, Alzheimer's disease, Parkinson's disease, dyskinesia, dementia, bipolar disorder, anxiety, depression, major depressive disorder, dysthymia, affective disorder, obsessive-compulsive disorder, attention deficit disorder, attention deficit hyperactivity disorder, vertigo, pain, sensitization accompanying neuropathic pain, fibromyalgia, migraine, cognitive impairment, cognitive deficit in Alzheimer's disease, cognitive deficit in Parkinson's disease, restless leg syndrome, multiple sclerosis, substance abuse, substance dependency, autism, obesity, undesirable weight retention, undesirable weight gain, metabolic syndrome, diabetes, impaired glucose tolerance or hyperglycemia.

24. The method of claim 19 , wherein the subject is a human.
