Abstract

There are provided inter alia multisubstituted aromatic compounds useful for the inhibition of kallikrein, which compounds include substituted pyrazolyl or substituted triazolyl. There are additionally provided pharmaceutical compositions. There are additionally provided methods of treating and preventing certain diseases or disorders, which disease or disorder is amenable to treatment or prevention by the inhibition of kallikrein.

Claims (51)

1. A method for treating a disease or disorder, comprising administering a kallikrein-inhibiting compound to a subject in need thereof in an amount effective to treat said disease or disorder, wherein the kallikrein-inhibiting compound has the following formula:

or pharmaceutically acceptable salt thereof;

wherein

L 1 is —NR 5 —;

L 2 is a bond;

L 3 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

L 4 is a bond, substituted or unsubstituted alkylene, substituted or unsubstituted heteroalkylene, —S—, —SO—, —SO 2 —, —O—, —NHSO 2 —, or —NR 5 —;

R 1 is substituted alkyl having one or more substituent groups, wherein any substituent group for said R 1 substituted alkyl is selected from the group consisting of —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, and substituted or unsubstituted heteroaryl;

R 2 is substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted heterocycloalkenyl, substituted or unsubstituted aryl, substituted or unsubstituted fused ring aryl, or substituted or unsubstituted heteroaryl, wherein the substituted alkyl, substituted cycloalkyl, substituted cycloalkenyl, substituted heterocycloalkyl, substituted heterocycloalkenyl, substituted aryl, substituted fused ring aryl, or substituted heteroaryl has one or more substituent group selected from the group consisting of oxo, —OH, —NH 2 , —SH, —CN, —CF 3 , —NO 2 , -halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, and substituted or unsubstituted heterocycloalkyl;

R 3 is substituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted cycloalkenyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein when R 3 is substituted heteroaryl, any substituent group of said R 3 substituted heteroaryl is selected from the group consisting of NH 2 , —SH, —CN, —CF 3 , —NO 2 , halogen, —COOH, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, and substituted or unsubstituted heteroaryl;

R 4 is hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsub stituted heterocycloalkyl, substituted aryl, or substituted or unsubstituted heteroaryl; and

R 5 is hydrogen, or substituted or unsubstituted alkyl,

wherein the disease or disorder is an ophthalmic disease, a dermatological condition, or hereditary angioedema.

2. The method according to claim 1 , wherein said compound is selected from the group consisting of:

Compound 19, 1-(5-{[(4-fluorophenyl)methyl]amino}-3-phenyl-1H-pyrazol-1-yl)-2,2-dimethylpropan-1-one;

Compound 20, 1-(5-{[(4-fluorophenyl)methyl]amino}-3-(pyridin-2-yl)-1H-pyrazol-1-yl)-2,2-dimethylpropan-1-one;

Compound 266, 1-(5-{[(4-fluorophenyl)methyl]amino}-3-(pyridazin-3-yl)-1H-pyrazol-1-yl)-2,2-dimethylpropan-1-one;

Compound 267, 1-(5-{[(4-fluorophenyl)methyl]amino-3-(pyrimidin-4-yl)-1H-pyrazol-1-yl)-2,2-dimethylpropan-1-one;

Compound 268, 1-(5-{[(4-fluorophenyl)methyl]amino}-3-phenyl-1H-pyrazol-1-yl)propan-1-one;

Compound 269, 1-(5-{[(5-chlorothiophen-2-yl)methyl]amino}-3-(3-fluoropyridin-2-yl)-1H-pyrazol-1-yl)-2,2-dimethylpropan-1-one;

Compound 291, N-[(5-chlorothiophen-2-yl)methyl]-3-(3-fluoropyridin-2-yl)-1-[(furan-3-yl)carbonyl]-1H-pyrazol-5-amine;

Compound 292, N-[(5-chlorothiophen-2-yl)methyl]-3-(pyridazin-3-yl)-1-[(thiophen-3-yl)carbonyl]-1H-pyrazol-5-amine; and

Compound 293, N-[(5-chlorothiophen-2-yl)methyl]-3-(pyridin-2-yl)-1-[(thiophen-3-yl)carbonyl]-1H-pyrazol-5-amine.

3. The method according to claim 1 , wherein said disease or disorder comprises an ophthalmic disease.

4. The method of claim 3 , wherein said ophthalmic disease comprises at least one of diabetic macular edema, age-related macular degeneration, and diabetic retinopathy.

5. The method according to claim 3 , wherein said compound or pharmaceutical composition is administered orally, in the form of an ophthalmic composition applied topically to the eye, or in the form of an ophthalmic composition via intravitreal injection.

6. The method according to claim 5 , wherein said ophthalmic composition is in the form of eye drops.

7. The method according to claim 3 , wherein said compound acts by reducing retinal vascular permeability.

8. The method according to claim 1 , wherein said disease or disorder comprises a dermatological condition.

9. The method according to claim 1 , wherein said disease or disorder comprises hereditary angioedema.

10. The method according to claim 9 , wherein said disease or disorder is hereditary angioedema and said compound is administered orally.

11. The method according to claim 8 , wherein said dermatological condition comprises at least one of atopic dermatitis, psoriasis, and Netherton Syndrome.

12. The method according to claim 1 , wherein said compound acts by inhibiting kallikrein.

13. The method according to claim 12 , wherein said compound acts by inhibiting tissue kallikrein.

14. The method according to claim 12 , wherein said compound acts by inhibiting plasma kallikrein.
