Abstract

The invention provides methods and compositions for use of desmopressin in combination with a beta-3-adrenergic receptor agonist. The methods and compositions are useful in the treatment of nocturia and other urinary frequency disorders.

Claims (22)

1. A method of inhibiting the urge to urinate in an adult human subject over an interval of about two hours to no more than about eight hours, comprising administering to the adult human subject in need thereof a pharmaceutical composition comprising pharmaceutically active agents, the pharmaceutically active agents consisting of an effective, low dose amount of desmopressin and a beta-3-adrenergic receptor agonist so that both exert physiological activity during an overlapping time period, wherein the beta-3-adrenergic receptor agonist is administered at a daily dosage of about 1 mg to about 20 mg.

2. The method of claim 1 , wherein the method inhibits the urge to urinate in a human subject over an interval of about 4 hours to about 7 hours.

3. The method of claim 1 , wherein an antidiuretic effect is achieved over an interval of about two hours to no more than about six hours.

4. The method of claim 1 , wherein the administering achieves in the subject a blood plasma concentration of desmopres sin that does not exceed 15 pg/mL.

5. The method of claim 1 , wherein the administering achieves in the subject a blood plasma concentration of desmopressin in the range of about 0.5 pg/mL to about 5 pg/mL.

6. The method of claim 1 , wherein the pharmaceutical composition is administered transdermally or intradermally.

7. The method of claim 1 , wherein the pharmaceutical composition is administered intranasally.

8. The method of claim 1 , wherein the pharmaceutical composition is administered sublingually across the oral mucosa.

9. The method of claim 1 , wherein the beta-3-adrenergic receptor agonist is amibegron, fasobegron, mirabegron, ritobegron, solabegron, BRL-37344, CL-316,243, GS-332, KRP-204, L-796568, or a pharmaceutically acceptable salt thereof.

10. The method of claim 1 , wherein the beta-3-adrenergic receptor agonist is mirabegron.

11. The method of claim 1 , wherein the pharmaceutical composition further comprises a pharmaceutically acceptable carrier.

12. The method of claim 1 , wherein the beta-3-adrenergic receptor agonist is administered at a daily dosage ranging from 1 mg to 5 mg.

13. The method of claim 1 , wherein the beta-3-adrenergic receptor agonist is administered at a daily dosage ranging from 5 mg to 10 mg.

14. The method of claim 1 , wherein the beta-3-adrenergic receptor agonist is administered at a daily dosage ranging from 10 mg to 20 mg.

15. The method of claim 12 , wherein the beta-3-adrenergic receptor agonist is mirabegron.

16. The method of claim 13 , wherein the beta-3-adrenergic receptor agonist is mirabegron.

17. The method of claim 14 , wherein the beta-3-adrenergic receptor agonist is mirabegron.

18. The method of claim 1 , wherein the desmopressin is administered in an amount that delivers to the blood stream a concentration of desmopres sin no more than 2 ng/kg.

19. The method of claim 12 , wherein the desmopressin is administered in an amount that delivers to the blood stream a concentration of desmopres sin no more than 2 ng/kg.

20. The method of claim 13 , wherein the desmopres sin is administered in an amount that delivers to the blood stream a concentration of desmopres sin no more than 2 ng/kg.

21. The method of claim 14 , wherein the desmopres sin is administered in an amount that delivers to the blood stream a concentration of desmopres sin no more than 2 ng/kg.

22. The method of claim 1 , wherein the adult human subject suffers from adult primary nocturnal enuresis, overactive bladder, incontinence, nocturia, or urinary urgency or frequency during waking hours.
