Abstract

Compositions and methods, including vaccines and pharmaceutical compositions for inducing or enhancing an immune response are disclosed based on the discovery of useful immunological adjuvant properties in a synthetic, glycopyranosyl lipid adjuvant (GLA) that is provided in substantially homogeneous form. Chemically defined, synthetic GLA offers a consistent vaccine component from lot to lot without the fluctuations in contaminants or activity that compromise natural-product adjuvants. Also provided are vaccines and pharmaceutical compositions that include GLA and one or more of an antigen, a Toll-like receptor (TLR) agonist, a co-adjuvant and a carrier such as a pharmaceutical carrier.

Claims (34)

1. A method of eliciting or enhancing a desired antigen-specific immune response in a subject, the method comprising administering to the subject a pharmaceutical composition that comprises (a) one or more antigens, or at least one nucleic acid sequence encoding a polypeptide antigen; (b) a glucopyranosyl lipid adjuvant (GLA) having the formula:

where: R1, R3, R5 and R6 are C11-C20 alkyl; and R2 and R4 are C12-C20 alkyl; or a pharmaceutically acceptable salt thereof; and (c) at least one of a surfactant, biodegradable oil, pharmaceutically acceptable carrier, excipient and diluent; thereby eliciting or enhancing a desired antigen-specific immune response.

2. A method for stimulating an immune response in a subject comprising administering a pharmaceutical composition comprising: (a) GLA having the formula:

where: R1, R3, R5 and R6 are C11-C20 alkyl; and R2 and R4 are C12-C20 alkyl; or a pharmaceutically acceptable salt thereof; and (b) at least one of a surfactant, biodegradable oil, pharmaceutically acceptable carrier, excipient and diluent; in an amount effective to stimulate an immune response.

3. The method of claim 1 or 2 wherein the pharmaceutical composition is in an emulsion form.

4. The method of claim 1 or 2 wherein the pharmaceutical composition is in an oil-in-water emulsion form.

5. The method of claim 4 wherein the pharmaceutical composition comprises squalene.

6. The method of claim 1 or 2 wherein the pharmaceutical composition further comprises at least one additional component selected from the group consisting of:

a toll-like receptor (TLR) agonist;

a saponin or saponin mimetic;

a co-adjuvant; and

a carrier that comprises at least one of an oil and an immunostimulatory complex.

7. The method of claim 6 wherein the saponin comprises QS21.

8. The method of claim 1 or 2 wherein the pharmaceutical composition comprises oil droplets having a size of less than 1 micron in diameter.

9. The method of claim 1 or 2 wherein the pharmaceutical composition is in a liquid form.

10. The method of claim 1 or 2 wherein the pharmaceutical composition is in a solution form or in a suspension form.

11. The method of claim 1 or 2 wherein the pharmaceutical composition comprises a stable aqueous suspension of particles of less than 0.2 μm diameter and at least one component selected from the group consisting of phospholipid, fatty acid, surfactant, detergent, saponin, and fluoridated lipid.

12. The method of claim 1 or 2 wherein the pharmaceutical composition comprises liposomes.

13. The method of claim 1 or 2 wherein the pharmaceutical composition comprises microspheres.

14. The method of claim 1 or 2 wherein the pharmaceutical composition is in an aerosol form.

15. The method of claim 1 or 2 wherein the pharmaceutical composition is in a lyophilizate form.

16. The method of claim 1 or 2 wherein the pharmaceutical composition comprises a preservative, an antibacterial agent, or an antioxidant, or any combination of the foregoing.

17. The method of claim 1 or 2 wherein the pharmaceutical composition comprises a dispersing agent and/or a suspending agent.

18. The method of claim 1 or 2 wherein the pharmaceutical composition comprises a buffer.

19. The method of claim 1 or 2 wherein the pharmaceutical composition comprises sterile water.

20. The method of claim 1 or 2 wherein the pharmaceutical composition comprises alpha tocopherol.

21. The method of claim 1 or 2 wherein the pharmaceutical composition comprises a synthetic mono or diglyceride.

22. The method of claim 1 or 2 wherein the pharmaceutical composition comprises an aluminum salt.

23. The method of claim 1 or 2 wherein the pharmaceutical composition comprises a recombinant viral vector.

24. The method of claim 1 or 2 wherein the pharmaceutical composition comprises at least one of petrolatum, lanolin, polyethylene glycol, beeswax, and mineral oil.

25. The method of claim 1 or 2 wherein the pharmaceutical composition is formulated for injection.

26. The method of claim 1 or 2 , wherein: R1, R3, R5 and R6 are undecyl; and R2 and R4 are tridecyl.

27. The method of claim 1 or 2 wherein the GLA is a pharmaceutically acceptable salt.

28. The method of claim 26 wherein the GLA is a pharmaceutically acceptable salt.
