Compositions and methods for treating gluten intolerance and disorders arising therefrom
The invention described herein relates to methods and compositions for treatment of gluten intolerance and related conditions (e.g., celiac disease and gluten sensitivity), or inhibition of inflammation and/or immune response in the intestine due to antigenic food peptides, by administration of a pharmaceutical composition comprising one or more Nepenthes enzymes.
1. A pharmaceutical composition comprising recombinant neprosin and a pharmaceutically acceptable buffering agent, wherein
the recombinant neprosin is a protein having at least 90% sequence identity to amino acids 25 to 380 of the amino acid sequence of SEQ ID NO: 1 and having prolyl endopeptidase activity, wherein the recombinant neprosin has a different glycosylation pattern than natural neprosin.
2. The pharmaceutical composition of claim 1 , wherein the amino acid sequence of the neprosin comprises an amino acid sequence having at least 90% sequence identity to the amino acid sequence of SEQ ID NO: 1 and having prolyl endopeptidase activity.

The pharmaceutical composition of claim 2 , further comprising at least one additional Nepenthes enzyme or variant thereof, wherein the at least one additional Nepenthes enzyme comprises the amino acid sequence of SEQ ID NO: 5, SEQ ID NO: 6, SEQ ID NO: 7, SEQ ID NO: 8, SEQ ID NO: 9, SEQ ID NO: 21, or SEQ ID NO: 22 or a variant thereof comprising at least 85% sequence identity to the amino acid sequence of SEQ ID NO: 5, SEQ ID NO: 6, SEQ ID NO: 7, SEQ ID NO: 8, SEQ ID NO: 9, SEQ ID NO: 21, or SEQ ID NO: 22 and having protease activity.
4. The pharmaceutical composition of claim 3 , wherein the at least one additional Nepenthes enzyme or variant thereof is nepenthesin I, nepenthesin II, and/or a variant thereof.
5. The pharmaceutical composition of claim 1 , wherein said composition is a sustained release formulation.
6. The pharmaceutical composition of claim 3 , wherein the neprosin and/or Nepenthes enzyme(s) comprises a propeptide.
7. A solid pharmaceutical formulation comprising the composition of claim 1 , wherein the neprosin is present in multiple layers such that the neprosin is released continuously while the formulation is present in the stomach.
8. The pharmaceutical composition of claim 1 , wherein the pharmaceutical composition is formulated for oral administration.
9. The pharmaceutical composition of claim 1 , wherein the neprosin comprises the amino acid sequence of SEQ ID NO: 1.
10. The pharmaceutical composition of claim 3 , wherein the at least one additional Nepenthes enzyme or variant thereof is nepenthesin II.
11. The pharmaceutical composition of claim 3 , wherein the at least one additional Nepenthes enzyme or variant thereof is nepenthesin I.
12. The pharmaceutical composition of claim 3 , wherein the ratio of neprosin to the at least one additional Nepenthes enzyme or variant thereof is between 100:1 and 1:100.
13. The pharmaceutical composition of claim 1 , wherein the pharmaceutical composition is in liquid form.
14. The pharmaceutical composition of claim 1 , wherein the buffering agent maintains the pharmaceutical composition at a pH of 6.5 or above when the pharmaceutical composition is in liquid form prior to administration to a subject.
15. The pharmaceutical composition of claim 14 , wherein the buffering agent maintains the composition at a pH of about 7.0 or above.
