Abstract

Provided are cells containing exogenous antigen and uses thereof.

Claims (29)

1. A method of inducing immune tolerance in a human subject suffering from immune activation in response to an antigen that mediates an autoimmune disease, the method comprising: administering to the human subject a pharmaceutical composition comprising an enucleated erythroid cell comprising at least 1,000 copies of an exogenous fusion polypeptide comprising the antigen, or an antigenic fragment thereof, fused to an intracellular domain of a transmembrane protein, wherein:

the antigen, or the antigenic fragment thereof, is localized on the intracellular side of the enucleated erythroid cell,

the exogenous fusion polypeptide does not comprise a ligand-binding domain,

the exogenous fusion polypeptide was expressed from a transgene introduced into a nucleated erythroid cell precursor used to generate the enucleated erythroid cell,

the enucleated erythroid cell is not a hypotonically dialysed cell; and

the pharmaceutical composition is administered in an amount and frequency sufficient to mediate the induction of the immune tolerance in the subject.

2. The method of claim 1 , wherein the enucleated erythroid cell comprises at least 10,000 copies of the antigen.

3. The method of claim 1 , wherein the antigen is selected from the group consisting of: insulin, proinsulin, preproinsulin, GAD65, IA-2, myelin oligodendrocyte glycoprotein, myelin basic protein, and proteolipid protein, or an antigenic fragment thereof.

4. The method of claim 1 , wherein the enucleated erythroid cell is a reticulocyte.

5. The method of claim 1 , wherein the pharmaceutical composition comprises a population of erythroid cells that is greater than 60% enucleated.

6. The method of claim 1 , wherein the enucleated erythroid cell is an erythrocyte.

7. The method of claim 1 , wherein the antigen is a viral capsid protein, or an antigenic fragment thereof.

8. The method of claim 1 , wherein the antigen is insulin or an antigenic fragment thereof.

9. The method of claim 1 , wherein the antigen is proinsulin or an antigenic fragment thereof.

10. The method of claim 1 , wherein the antigen is preproinsulin or an antigenic fragment thereof.

11. The method of claim 1 , wherein the antigen is GAD65 or an antigenic fragment thereof.

12. A method of inducing immune tolerance in a human subject suffering from immune activation in response to an antigen that mediates an autoimmune disease, the method comprising administering to the human subject a pharmaceutical composition comprising a population of enucleated erythroid cells that comprise an exogenous fusion polypeptide comprising the antigen, or an antigenic fragment thereof, fused to an intracellular domain of a transmembrane protein, wherein:

the antigen, or the antigenic fragment thereof, is localized on the intracellular side of the enucleated erythroid cells;

the exogenous fusion polypeptide does not comprise a ligand-binding domain;

the exogenous fusion polypeptide was expressed from a transgene introduced into a nucleated erythroid cell precursor used to generate the population of enucleated erythroid cells;

the enucleated erythroid cells are not hypotonically dialysed cells;

the pharmaceutical composition comprises a population of erythroid cells that is greater than 70% enucleated; and

the pharmaceutical composition is administered in an amount and frequency to mediate the induction of the immune tolerance in the subject.

13. The method of claim 12 , wherein the antigen is selected from the group consisting of: insulin, proinsulin, preproinsulin, GAD65, IA-2, myelin oligodendrocyte glycoprotein, myelin basic protein, and proteolipid protein, or an antigenic fragment thereof.

14. The method of claim 12 , wherein the antigen is a viral capsid protein, or an antigenic fragment thereof.

15. The method of claim 12 , wherein the antigen is insulin or an antigenic fragment thereof.

16. The method of claim 12 , wherein the antigen is proinsulin or an antigenic fragment thereof.

17. The method of claim 12 , wherein the antigen is preproinsulin or an antigenic fragment thereof.

18. The method of claim 12 , wherein the antigen is GAD65 or an antigenic fragment thereof.
