Abstract

Compositions comprising stem cells delivered into infarcted myocardium by endocardial injection engraft and differentiate into myocytes, endothelial cells, and vascular smooth muscle, and do so without the requirement for survival enhancing modification. These cells engraft whether injected acutely (days) or late (months) after myocardial infarction, and the efficiency of engraftment correlates with the functional recovery of the heart. The stem cells also recruit endogenous cardiac precursor cells, reconstitute myocardial stem cell niches, and enhance endogenous cell differentiation into myocytes.

Claims (17)

1. A pharmaceutical composition for treating heart disease comprising mesenchymal stem cells and c-kit + cardiac stem cells and a pharmaceutically acceptable carrier.

2. The pharmaceutical composition of claim 1 , further comprising cardiac tissue.

3. The pharmaceutical composition of claim 1 , wherein composition is a solution with a concentration of 10-40×10 6 mesenchymal stem cells/ml.

4. The pharmaceutical composition of claim of claim 1 , wherein the composition comprises 1 million mesenchymal stem cells.

5. The pharmaceutical composition of claim 1 , wherein the composition is in a unit dosage form of 20-50 μl.

6. The pharmaceutical composition of claim 1 , wherein the composition is suitable for injection.

7. The pharmaceutical composition of claim 1 , wherein the carrier comprises phosphate buffered saline (PBS).

8. The pharmaceutical composition of claim 1 , further comprising a cardiotropic factor.

9. The pharmaceutical composition of claim 1 , wherein the carrier comprises culture media.

10. The pharmaceutical composition of claim 9 , wherein the culture media is serum free.

11. The pharmaceutical composition of claim 1 , wherein the carrier comprises physiological saline.

12. The pharmaceutical composition of claim 1 , wherein the carrier comprises 5% dextrose in water.

13. The pharmaceutical composition of claim 1 , wherein the heart disease to be treated is cardiomyopathy, hypertrophic cardiomyopathy, dilated cardiomyopathy, myocarditis, myocardial infarction, or congestive heart failure.

14. The pharmaceutical composition of claim 1 , wherein the heart disease to be treated is cardiomyopathy.

15. The pharmaceutical composition of claim 1 , wherein the heart disease to be treated is myocarditis.

16. The pharmaceutical composition of claim 1 , wherein the heart disease to be treated is myocardial infarction.

17. The pharmaceutical composition of claim 1 , wherein the heart disease to be treated is congestive heart failure.
