Abstract

The present invention provides novel microfluidic substrates and methods that are useful for performing biological, chemical and diagnostic assays. The substrates can include a plurality of electrically addressable, channel bearing fluidic modules integrally arranged such that a continuous channel is provided for flow of immiscible fluids.

Claims (18)

1. A method for preparing nucleic acids for sequencing on a chip, the method comprising:

providing a plurality of nucleic acid fragments and a plurality of microbeads to a continuous microfluidic droplet formation chip in fluidic communication with a microfluidic PCR plate;

encapsulating, using the continuous microfluidic droplet formation chip, the plurality of nucleic acids fragments and the plurality of microbeads in droplets, wherein each droplet contains one of the plurality of nucleic acid fragments and one microbead, such that the droplets continuously flow into the microfluidic PCR plate as they are formed;

amplifying the nucleic acids fragments by flowing the droplets through a serpentine path in the microfluidic PCR plate configured to expose the droplets to thermocycling, wherein the amplifying also generates bead-bound nucleic acid fragments;

breaking the emulsion by centrifugation and recovering the bead-bound nucleic acid fragments; and

depositing the bead-bound nucleic acid fragments onto a chip for sequencing.

2. The method of claim 1 , wherein the nucleic acid comprises DNA or RNA.

3. The method of claim 1 , wherein the nucleic acid comprises an adaptor.

4. The method of claim 1 , wherein the droplets comprise primers.

5. The method of claim 4 , wherein the primers are coupled to the microbeads.

6. The method of claim 1 , further comprising shearing a nucleic acid to generate the nucleic acid fragments.

7. The method of claim 6 , wherein the nucleic acid comprises an entire genome.

8. The method of claim 1 , wherein the droplets are surrounded by an immiscible carrier fluid.

9. The method of claim 1 , wherein the amplification reaction comprises a polymerase chain reaction.

10. The method of claim 1 , wherein the sequencing chip comprises a plurality of wells.

11. The method of claim 10 , wherein each of the bead-bound nucleic acid fragments is deposited into a well.

12. The method of claim 1 , further comprising detecting droplets comprising bead-bound nucleic acid fragments.

13. The method of claim 12 , further comprising sorting the droplets comprising bead-bound nucleic acid fragments from droplets that do not comprise bead-bound nucleic acid fragments.
