Abstract

A system for fluorescence-based imaging of a target includes at least one excitation light source configured to emit a homogeneous field of excitation light and positioned to uniformly illuminate a target surface with the homogeneous field of excitation light during fluorescent imaging, a power source, and a portable housing configured to be held in a user's hand during imaging. The housing contains a lens, a filter, an image sensor, and a processor. The filter is configured to permit optical signals responsive to illumination of the target surface and having a wavelength corresponding to at least one of bacterial autofluorescence and tissue autofluorescence to pass through the filter to the image sensor. The at least one excitation light is adjacent to the housing so as to be positioned between the target surface and the image sensor during fluorescent imaging.

Claims (56)

1. A system for fluorescence-based imaging of a target, comprising:

at least one excitation light source configured to emit excitation light having a wavelength of 405 nm±10 nm and positioned to illuminate a target surface with the excitation light during fluorescent imaging;

a white light source positioned to illuminate the target surface with the white light during white light imaging; and

a portable housing configured to be held in a user's hand during imaging, the housing containing:

a lens,

a filter,

an image sensor, and

a processor,

wherein the lens is configured to direct optical signals responsive to illumination of the target surface with the excitation light toward the filter, the filter is configured to permit optical signals corresponding to autofluorescence of bacteria, fungus, yeast, and/or other microorganisms illuminated with the excitation light and having a wavelength between about 500 nm and about 550 nm and/or a wavelength greater than or equal to about 600 nm to pass through the filter to the image sensor, the image sensor is configured to detect the filtered optical signals, and the processor is configured to receive the detected optical signals and to output a representation of bacteria, fungus, yeast, and/or other microorganisms detected in and/or on the target surface based on the detected optical signals;

wherein the processor is further configured to spatially co-register one or more of absorbance data, reflectance data, endogenous fluorescence data, and exogenous fluorescence data, based on the detected optical signals, and output the co-registered data in the representation of the bacteria, fungus, yeast, and/or other microorganisms detected in and/or on the target surface; and

wherein the at least one excitation light source is adjacent to the housing so as to be positioned between the target surface and the image sensor during fluorescent imaging.

2. The system of claim 1 , wherein the at least one excitation light source is coupled to the housing.

3. The system of claim 1 , wherein the filter is further configured to block passage of optical signals having a wavelength of 405 nm±10 nm.

4. The system of claim 1 , wherein the filter is configured to permit optical signals having a wavelength between about 500 nm and about 550 nm and optical signals having a wavelength between about 600 nm and about 660 nm to pass through the filter to the image sensor.

5. The system of claim 1 , wherein the at least one excitation light source includes first and second LED light arrays, each array configured to emit light having a wavelength of 405 nm±10 nm.

6. The system of claim 1 , further comprising a display.

7. The system of claim 1 , wherein the processor is configured to wirelessly transmit and/or receive data.

8. The system of claim 1 , further comprising one or more of a power source, a heat sink, a thermal sensor, and a range finder.

9. The system of claim 1 , wherein the processor is configured to output data regarding detection of:

a location, a population, a quantity, a distribution, a colonization, a contamination, a critical colonization, an infection, and/or an extent of bacteria, fungus, yeast, and/or other microorganisms when present in the target.

The system of claim 1 , wherein the filter is further configured to permit optical signals responsive to illumination of the target surface with the excitation light and having a wavelength corresponding to tissue autofluorescence and tissue fluorescence to pass through the filter to the image sensor and, wherein, the processor is further configured to receive the detected optical signals corresponding to tissue autofluorescence and tissue fluorescence and to output a representation of tissue components detected in and/or on the target surface based on the detected optical signals corresponding to tissue autofluorescence and tissue fluorescence.

11. The system of claim 1 , wherein the at least one excitation light source includes a first blue/violet excitation light source configured to emit excitation light having a wavelength of 405 nm±10 nm and a second near infrared or infrared excitation light source.

12. A system for fluorescence-based imaging of a target, comprising:

first and second LED light arrays configured to emit violet or blue excitation light and positioned to illuminate a target surface with the excitation light during fluorescent imaging;

a white light source positioned to illuminate the target surface with the white light during white light imaging; and

a portable housing configured to be held in a user's hand during imaging, the housing containing:

a lens,

a filter,

an image sensor, and

a processor,

wherein the lens is configured to direct optical signals responsive to illumination of the target surface with the excitation light toward the filter, the filter is configured to permit optical signals corresponding to autofluorescence of bacteria, fungus, yeast, and/or other microorganisms illuminated with the excitation light and having a wavelength between about 500 nm and about 550 nm and a wavelength greater than or equal to about 600 nm to pass through the filter to the image sensor, the image sensor is configured to detect the filtered optical signals, and the processor is configured to receive the detected optical signals and to output a representation of bacteria, fungus, yeast, and/or other microorganisms in detected in and/or on the target surface to a display based on the detected optical signals;

wherein the processor is further configured to spatially co-register one or more of absorbance data, reflectance data, endogenous fluorescence data, and exogeneous fluorescence data, based on the detected optical signals, and output the co-registered data in the representation of the bacteria, fungus, yeast, and/or other microorganisms detected in and/or on the target surface; and

wherein the lens, the filter, and the image sensor are positioned to share a common axis passing through the housing, and wherein the first and second LED light arrays are positioned on opposite sides of the common axis.

13. The system of claim 12 , further comprising one or more of a power source a heat sink, a thermal sensor, and a range finder.

14. The system of claim 12 , wherein the first and second LED light arrays are connected to the housing.

15. The system of claim 12 , wherein the filter is configured to permit optical signals having a wavelength between about 500 nm and about 550 nm and optical signals having a wavelength between about 600 nm and about 660 nm to pass through the filter to the image sensor.

16. The system of claim 12 , wherein the first and second LED light arrays are each configured to emit light having a wavelength of 405 nm±10 nm.

17. The system of claim 12 , wherein the processor is configured to output data regarding detection of

a location, a population, a quantity, a distribution, a colonization, a contamination, a critical colonization, an infection, and/or an extent of bacteria, fungus, yeast, and/or other microorganisms when present in the illuminated target.

The system of claim 12 , wherein system further comprises a memory storing a pre-determined look-up table of fluorescence, absorbance, and reflectance spectral data of biological, tissue, cellular, and molecular components and/or biomarkers and non-biological materials for comparison with fluorescence, absorbance, and reflectance detected in the target.

The system of claim 12 , wherein the filter is further configured to permit optical signals corresponding to autofluorescence and fluorescence of tissue of the target surface illuminated with the excitation light to pass through the filter to the image sensor and wherein the processor is further configured to receive the detected optical signals corresponding to tissue autofluorescence and tissue fluorescence and to output a representation of tissue components detected in and/or on the target surface based on the detected optical signals corresponding to tissue autofluorescence and tissue fluorescence.

20. The system of claim 12 , further comprising a display.

21. A method of obtaining fluorescence-based data regarding a target, comprising:

Illuminating a target surface with white light emitted by a white light source;

illuminating the target surface with excitation light emitted by at least one excitation light source, the at least one excitation light source emitting one or more wavelengths or wavelength bands of blue or violet excitation light causing at least one biomarker in bacteria, fungus, yeast, and/or other microorganisms in and/or on the target surface to fluoresce;

directing optical signals responsive to illumination of the target surface with the excitation light to a filter with a lens of a portable housing configured to be held in a user's hand during imaging;

filtering the optical signals to permit optical signals corresponding to autofluorescence of bacteria, fungus, yeast, and/or other microorganisms illuminated with the excitation light and having a wavelength between about 500 nm and about 550 nm and/or a wavelength greater than or equal to about 600 nm to pass through the filter to an image sensor contained in the portable housing;

detecting the filtered optical signals with the image sensor;

receiving the detected optical signals at a processor contained in the portable housing and, in response, outputting a representation of bacteria, fungus, yeast, and/or other microorganisms detected in and/or on the target surface, based on the detected optical signals; and

spatially co-registering one or more of absorbance data, reflectance data, endogenous fluorescence data, and exogenous fluorescence data, based on the detected optical signals, with the processor and outputting the co-registered data in the representation of the bacteria, fungus, yeast, and/or other microoganisms detected in and/or on the target surface.

The method of claim 21 , wherein uniformly illuminating the target surface with excitation light includes illuminating the target surface with excitation light from first and second excitation light sources configured to emit blue or violet excitation light and positioned on opposites sides of a common axis passing through the lens, the filter, and the image sensor.

The method of claim 22 , wherein illuminating the target surface with excitation light from first and second excitation light sources configured to emit blue or violet excitation light further includes illuminating the target surface with excitation light from first and second LED light arrays, each array configured to emit light having a wavelength of 405 nm±10 nm.

24. The method of claim 21 , wherein the steps of directing, filtering, and detecting include moving optical signals along a common axis shared by the lens, the filter, and the image sensor and passing through the portable housing.

The method of claim 21 , wherein spatially co-registering the endogenous fluorescence data and/or the exogenous fluorescence data comprises co-registering the fluorescence data, based on the detected optical signals corresponding to autofluorescence of the bacteria, fungus, yeast, and/or other microorganisms detected in and/or on the target surface, relative to one or more of target topography, target anatomy, target area, target depth, target volume, and target margins, in the representation of bacteria, fungus, yeast, and/or other microorganisms illuminated detected in and/or on the target surface.

26. The method of claim 21 , further comprising, based on the detected optical signals corresponding to autofluorescence of bacteria, fungus, yeast, and/or other microorganisms in and/or on the target surface, outputting data regarding detection of

a location, a population, a quantity, a distribution, a colonization, a contamination, a critical colonization, an infection, and/or an extent of bacteria, fungus, yeast, and/or other microorganisms when present in the target.
