Middle east respiratory syndrome coronavirus neutralizing antibodies and methods of use thereof
The present invention provides antibodies that neutralize MERS-CoV and methods of use thereof. The invented antibody is used to treat MERS-CoV infections and symptoms thereof.
1. A method of delaying the onset of one or more symptoms of a MERS-CoV infection, comprising administering to a person at risk of suffering from said infection, a therapeutically effective amount of a monoclonal antibody or a scFv antibody, wherein said monoclonal antibody or scFv antibody binds to an epitope in the receptor binding domain (RBD) of the spike protein of a Middle East Respiratory Syndrome coronavirus (MERS-CoV), and neutralizes MERS-CoV, and wherein the monoclonal antibody or scFv antibody comprises a VH amino acid sequence having SEQ ID NO: 14, and a VL amino acid sequence having SEQ ID NO: 16.
2. A method of delaying the onset of one or more symptoms of a MERS-CoV infection, comprising administering to a person at risk of suffering from said infection, a therapeutically effective amount of a monoclonal antibody or a scFv antibody, and wherein the monoclonal antibody or scFv antibody comprises a heavy chain with three CDRs comprising the amino acid sequences GGTFSSYA (SEQ ID NO:35), IIPIFGTA (SEQ ID NO:38), and ARVGYCSSTSCHIGAFDI (SEQ ID NO:39) respectively and a light chain with three CDRs comprising the amino acid sequences QSVSSS (SEQ ID NO:55), DSS (SEQ ID NO:56), and QQYSSSPYT (SEQ ID NO:57) respectively.
3. A method of neutralizing Middle East Respiratory Syndrome coronavirus (MERS-CoV), comprising contacting an epitope in the receptor binding domain (RBD) of the spike protein of a Middle East Respiratory Syndrome coronavirus (MERS-CoV) with a monoclonal antibody or scFv antibody, wherein the monoclonal antibody or scFv antibody comprises a heavy chain with three CDRs comprising the amino acid sequences GGTFSSYA (SEQ ID NO:35), IIPIFGTA (SEQ ID NO:38), and ARVGYCSSTSCHIGAFDI (SEQ ID NO:39) respectively and a light chain with three CDRs comprising the amino acid sequences QSVSSS (SEQ ID NO:55), DSS (SEQ ID NO:56), and QQYSSSPYT (SEQ ID NO:57) respectively, wherein the monoclonal antibody or scFv antibody neutralizes MERS-CoV.
4. A method of neutralizing Middle East Respiratory Syndrome coronavirus (MERS-CoV), comprising contacting an epitope in the receptor binding domain (RBD) of the spike protein of a Middle East Respiratory Syndrome coronavirus (MERS-CoV) with a monoclonal antibody or scFv antibody, wherein the monoclonal antibody or scFv antibody comprises a VH amino acid sequence having SEQ ID NO: 14 and a VL amino acid sequence having SEQ ID NO: 16.
