Treatment of immune disease by mucosal delivery of antigens using genetically modified
The present invention relates to the treatment of autoimmune and allergic diseases by mucosal delivery by micro-organism, in particular Lactococcus lactis , of secreted immunodominant antigens.
1. A method for treating type 1 diabetes in a subject having established autoimmunity to a beta cell autoantigen, the method comprising orally administering to the subject a composition comprising lactic acid bacteria comprising a nucleic acid sequence integrated into a chromosome of the lactic acid bacteria, wherein said nucleic acid sequence encodes said beta cell autoantigen, and wherein said beta cell autoantigen is constitutively expressed and secreted by said lactic acid bacteria.
2. The method of claim 1 , wherein said beta cell autoantigen is an insulin, a proinsulin, or an insulin fragment.
3. The method of claim 2 , wherein said beta cell autoantigen is a human proinsulin B24-C36, a human insulin, or an InsB 9-23 .
4. The method of claim 1 , wherein said lactic acid bacteria is Lactococcus lactis.
5. The method of claim 1 , wherein said oral administration to said subject:
a. induces regulatory T-cells (Treg) in said subject;
b. reduces proliferation of spleen and inguinal lymph node cells in said subject; and/or
c. suppresses an inflammatory T cell response that is specific to said beta cell autoantigen, in said subject.
6. The method of claim 5 , wherein said Treg cells are Foxp3 + Treg cells.
7. The method of claim 1 , wherein said lactic acid bacteria is:
(i) administered daily;
(ii) administered for at least 1 week;
(iii) administered at least once a day;
(iv) administered in a dose of at least 10 femtogram to 100 mg per day; and/or
(v) administered in a formulation selected from the group consisting of: a spray, a capsule, an aerosol, a lozenges, a bolus, a tablet, a sachet, a liquid, a suspension, an emulsion, and a troche.
8. The method of claim 1 , wherein said lactic acid bacteria is administered to said subject daily.
9. The method of claim 1 , wherein said composition is a medicament, a medical food, a nutraceutical, ora beverage.
10. The method of claim 2 , wherein said lactic acid bacteria is Lactococcus lactis.
11. The method of claim 3 , wherein said lactic acid bacteria is Lactococcus lactis.
12. The method of claim 5 , wherein said lactic acid bacteria is Lactococcus lactis.
13. The method of claim 6 , wherein said lactic acid bacteria is Lactococcus lactis.
14. The method of claim 7 , wherein said lactic acid bacteria is Lactococcus lactis.
15. The method of claim 8 , wherein said lactic acid bacteria is Lactococcus lactis.
16. The method of claim 9 , wherein said lactic acid bacteria is Lactococcus lactis.
