Abstract

The invention relates to a method for synthesising long nucleic acids, including at least one cycle of elongating initial fragments of nucleic acids, including a) a phase comprising the enzymatic addition of nucleotides to said fragments, b) a phase comprising the purification of the fragments having a correct sequence, c) an optional phase of enzymatic amplification, each cycle being performed in a reaction medium which is compatible with enzymatic addition and amplification, such as an aqueous medium, the synthesis method also comprising, at the end of all the elongation cycles, a last step of final amplification. The invention also relates to the use of the method for the production of genes, or sequences of synthetic nucleic acids, DNA or RNA. The invention further relates to a kit for implementing said method.

Claims (34)

1. A process for the synthesis of a nucleic acid of a desired sequence, comprising:

a) attaching initial nucleic acid fragments or elongated fragments to a first support;

b) repeating cycles of (i) contacting the attached initial nucleic acid fragments or the elongated fragments with a modified nucleoside triphosphate and a template-independent DNA polymerase so that initial nucleic acid fragments or elongated fragments are elongated by a modified nucleotide, wherein the modified nucleotide comprises a protective group that prevents multiple additions of nucleotides and that specifically binds to a second support, and (ii) deprotecting the elongated fragments, until at least one of the initial nucleic acid fragments or previously elongated fragments are elongated by Xi nucleotides in the ith cycle;

c) detaching the elongated initial nucleic acid fragments or the elongated fragments from the first support;

d) purifying correctly elongated fragments by specifically binding their protection groups to the second support;

e) deprotecting the protection groups to release the correctly elongated fragments; and

f) repeating a) through e) until a nucleic acid of a desired sequence is obtained.

2. The process of claim 1 , further comprising amplifying said released correctly elongated fragments prior to repeating said step of attaching.

3. The process of claim 1 , wherein said Xi nucleotides are between 1 and 5 nucleotides for every said cycle i.

4. The process of claim 1 , wherein said step of attaching comprises hybridizing a segment of said initial nucleic acid fragments or elongated fragments to a complementary oligonucleotide attached to said first support.

5. The process of claim 1 , wherein said template-independent DNA polymerase is a terminal deoxynucleotidyl transferase, a telomerase, a polymerase η or ζ, a PNPase, or a template-independent RNA polymerase.

6. The process of claim 5 wherein said template-independent DNA polymerase is a terminal deoxynucleotidyl transferase.

7. The process of claim 1 , wherein said modified nucleoside triphosphate is a 3′-O-protected nucleoside triphosphate.

8. The process of claim 1 , wherein said step of purifying comprises specifically binding a 3′-O-protection group to said second support.

9. The process of claim 1 , wherein said modified nucleoside triphosphate has a 3′-O-protection group and a protected nitrogenous base.

10. A process for the synthesis of a nucleic acid of a desired sequence, comprising:

a) attaching initial nucleic acid fragments or correct-sequence elongated fragments to a first support;

b) elongating the initial nucleic acid fragments or correct-sequence elongated fragments by a predetermined number of nucleotides by repeated cycles of reacting said fragments with a modified nucleoside triphosphate and a template-independent DNA polymerase, wherein said modified nucleotide comprises a protective group that prevents multiple additions of nucleotides and that specifically binds to a second support;

c) detaching the elongated initial nucleic acid fragments or the elongated fragments from the first support;

d) purifying correct-sequence elongated fragments by specifically binding their protection groups to the second support;

e) deprotecting the specifically bound protection groups to release the correct-sequence elongated fragments; and

f) repeating steps a) through e) until a nucleic acid of a desired sequence is obtained.

11. The process of claim 10 , wherein said protective group that prevents multiple additions of nucleotides and said protective group that specifically binds to a second support are the same.

12. The process of claim 10 , further comprising amplifying said released correct-sequence fragments prior to repeating said step of attaching.

13. The process of claim 10 wherein said template-independent DNA polymerase is a terminal deoxynucleotidyl transferase and wherein said protective group that prevents multiple additions of nucleotides is a 3′-O-protection group.

14. A process for the synthesis of a nucleic acid of a desired sequence, comprising: elongating initial nucleic acid fragments or correct-sequence elongated fragments attached to a first support and having free 3′-hydroxyls by:

(a) reacting said fragments with a modified nucleoside triphosphate and a template-independent DNA polymerase, wherein said modified nucleoside triphosphate comprises a protective group that prevents multiple additions of nucleotides and that specifically binds to a second support,

(b) removing the protective group that prevents multiple additions of nucleotides;

(c) repeating steps (a) and (b).

15. The process of claim 14 further including steps of:

(d) detaching said elongated fragments from said first support; and

(e) purifying elongated fragments by specifically binding a protection group thereof specific for a second support.

16. The process of claim 15 further including the step of repeating said steps (a) through (c).

17. The process of claim 14 , wherein said template-independent DNA polymerase is a terminal deoxynucleotidyl transferase and wherein said protective group that prevents multiple additions of nucleotides is a 3′-O-protection group.
