Detection units and methods for detecting a target analyte
The present application relates to detection units and methods for detecting one or more target analytes in a sample using a complex formed by a target and first and second probes, wherein the complex comprises an elongated region, a particle that is coupled to the first probe, and a solid support that is coupled to the second probe. Specific binding of a target analyte can be distinguished from non-specific binding of the particle by measuring the displacement of the particle.
1. A kit for detecting the presence of a target analyte in a sample, the kit comprising:
a) a particle;
b) a solid support;
c) a first probe having a first region configured to directly bind said analyte and a second region configured to couple to said particle, wherein the first probe cannot specifically couple to the solid support in the absence of analyte; and
d) a second probe having a first region configured to directly bind said analyte at a location different than the location that the first probe is configured to directly bind the analyte, and a second region configured to couple to said solid support, wherein the second probe cannot specifically couple to the particle in the absence of analyte;
wherein the first and second probes are configured such that the particle indirectly couples to the solid support when a complex is formed by the first and second probes binding to the analyte, and wherein at least one of the first probe, second probe or a region of the analyte between the locations where the first and second probes bind the analyte comprises an elongated region of about 1.0 micron to about 20 microns long.
2. The kit of claim 1 , wherein the first probe comprises an antibody and the second probe comprises an antibody.
3. The kit of claim 1 , wherein the first probe comprises a first nucleic acid and the second probe comprises a second nucleic acid.
4. The kit of claim 1 , wherein the first probe is coupled to the particle in the kit.
5. The kit of claim 1 , wherein the first probe is not coupled to the particle in the kit.
6. The kit of claim 1 , wherein the solid support is coupled to the second probe.
7. The kit of claim 1 , wherein the elongated region is on the first probe.
8. The kit of claim 1 , wherein the elongated region is on the second probe.
9. The kit of claim 1 , wherein the elongated region is on the portion of the analyte between the locations where the first and second probes bind to the analyte.
10. The kit of claim 1 , wherein the particle is a magnetic particle.
11. The kit of claim 10 , wherein the magnetic particle is superparamagnetic.
12. The kit of claim 1 , wherein the particle is fluorescent.
13. The kit of claim 1 , wherein the elongated region is about 1.0 micron to about 5 microns long.
14. The kit of claim 1 , wherein the elongated region is about 5 microns to about 20 microns long.
15. The kit of claim 1 , wherein the particle when indirectly coupled to the solid support via the complex is capable of displacing a measurable distance that is distinguishable from the displacement of a particle directly or non-specifically coupled to the solid support.
16. The kit of claim 1 , wherein the elongated region comprises double-stranded DNA between about 3,000 base pairs to about 60,000 base pairs long.
