Trimethylamine-containing compounds for diagnosis and prediction of disease
The present invention provides markers and methods for determining whether a subject, particularly a human subject, has or is at risk of developing, a disease such as cardiovascular disease, diabetes mellitus, insulin resistance, metabolic syndrome, NAFLD (Nonalcoholic Fatty Liver Disease) or NASH (Nonalcoholic Steatohepatitis) (e.g., within the ensuing year, two years, and/or three years). The present application also relates to the use of such markers and methods for monitoring the status of such diseases in a subject or the effects of therapeutic agents on subjects with such diseases.
1. A method of identifying a subject with elevated risk of cardiovascular disease and treating the subject with a cardiovascular disease therapeutic comprising:
a) identifying a subject as having elevated level of N6-Trimethyl-lysine (TML) in a biological sample obtained from said subject,
wherein said subject is a human,
wherein said identifying comprises comparing the level of TML from a biological sample from said subject to a control value of TML derived from a reference cohort; and
b) treating said subject with a cardiovascular disease therapeutic.
2. The method of claim 1 , further comprising identifying said subject as having elevated levels of trimethylamine-N-oxide (TMAO).
3. The method of claim 2 , wherein said identifying said subject as having elevated levels of TMAO comprises comparing the level of TMAO in a biological sample from said subject to a control value of TMAO derived from a reference cohort.
4. The method of claim 1 , wherein said reference cohort comprises: i) the general population, or ii) individuals characterized as free of symptoms of cardiovascular disease.
