Abstract

Anti-RSV antibodies with neutralizing potency against RSV subtype A and RSV subtype B are provided, as well as methods for their identification, isolation, generation, and methods for their preparation and use are provided.

Claims (43)

1. An isolated antibody or an antigen-binding fragment thereof comprising:

a CDRH1 comprising the amino acid sequence FSFRSYSMN (SEQ ID NO: 563);

a CDRH2 comprising the amino acid sequence SIS-Xaa1-SS-Xaa2-YINYADSVKG (SEQ ID NO: 2225), wherein Xaa1 is S or A and wherein Xaa2 is N or T;

a CDRH3 comprising the amino acid sequence ARDL-Xaa3-PVERGPAFDI (SEQ ID NO: 2226), wherein Xaa3 is S or L;

a CDRL1 comprising the amino acid sequence TGSSSNIG-Xaa4-GYDVH (SEQ ID NO: 2227), wherein Xaa4 is A or R;

a CDRL2 comprising the amino acid sequence Xaa5-NSNRPS (SEQ ID NO: 2228), wherein Xaa5 is A or R; and

a CDRL3 comprising the amino acid sequence QSYDSRLGGS-Xaa6 (SEQ ID NO: 2229), wherein Xaa6 is V or N; and

wherein the antibody or antigen binding fragment thereof is not Antibody 267.

2. The isolated antibody or antigen-binding fragment thereof of claim 1 , wherein:

a) the CDRH1 comprises the amino acid sequence of SEQ ID NO: 2147, the CDRH2 comprises the amino acid sequence of SEQ ID NO: 2148, the CDRH3 comprises the amino acid sequence of SEQ ID NO: 2149, the CDRL1 comprises the amino acid sequence of SEQ ID NO: 2152, the CDRL2 comprises the amino acid sequence of SEQ ID NO: 2153, and the CDRL3 comprises the amino acid sequence of SEQ ID NO: 2154;

b) the CDRH1 comprises the amino acid sequence of SEQ ID NO: 2157, the CDRH2 comprises the amino acid sequence of SEQ ID NO: 2158, the CDRH3 comprises the amino acid sequence of SEQ ID NO: 2159, the CDRL1 comprises the amino acid sequence of SEQ ID NO: 2162, the CDRL2 comprises the amino acid sequence of SEQ ID NO: 2163, and the CDRL3 comprises the amino acid sequence of SEQ ID NO: 2164;

c) the CDRH1 comprises the amino acid sequence of SEQ ID NO: 2167, the CDRH2 comprises the amino acid sequence of SEQ ID NO: 2168, the CDRH3 comprises the amino acid sequence of SEQ ID NO: 2169, the CDRL1 comprises the amino acid sequence of SEQ ID NO: 2172, the CDRL2 comprises the amino acid sequence of SEQ ID NO: 2173, and the CDRL3 comprises the amino acid sequence of SEQ ID NO: 2174;

d) the CDRH1 comprises the amino acid sequence of SEQ ID NO: 2177, the CDRH2 comprises the amino acid sequence of SEQ ID NO: 2178, the CDRH3 comprises the amino acid sequence of SEQ ID NO: 2179, the CDRL1 comprises the amino acid sequence of SEQ ID NO: 2182, the CDRL2 comprises the amino acid sequence of SEQ ID NO: 2183, and the CDRL3 comprises the amino acid sequence of SEQ ID NO: 2184;

e) the CDRH1 comprises the amino acid sequence of SEQ ID NO: 2187, the CDRH2 comprises the amino acid sequence of SEQ ID NO: 2188, the CDRH3 comprises the amino acid sequence of SEQ ID NO: 2189, the CDRL1 comprises the amino acid sequence of SEQ ID NO: 2192, the CDRL2 comprises the amino acid sequence of SEQ ID NO: 2193, and the CDRL3 comprises the amino acid sequence of SEQ ID NO: 2194;

f) the CDRH1 comprises the amino acid sequence of SEQ ID NO: 2197, the CDRH2 comprises the amino acid sequence of SEQ ID NO: 2198, the CDRH3 comprises the amino acid sequence of SEQ ID NO: 2199, the CDRL1 comprises the amino acid sequence of SEQ ID NO: 2202, the CDRL2 comprises the amino acid sequence of SEQ ID NO: 2203, and the CDRL3 comprises the amino acid sequence of SEQ ID NO: 2204;

g) the CDRH1 comprises the amino acid sequence of SEQ ID NO: 2207, the CDRH2 comprises the amino acid sequence of SEQ ID NO: 2208, the CDRH3 comprises the amino acid sequence of SEQ ID NO: 2209, the CDRL1 comprises the amino acid sequence of SEQ ID NO: 2212, the CDRL2 comprises the amino acid sequence of SEQ ID NO: 2213, and the CDRL3 comprises the amino acid sequence of SEQ ID NO: 2214; or

h) the CDRH1 comprises the amino acid sequence of SEQ ID NO: 2217, the CDRH2 comprises the amino acid sequence of SEQ ID NO: 2218, the CDRH3 comprises the amino acid sequence of SEQ ID NO: 2219, the CDRL1 comprises the amino acid sequence of SEQ ID NO: 2222, the CDRL2 comprises the amino acid sequence of SEQ ID NO: 2223, and the CDRL3 comprises the amino acid sequence of SEQ ID NO: 2224.

3. The isolated antibody or antigen-binding fragment thereof of claim 1 , wherein the antibody or antigen-binding fragment thereof comprises:

a) a heavy chain (HC) amino acid sequence of SEQ ID NO: 2146 and a light chain (LC) amino acid sequence of SEQ ID NO: 2151;

b) a heavy chain (HC) amino acid sequence of SEQ ID NO: 2156 and a light chain (LC) amino acid sequence of SEQ ID NO: 2161;

c) a heavy chain (HC) amino acid sequence of SEQ ID NO: 2166 and a light chain (LC) amino acid sequence of SEQ ID NO: 2171;

d) a heavy chain (HC) amino acid sequence of SEQ ID NO: 2176 and a light chain (LC) amino acid sequence of SEQ ID NO: 2181;

e) a heavy chain (HC) amino acid sequence of SEQ ID NO: 2186 and a light chain (LC) amino acid sequence of SEQ ID NO: 2191;

f) a heavy chain (HC) amino acid sequence of SEQ ID NO: 2196 and a light chain (LC) amino acid sequence of SEQ ID NO: 2201;

g) a heavy chain (HC) amino acid sequence of SEQ ID NO: 2206 and a light chain (LC) amino acid sequence of SEQ ID NO: 2211; or

h) a heavy chain (HC) amino acid sequence of SEQ ID NO: 2216 and a light chain (LC) amino acid sequence of SEQ ID NO: 2221.

4. A pharmaceutical composition comprising:

one or more of the isolated antibodies or antigen-binding fragments thereof according to claim 1 ; and

a pharmaceutically acceptable carrier and/or excipient.

A method of treating a Respiratory Syncytial Virus (RSV) infection, or at least one symptom associated with RSV infection, comprising administering to a patient in need thereof or suspected of being in need thereof one or more antibodies or antigen-binding fragments thereof according to claim 1 such that the RSV infection is treated, or the at least one symptom associated with RSV infection is treated, alleviated, or reduced in severity.

6. The method according to claim 5 , wherein the method further comprises administering to the patient a second therapeutic agent.

The method according to claim 6 , wherein the second therapeutic agent is selected from the group consisting of: an antiviral agent; a vaccine specific for RSV, a vaccine specific for influenza virus, or a vaccine specific for metapneumovirus (MPV); an siRNA specific for an RSV antigen or a metapneumovirus (MPV) antigen; a second antibody specific for an RSV antigen or a metapneumovirus (MPV) antigen; an anti-IL4R antibody, an antibody specific for an influenza virus antigen, an anti-RSV-G antibody and a NSAID.

8. A method comprising a step of:

administering the pharmaceutical composition according to claim 4 to a patient suffering from infection with respiratory syncytial virus (RSV).

9. An isolated antibody or an antigen-binding fragment thereof that is a variant of Antibody 267, wherein:

(i) the isolated antibody or antigen-binding fragment thereof comprises:

a CDRH1 comprising the amino acid sequence FSFRSYSMN (SEQ ID NO: 563);

a CDRH2 comprising the amino acid sequence SIS-Xaa1-SS-Xaa2-YINYADSVKG (SEQ ID NO: 2225), wherein Xaa1 is S or A and wherein Xaa2 is N or T;

a CDRH3 comprising the amino acid sequence ARDL-Xaa3-PVERGPAFDI (SEQ ID NO: 2226), wherein Xaa3 is S or L;

a CDRL1 comprising the amino acid sequence TGSSSNIG-Xaa4-GYDVH (SEQ ID NO: 2227), wherein Xaa4 is A or R;

a CDRL2 comprising the amino acid sequence Xaa5-NSNRPS (SEQ ID NO: 2228), wherein Xaa5 is A or R; and

a CDRL3 comprising the amino acid sequence QSYDSRLGGS-Xaa6 (SEQ ID NO: 2229), wherein Xaa6 is V or N; and

(ii) wherein Antibody 267 has a light chain variable region as set forth in SEQ ID NO: 570 and a heavy chain variable region as set forth in SEQ ID NO: 562.
