Abstract

In one embodiment, a masked monoclonal antibody (mAb) is provided, the mAb, encoded by a nucleic acid sequence or an amino acid sequence molecule comprising a signal sequence, a masking epitope sequence, a linker sequence that is cleavable by a protease specific to a target tissue; and an antibody or a functional fragment thereof. In another embodiment, a masked monoclonal antibody (mAb) is provided, which includes a therapeutic mAb and a mask, the mask comprising protein A and protein L attached by a protease cleavable linker.

Claims (23)

1. A non-covalent masked antibody complex comprising:

(i) an antibody; and

(ii) a protein A-protein L mask comprising:

(a) a protein A non-covalently bound to the heavy chain of the variable region of said antibody; and

(b) a protein L non-covalently bound to the light chain of the variable region of said antibody; wherein said protein A and said protein L are bound together by a protease cleavable linker cleavable by a tumor-specific protease,

wherein said antibody is alemtuzumab, bevacizumab, cetuximab, edrecolomab, gemtuzumab, ibritumomab tiuxetan, matuzumab, panitumumab, rituximab, or trastuzumab.

2. A pharmaceutical composition comprising a pharmaceutical excipient and the complex of claim 1 .

3. A method of treating cancer, said method comprising administering a therapeutically effective amount of the complex of claim 1 to a subject in need thereof.

4. A non-covalently masked antibody complex comprising:

(i) an antibody; and

(ii) a protein A-protein L mask comprising:

(a) a protein A bound to the heavy chain of the variable region of said antibody; and

(b) a protein L bound to the light chain of the variable region of said antibody;

wherein said protein A and said protein L are bound together by a tumor associated protease MMP-9 linker comprising the amino acid sequence of SEQ ID NO:10; and

wherein said antibody is alemtuzumab, bevacizumab, cetuximab, edrecolomab, gemtuzumab, ibritumomab tiuxetan, matuzumab, panitumumab, rituximab, or trastuzumab.

5. A pharmaceutical composition comprising (1) a non-covalently masked antibody complex comprising:

(i) an antibody; and

(ii) a protein A-protein L mask comprising:

(a) a protein A bound to the heavy chain of the variable region of said antibody; and

(b) a protein L bound to the light chain of the variable region of said antibody;

wherein said protein A and said protein L are bound together by a tumor associated protease MMP-9 linker comprising the amino acid sequence of SEQ ID NO:10; and

wherein said antibody is trastuzumab;

and (2) a pharmaceutically acceptable carrier.
