Abstract

The present invention provides immunogenic compositions having one or more polysaccharide-protein conjugates in which one or more polysaccharides from Streptococcus pneumoniae bacterial capsules are conjugated to a carrier protein in an aprotic solvent such as dimethylsulfoxide (DMSO). The present invention also provides methods for providing an enhanced immune response to a pneumococcal polysaccharide protein conjugate vaccine comprising administering to a human subject an immunogenic composition comprising polysaccharide-protein conjugates prepared in DMSO conditions.

Claims (14)

1. An immunogenic composition consisting essentially of polysaccharide from S. pneumoniae serotypes 1, 3, 4, 5, 6A, 6B, 7F, 9V, 14, 18C, 19A, 19F, 22F, 23F and 33F each conjugated to a carrier protein, wherein the conjugation reaction for S. pneumoniae serotypes 6A, 6B, 7F, 18C, 19A, 19F, and 23F is in an aprotic solvent and the conjugation reaction for S. pneumoniae serotypes 1, 3, 4, 5, 9V, 14, 22F and 33F is in an aqueous solvent.

2. The immunogenic composition of claim 1 , wherein the conjugation reaction is reductive amination.

3. The immunogenic composition of claim 1 , wherein the carrier protein is CRM 197 .

4. The immunogenic composition of claim 1 , wherein the aprotic solvent is dimethylsulfoxide (DMSO).

5. The immunogenic composition of claim 1 , further comprising about 0.2% w/v Polysorbate 20.

An immunogenic composition consisting essentially of polysaccharide from S. pneumoniae serotypes 1, 3, 4, 5, 6A, 6B, 7F, 9V, 14, 18C, 19A, 19F, 22F, 23F and 33F each conjugated to CRM 197 ; wherein the conjugation reaction for S. pneumoniae serotypes 6A, 6B, 7F, 18C, 19A, 19F, and 23F is in an aprotic solvent, wherein the aprotic solvent is DMSO; and the conjugation reaction for S. pneumoniae serotypes 1, 3, 4, 5, 9V, 14, 22F and 33F is in an aqueous solvent.

7. The immunogenic composition of claim 6 , further comprising about 0.2% w/v Polysorbate 20.

An immunogenic composition consisting of polysaccharide from S. pneumoniae serotypes 1, 3, 4, 5, 6A, 6B, 7F, 9V, 14, 18C, 19A, 19F, 22F, 23F and 33F each conjugated to a carrier protein, wherein the conjugation reaction for S. pneumoniae serotypes 6A, 6B, 7F, 18C, 19A, 19F, and 23F is in an aprotic solvent and the conjugation reaction for S. pneumoniae serotypes 1, 3, 4, 5, 9V, 14, 22F and 33F is in an aqueous solvent.

9. The immunogenic composition of claim 8 , wherein the conjugation reaction is reductive amination.

10. The immunogenic composition of claim 8 , wherein the carrier protein is CRM 197 .

11. The immunogenic composition of claim 8 , wherein the aprotic solvent is dimethylsulfoxide (DMSO).

12. The immunogenic composition of claim 8 , further comprising about 0.2% w/v Polysorbate 20.

An immunogenic composition consisting of polysaccharide from S. pneumoniae serotypes 1, 3, 4, 5, 6A, 6B, 7F, 9V, 14, 18C, 19A, 19F, 22F, 23F and 33F each conjugated to CRM 197 ; wherein the conjugation reaction for S. pneumoniae serotypes 6A, 6B, 7F, 18C, 19A, 19F, and 23F is in an aprotic solvent, wherein the aprotic solvent is DMSO; and the conjugation reaction for S. pneumoniae serotypes 1, 3, 4, 5, 9V, 14, 22F and 33F is in an aqueous solvent.

14. The immunogenic composition of claim 13 , further comprising about 0.2% w/v Polysorbate 20.
