Abstract

The present invention provides compositions and systems for delivery of nanocarriers to cells of the immune system. The invention provides vaccine nanocarriers capable of stimulating an immune response in T cells and/or in B cells, in some embodiments, comprising at least one immunomodulatory agent, and optionally comprising at least one targeting moiety and optionally at least one immunostimulatory agent. The invention provides pharmaceutical compositions comprising inventive vaccine nanocarriers. The present invention provides methods of designing, manufacturing, and using inventive vaccine nanocarriers and pharmaceutical compositions thereof. The invention provides methods of prophylaxis and/or treatment of diseases, disorders, and conditions comprising administering at least one inventive vaccine nanocarrier to a subject in need thereof.

Claims (26)

1. A dosage unit for inducing tolerance to an antigen in an individual, the dosage unit comprising

polymeric nanoparticles having a mean geometric diameter between about 100 nm and 400 nm and comprising rapamycin to induce regulatory T cells,

the polymeric nanoparticles formed by self-assembly of a polymer blend comprising

a) a hydrophobic polymer selected from the group consisting of polylactic acid and polylactide-co-glycolide, and

b) an amphiphilic polymer having a first end formed of a hydrophobic polymer selected from the group consisting of polylactic acid and polylactide-co-glycolide and a second end formed of a hydrophilic polyethylene glycol polymer,

wherein the hydrophilic polyethylene glycol polymer end of the amphiphilic polymer is present on the surface of the nanoparticles and the hydrophobic polymer end of the amphiphilic polymer forms the core of the nanoparticles,

wherein the polymeric nanoparticles are selectively taken up by the subcapsular sinus of the draining lymph nodes and induce tolerance to the antigen when the antigen is co-administered with the nanoparticles.

2. The dosage unit of claim 1 , wherein the rapamycin and the antigen are in or associated with the same polymeric nanoparticle.

The dosage unit of claim 2 , wherein the antigen is encapsulated within the polymeric nanoparticles, on the surface of the polymeric nanoparticles, or bound to polylactic acid, polylactide-co-glycolide, or amphiphilic polymer having a first end formed of a hydrophobic polymer selected from the group consisting of polylactic acid and polylactide-co-glycolide and a second end formed of a hydrophilic polyethylene glycol polymer.

The dosage unit of claim 1 , wherein the rapamycin is encapsulated within the polymeric nanoparticles, on the surface of the polymeric nanoparticles, and/or covalently bound to the polylactic acid, polylactide-co-glycolide, or amphiphilic polymer having a first end formed of a hydrophobic polymer selected from the group consisting of polylactic acid and polylactide-co-glycolide and a second end formed of a hydrophilic polyethylene glycol polymer.

5. The dosage unit of claim 1 , wherein the antigen is selected from the group consisting of degenerative disease antigens, atopic disease antigens, autoimmune disease antigens, alloantigens, xenoantigens, self-antigens, metabolic disease enzymes or enzymatic product thereof, and allergen.

6. The dosage unit of claim 5 , wherein the antigen is a metabolic disease enzyme or an enzymatic product thereof.

7. The dosage unit of claim 5 wherein the antigen is a T cell antigen.

8. The dosage unit of claim 5 wherein the antigen is a B cell antigen.

9. The dosage unit of claim 1 wherein the polymeric nanoparticles comprise polylactide-co-glycolide.

10. The dosage unit of claim 1 , wherein the diameter of the nanoparticles is between 200 and 350 nm.

11. The dosage unit of claim 1 , wherein the polymeric nanoparticles comprise a targeting agent on the polymeric nanoparticles' surface,

wherein the targeting agent is for delivery of the polymeric nanoparticles to antigen presenting cells and T cells.

12. The dosage unit of claim 1 , wherein the polymeric nanoparticles have a diameter between 200 nm and 400 nm.

13. A method of therapeutically or prophylactically treating an individual in need thereof comprising administering to the individual an effective amount of the dosage unit of claim 1 in combination with antigen to induce tolerance to the antigen.

14. The method of claim 13 , wherein the antigen is a metabolic disease enzyme or an enzymatic product thereof.

15. The method of claim 13 , wherein the antigen is an alloantigen or a xenoantigen.

16. The method of claim 13 , wherein the individual has an allergic disease or an autoimmune disease.

17. The method of claim 16 wherein the individual has a food allergy.

18. The method of claim 17 wherein the food allergy is against milk, eggs, peanuts, tree nuts, fish, shellfish, soy, or wheat.

19. The method of claim 16 wherein the autoimmune disease is selected from the group consisting of lupus, multiple sclerosis, rheumatoid arthritis, diabetes mellitus type I, inflammatory bowel disease, thyroiditis, and celiac disease.
