Abstract

In certain embodiments the invention is directed to a process for preparing an oxycodone hydrochloride composition having less than 25 ppm of 14-hydroxycodeinone.

Claims (19)

1. A process for purifying a composition comprising an oxycodone free base or acid addition salt thereof comprising performing the following steps sequentially:

(i) hydrogenating the composition in the presence of a hydrogenation catalyst under acidic conditions;

(ii) increasing the pH of the composition;

(iii) removing the hydrogenation catalyst; and

(iv) isolating the purified oxycodone free base or acid addition salt thereof.

2. The process of claim 1 , wherein the hydrogen pressure is vented after step (i) and prior to step (ii).

3. The process of claim 1 , wherein the process comprises purifying an oxycodone free base composition.

4. The process of claim 1 , wherein the process comprises purifying an oxycodone acid addition salt composition.

5. The process of claim 4 , wherein the acid addition salt is hydrochloric acid.

6. The process of claim 4 , wherein the acid addition salt is sulfuric acid.

7. The process of claim 1 , wherein the composition further comprises 8α,14-dihydroxy-7,8-dihydrocodeinone prior to step (i).

8. The process of claim 7 , wherein the amount of 8α,14-dihydroxy-7,8-dihydrocodeinone is reduced during step (i).

9. The process of claim 8 , wherein the wherein the ratio of 8α,14-dihydroxy-7,8-dihydrocodeinone to oxycodone is 0.04% or less as measured by HPLC following step (iv).

10. The process of claim 7 , wherein the composition further comprises 14-hydroxycodeinone prior to step (i).

11. The process of claim 10 , wherein the amount of 14-hydroxycodeinone is reduced during step (i).

12. The process of claim 11 , wherein the amount of 14-hydroxycodeinone in the composition following step (iv) is less than 100 ppm.

13. The process of claim 11 , wherein the amount of 14-hydroxycodeinone in the composition following step (iv) is less than 25 ppm.

14. The process of claim 11 , wherein the amount of 14-hydroxycodeinone in the composition following step (iv) is less than 10 ppm.

15. The process of claim 14 , further comprising recrystallizing the purified oxycodone free base or acid addition salt thereof isolated in step (iv), wherein the amount of 14-hydroxycodeinone following the recrystallization is less than 10 ppm.
