Abstract

Improved yeast cell with increased succinic acid production based on yield and titer. Increased activity of one or more enzymes involved in the pentose phosphate pathway, reducing flux through phosphoglucose isomerase, increasing flux to cytoplasmic acetyl-CoA, installation of a malic enzyme, and/or installation of a formate dehydrogenase leads to increased production of succinic acid.

Claims (6)

1. A genetically modified yeast cell with increased succinic acid production, comprising at least one genetic modification that increases flux through the pentose phosphate pathway and a genetic modification that increases activity of enzymes for a cytoplasmic reductive pathway from phosphoenolpyruvate to succinate wherein said enzymes for a cytoplasmic reductive pathway comprising phosphoenolpyruvate carboxykinase (PCK), malate dehydrogenase (MDH), fumarase (FUM), and fumarate reductase (FRD).

2. The genetically modified yeast cell with increased succinic acid production of claim 1 , wherein said at least one genetic modification that increases flux through the pentose phosphate pathway increases the activity of glucose-6-phosphate dehydrogenase (ZWF).

3. The genetically modified yeast cell with increased succinic acid production of claim 1 , wherein said at least one genetic modification that increases flux through the pentose phosphate pathway decreases the activity of phosphoglucose isomerase (PGI).

The genetically modified yeast cell with increased succinic acid production of claim 1 , wherein said at least one genetic modification that increases flux through the pentose phosphate pathway decreases the activity of phosphoglucose isomerase (PGI) and increases the activity of one or more enzymes chosen from the group consisting of glucose-6-phosphate dehydrogenase (ZWF), 6-phosphogluconate lactonase (PGL), and 6-phosphogluconate dehydrogenase (GND).

5. The genetically modified yeast cell with increased succinic acid production of claim 1 , further comprising at least one genetic modification that increases the activity of malic enzyme (MAE).

6. The genetically modified yeast cell with increased succinic acid production of claim 5 , further comprising at least one genetic modification that decreases the activity of a pyruvate carboxylase (PYC).
