Abstract

The present disclosure relates to compositions and methods for treating disease. More particularly, the disclosure relates to synthetic proteins and their use for treating cancer.

Claims (22)

1. A synthetic protein, comprising:

a synthetic Epidermal Growth Factor (sEGF) growth factor comprising at least one synthetic targeted signaling pathway (sTSP) domain of a human Epidermal Growth Factor (hEGF) TSP (hTSP) domain, wherein the at least one sTSP is 95%, 96%, 97%, 98%, 99% or 100% identical to the amino acid sequence NTENDCPLSHEAYCLHDGVCMYIEALDKYACNCVVGYVGERCQFRDLRWWDAR (SEQ ID NO: 33), excluding the following amino acid changes: T2S, E3D, N4S, DSE, E11D, A12G, V38I, F44Y, R48K, F511, and A52L;

at least one linker; and

an immunogenic polypeptide, wherein the at least one linker includes a first linker that separates the sTSP from the immunogenic polypeptide.

2. The synthetic protein according to claim 1 , wherein the immunogenic polypeptide comprises a cholera toxin B (CT-B) protein.

The synthetic protein according to claim 1 , wherein the first linker is selected from the group consisting of SSG (SEQ ID NO: 13), GSSG (SEQ ID NO: 14), SSGGG (SEQ ID NO: 15), SGG (SEQ ID NO: 16), GGSGG (SEQ ID NO: 17), GGGGS (SEQ ID NO: 18), SSGGGSGG (SEQ ID NO: 19), SSGGGGSGGG (SEQ ID NO: 20), TSGGGSG (SEQ ID NO: 21), TSGGGGSGG (SEQ ID NO: 22), SSGGGSGGSSG (SEQ ID NO: 23), GGSGGTSGGGSG (SEQ ID NO: 24), SGGTSGGGGSGG (SEQ ID NO: 25), GGSGGTSGGGGSGG (SEQ ID NO: 26), SSGGGGSGGGSSG (SEQ ID NO: 27), SSGGGSGGSSGGG (SEQ ID NO: 28), SGGGGSGGGSSGGG (SEQ ID NO: 29) and GGSGGTSGGGGGSG (SEQ ID NO: 30).

4. The synthetic protein according to claim 3 , wherein the first linker is GGSGGTSGGGGGSG (SEQ ID NO: 30).

5. The synthetic protein according to claim 1 , wherein the synthetic growth factor comprises a first sTSP domain and a second sTSP domain.

6. The synthetic protein according to claim 5 , wherein the at least one linker includes a second linker that separates the first sTSP domain and the second sTSP domain, and the first linker separates the second sTSP domain from the immunogenic polypeptide.

The synthetic protein according to claim 6 , wherein the second linker is selected from the group consisting of SSG (SEQ ID NO: 13), GSSG (SEQ ID NO: 14), SSGGG (SEQ ID NO: 15), SGG (SEQ ID NO: 16), GGSGG (SEQ ID NO: 17), GGGGS (SEQ ID NO: 18), SSGGGSGG (SEQ ID NO: 19), SSGGGGSGGG (SEQ ID NO: 20), TSGGGSG (SEQ ID NO: 21), TSGGGGSGG (SEQ ID NO: 22), SSGGGSGGSSG (SEQ ID NO: 23), GGSGGTSGGGSG (SEQ ID NO: 24), SGGTSGGGGSGG (SEQ ID NO: 25), GGSGGTSGGGGSGG (SEQ ID NO: 26), SSGGGGSGGGSSG (SEQ ID NO: 27), SSGGGSGGSSGGG (SEQ ID NO: 28), and SSGGGGSGGGSSGGG (SEQ ID NO: 29).

8. The synthetic protein according to claim 6 , wherein the second linker is GSSG (SEQ ID NO: 14).

9. The synthetic protein according to claim 1 , wherein the synthetic protein has the amino acid sequence of SEQ ID NO:2.

10. An immunogenic composition, comprising the synthetic protein of claim 9 and one or more components selected from the group consisting of an adjuvant, a carrier, an excipient, a diluent, and combinations thereof.

11. An immunogenic composition, comprising the synthetic protein of claim 1 and one or more components selected from the group consisting of an adjuvant, a carrier, an excipient, a diluent, and combinations thereof.

12. The immunogenic composition according to claim 11 , wherein the immunogenic polypeptide comprises a cholera toxin B (CT-B) protein.

The immunogenic composition according to claim 11 , wherein the first linker is selected from the group consisting of SSG (SEQ ID NO: 13), GSSG (SEQ ID NO: 14), SSGGG (SEQ ID NO: 15), SGG (SEQ ID NO: 16), GGSGG (SEQ ID NO: 17), GGGGS (SEQ ID NO: 18), SSGGGSGG (SEQ ID NO: 19), SSGGGGSGGG (SEQ ID NO: 20), TSGGGSG (SEQ ID NO: 21), TSGGGGSGG (SEQ ID NO: 22), SSGGGSGGSSG (SEQ ID NO: 23), GGSGGTSGGGSG (SEQ ID NO: 24), SGGTSGGGGSGG (SEQ ID NO: 25), GGSGGTSGGGGSGG (SEQ ID NO: 26), SSGGGGSGGGSSG (SEQ ID NO: 27), SSGGGSGGSSGGG (SEQ ID NO: 28), SSGGGGSGGGSSGGG (SEQ ID NO: 29) and GGSGGTSGGGGGSG (SEQ ID NO: 30).

14. The immunogenic composition according to claim 13 , wherein the first linker is GGSGGTSGGGGGSG (SEQ ID NO: 30).

15. The immunogenic composition according to claim 11 , wherein the synthetic Epidermal Growth Factor (sEGF) growth factor comprises a first sTSP domain and a second sTSP domain.

16. The immunogenic composition according to claim 15 , wherein the at least one linker includes a second linker that separates the first sTSP domain and the second sTSP domain, and the first linker separates the second sTSP domain from the immunogenic polypeptide.

The immunogenic composition according to claim 16 , wherein the second linker is selected from the group consisting of SSG (SEQ ID NO: 13), GSSG (SEQ ID NO: 14), SSGGG (SEQ ID NO: 15), SGG (SEQ ID NO: 16), GGSGG (SEQ ID NO: 17), GGGGS (SEQ ID NO: 18), SSGGGSGG (SEQ ID NO: 19), SSGGGGSGGG (SEQ ID NO: 20), TSGGGSG (SEQ ID NO: 21), TSGGGGSGG (SEQ ID NO: 22), SSGGGSGGSSG (SEQ ID NO: 23), GGSGGTSGGGSG (SEQ ID NO: 24), SGGTSGGGGSGG (SEQ ID NO: 25), GGSGGTSGGGGSGG (SEQ ID NO: 26), SSGGGGSGGGSSG (SEQ ID NO: 27), SSGGGSGGSSGGG (SEQ ID NO: 28), and SSGGGGSGGGSSGGG (SEQ ID NO: 29).

18. The immunogenic composition according to claim 17 , wherein the second linker is GSSG (SEQ ID NO: 14).

19. The immunogenic composition according to claim 11 , wherein the synthetic protein has the amino acid sequence of SEQ ID NO:2.
