Abstract

The present disclosure relates to bifunctional compounds, which find utility as modulators of estrogen receptor (target protein). In particular, the present disclosure is directed to bifunctional compounds, which contain on one end at least one of a Von Hippel-Lindau ligand, a cereblon ligand, inhibitors of apoptosis proteins ligand, mouse double-minute homolog 2 ligand, or a combination thereof, which binds to the respective E3 ubiquitin ligase, and on the other end a moiety which binds the target protein, such that the target protein is placed in proximity to the ubiquitin ligase to effect degradation (and inhibition) of target protein. The present disclosure exhibits a broad range of pharmacological activities associated with degradation/inhibition of target protein. Diseases or disorders that result from aggregation or accumulation of the target protein are treated or prevented with compounds and compositions of the present disclosure.

Claims (30)

1. A method of treating breast cancer in a subject comprising administering to a subject in need thereof a composition comprising a pharmaceutically acceptable carrier and an effective amount of a compound having the structure:

or a pharmaceutically acceptable salt, enantiomer, stereoisomer, isotopic derivative, or prodrug of any of the foregoing.

2. A method of treating breast cancer in a subject comprising administering to a subject in need thereof a composition comprising a pharmaceutically acceptable carrier and an effective amount of a compound having the structure:

or a pharmaceutically acceptable salt, enantiomer, or stereoisomer thereof.

3. A method of treating breast cancer in a subject comprising administering to a subject in need thereof a composition comprising a pharmaceutically acceptable carrier and an effective amount of a compound having the structure:

or a pharmaceutically acceptable salt, enantiomer, or stereoisomer thereof.

4. A method of treating breast cancer in a subject comprising administering to a subject in need thereof a composition comprising a pharmaceutically acceptable carrier and an effective amount of a compound having the structure:

or a pharmaceutically acceptable salt, enantiomer, or stereoisomer thereof.

5. A method of treating breast cancer in a subject comprising administering to a subject in need thereof a composition comprising a pharmaceutically acceptable carrier and an effective amount of a compound having the structure:

or a pharmaceutically acceptable salt, enantiomer, or stereoisomer thereof.

6. The method of claim 1 , wherein the composition further comprises an effective amount of at least one additional anti-cancer agent.

7. The method of claim 2 , wherein the composition further comprises an effective amount of at least one additional anti-cancer agent.

8. The method of claim 3 , wherein the composition further comprises an effective amount of at least one additional anti-cancer agent.

9. The method of claim 4 , wherein the composition further comprises an effective amount of at least one additional anti-cancer agent.

10. The method of claim 5 , wherein the composition further comprises an effective amount of at least one additional anti-cancer agent.

11. The method of claim 6 , wherein the anti-cancer agent is docetaxel, goserelin acetate, triptorelin, buserelin, flutamide, bicalutamide, nilutamide, pamidronate, zolendronate, everolimus, pazopanib, carboplatin, cisplatin, oxaliplatin, epithilone B, fulvestrant, acolbifene, lasofoxifene, idoxifene, topotecan, pemetrexed, erlotinib, ticilimumab, ipilimumab, vorinostat, etoposide, gemcitabine, doxorubicin, 5′-deoxy-5-fluorouridine, vincristine, temozolomide, capecitabine, camptothecin, PD0325901, tamoxifen, toremifene, anastrazole, letrozole, bevacizumab, raloxifene, alpelisib, paclitaxel, abraxane, trastuzumab, palbociclib, ribociclib, or abemaciclib.

12. The method of claim 11 , wherein the anti-cancer agent is everolimus, capecitabine, docetaxel, paclitaxel, or abraxane.

13. The method of claim 7 , wherein the anti-cancer agent is docetaxel, goserelin acetate, triptorelin, buserelin, flutamide, bicalutamide, nilutamide, pamidronate, zolendronate, everolimus, pazopanib, carboplatin, cisplatin, oxaliplatin, epithilone B, fulvestrant, acolbifene, lasofoxifene, idoxifene, topotecan, pemetrexed, erlotinib, ticilimumab, ipilimumab, vorinostat, etoposide, gemcitabine, doxorubicin, 5′-deoxy-5-fluorouridine, vincristine, temozolomide, capecitabine, camptothecin, PD0325901, tamoxifen, toremifene, anastrazole, letrozole, bevacizumab, raloxifene, alpelisib, paclitaxel, abraxane, trastuzumab, palbociclib, ribociclib, or abemaciclib.

14. The method of claim 13 , wherein the anti-cancer agent is everolimus, capecitabine, docetaxel, paclitaxel, or abraxane.

The method of claim 8 , wherein the anti-cancer agent is docetaxel, goserelin acetate, triptorelin, buserelin, flutamide, bicalutamide, nilutamide, pamidronate, zolendronate, everolimus, pazopanib, carboplatin, cisplatin, oxaliplatin, epithilone B, fulvestrant, acolbifene, lasofoxifene, idoxifene, topotecan, pemetrexed, erlotinib, ticilimumab, ipilimumab, vorinostat, etoposide, gemcitabine, doxorubicin, 5′-deoxy-5-fluorouridine, vincristine, temozolomide, capecitabine, camptothecin, PD0325901, tamoxifen, toremifene, anastrazole, letrozole, bevacizumab, oserelin acetate, raloxifene, alpelisib, paclitaxel, abraxane, trastuzumab, palbociclib, ribociclib, or abemaciclib.

16. The method of claim 15 , wherein the anti-cancer agent is everolimus, capecitabine, docetaxel, paclitaxel, or abraxane.

The method of claim 9 , wherein the anti-cancer agent is docetaxel, goserelin acetate, triptorelin, buserelin, flutamide, bicalutamide, nilutamide, pamidronate, zolendronate, everolimus, pazopanib, carboplatin, cisplatin, oxaliplatin, paclitaxel, epithilone B, fulvestrant, acolbifene, lasofoxifene, idoxifene, topotecan, pemetrexed, erlotinib, ticilimumab, ipilimumab, vorinostat, etoposide, gemcitabine, doxorubicin, 5′-deoxy-5-fluorouridine, vincristine, temozolomide, capecitabine, camptothecin, PD0325901, tamoxifen, toremifene, anastrazole, letrozole, bevacizumab, raloxifene, alpelisib, abraxane, trastuzumab, palbociclib, ribociclib, or abemaciclib.

18. The method of claim 17 , wherein the anti-cancer agent is everolimus, capecitabine, docetaxel, paclitaxel, or abraxane.

The method of claim 10 , wherein the anti-cancer agent is docetaxel, goserelin acetate, triptorelin, buserelin, flutamide, bicalutamide, nilutamide, pamidronate, zolendronate, everolimus, pazopanib, carboplatin, cisplatin, oxaliplatin, epithilone B, fulvestrant, acolbifene, lasofoxifene, idoxifene, topotecan, pemetrexed, erlotinib, ticilimumab, ipilimumab, vorinostat, etoposide, gemcitabine, doxorubicin, 5′-deoxy-5-fluorouridine, vincristine, temozolomide, capecitabine, camptothecin, PD0325901, tamoxifen, toremifene, anastrazole, letrozole, bevacizumab, raloxifene, alpelisib, paclitaxel, abraxane, trastuzumab, palbociclib, ribociclib, or abemaciclib.

20. The method of claim 19 , wherein the anti-cancer agent is everolimus, capecitabine, docetaxel, paclitaxel, or abraxane.

The method of claim 6 , wherein the anti-cancer agent is a FLT-3 inhibitor, a VEGFR inhibitor, an EGFR TK inhibitor, an aurora kinase inhibitor, a PIK-1 modulator, a Bcl-2 inhibitor, an HDAC inhibitor, a c-MET inhibitor, a PARP inhibitor, a CDK inhibitor, an anti-HGF antibody, a PI3 kinase inhibitor, an AKT inhibitor, an mTORC1/2 inhibitor, a JAK/STAT inhibitor, a checkpoint-1 inhibitor, a checkpoint-2 inhibitor, a focal adhesion kinase inhibitor, a MAP kinase inhibitor, or a VEGF trap antibody.

The method of claim 7 , wherein the anti-cancer agent is a FLT-3 inhibitor, a VEGFR inhibitor, an EGFR TK inhibitor, an aurora kinase inhibitor, a PIK-1 modulator, a Bcl-2 inhibitor, an HDAC inhibitor, a c-MET inhibitor, a PARP inhibitor, a CDK inhibitor, an anti-HGF antibody, a PI3 kinase inhibitor, an AKT inhibitor, an mTORC1/2 inhibitor, a JAK/STAT inhibitor, a checkpoint-1 inhibitor, a checkpoint-2 inhibitor, a focal adhesion kinase inhibitor, a MAP kinase inhibitor, or a VEGF trap antibody.

The method of claim 8 , wherein the anti-cancer agent is a FLT-3 inhibitor, a VEGFR inhibitor, an EGFR TK inhibitor, an aurora kinase inhibitor, a PIK-1 modulator, a Bcl-2 inhibitor, an HDAC inhibitor, a c-MET inhibitor, a PARP inhibitor, a CDK inhibitor, an anti-HGF antibody, a PI3 kinase inhibitor, an AKT inhibitor, an mTORC1/2 inhibitor, a JAK/STAT inhibitor, a checkpoint-1 inhibitor, a checkpoint-2 inhibitor, a focal adhesion kinase inhibitor, a MAP kinase inhibitor, or a VEGF trap antibody.
