Abstract

Disclosed herein is a vaccine comprising a Middle East Respiratory Syndrome coronavirus (MERS-CoV) antigen. The antigen can be a consensus antigen. The consensus antigen can be a consensus spike antigen. Also disclosed herein is a method of treating a subject in need thereof, by administering the vaccine to the subject.

Claims (30)

1. A nucleic acid molecule comprising a nucleotide sequence selected from the group consisting of:

(a) an immunogenic fragment of SEQ ID NO: 1, wherein the fragment encodes a sequence having 100% identity to consecutive amino acids over at least 60% of the full length of SEQ ID NO: 2;

(b) an immunogenic fragment of SEQ ID NO: 3, wherein the fragment encodes a sequence having 100% identity to consecutive amino acids over at least 60% of the full length of SEQ ID NO: 2;

(c) a nucleotide sequence having at least 90% identity over an entire length of the nucleic acid sequence set forth in SEQ ID NO: 1;

(d) a nucleotide sequence having at least 90% identity over an entire length of the nucleic acid sequence set forth in SEQ ID NO: 3;

(e) an immunogenic fragment of SEQ ID NO: 1, wherein the fragment comprises a sequence having 100% identity to consecutive bases over at least 60% of the full length of SEQ ID NO:1; and

(f) an immunogenic fragment of SEQ ID NO: 3, wherein the fragment comprises a sequence having 100% identity to consecutive bases over at least 60% of the full length of SEQ ID NO: 3.

2. The nucleic acid molecule of claim 1 , wherein the nucleic acid molecule comprises an expression vector.

3. The nucleic acid molecule of claim 1 , wherein the nucleic acid molecule is incorporated into a viral particle.

4. An immunogenic composition comprising the nucleic acid molecule of claim 1 .

5. The immunogenic composition of claim 4 , further comprising at least one selected from the group consisting of a pharmaceutically acceptable excipient and an adjuvant.

6. A method of inducing an immune response against a Middle East Respiratory Syndrome coronavirus (MERS-CoV) in a subject in need thereof, the method comprising administering the nucleic acid molecule of claim 1 to the subject.

7. The method of claim 6 , wherein administering includes at least one of electroporation and injection.

8. A method of protecting a subject in need thereof from infection with a Middle East Respiratory Syndrome coronavirus (MERS-CoV), the method comprising administering the nucleic acid molecule of claim 1 to the subject.

9. The method of the claim 8 , wherein administering includes at least one of electroporation and injection.

10. A method of treating a subject in need thereof against Middle East Respiratory Syndrome coronavirus (MERS-CoV), the method comprising administering the nucleic acid molecule of claim 1 to the subject, wherein the subject is thereby resistant to one or more MERS-CoV strains.

11. The method of claim 10 , wherein administering includes at least one of electroporation and injection.

12. A method of inducing an immune response against a Middle East Respiratory Syndrome coronavirus (MERS-CoV) in a subject in need thereof, the method comprising administering a peptide to the subject, wherein the peptide is selected from the group consisting of:

a) the peptide comprises an amino acid sequence as set forth in SEQ ID NO:2, and

b) the peptide comprises an amino acid sequence as set forth in SEQ ID NO:4.

13. The method of claim 12 , wherein administering includes at least one of electroporation and injection.

14. A method of protecting a subject in need thereof from infection with a Middle East Respiratory Syndrome coronavirus (MERS-CoV), the method comprising administering a peptide to the subject, wherein the peptide is selected from the group consisting of:

a) the peptide comprises an amino acid sequence as set forth in SEQ ID NO:2, and

b) the peptide comprises an amino acid sequence as set forth in SEQ ID NO:4.

15. The method of the claim 14 , wherein administering includes at least one of electroporation and injection.

16. A method of treating a subject in need thereof against Middle East Respiratory Syndrome coronavirus (MERS-CoV), the method comprising administering a peptide to the subject, wherein the peptide is selected from the group consisting of:

a) the peptide comprises an amino acid sequence as set forth in SEQ ID NO:2, and

b) the peptide comprises an amino acid sequence as set forth in SEQ ID NO:4,

wherein the subject is thereby resistant to one or more MERS-CoV strains.

17. The method of claim 16 , wherein administering includes at least one of electroporation and injection.
