Abstract

Exemplary systems are for imaging of samples, for example using an array of micro optical elements. In some embodiments, an optical chip including an array of micro optical elements moves relative to an imaging window and a detector in order to scan over a sample to produce an image. A focal plane can reside within a sample or on its surface during imaging. In some embodiments, detecting optics are used to detect back-emitted light collected by an array of micro optical elements that is generated by an illumination beam impinging on a sample. In some embodiments, an imaging system has a large field of view and a large optical chip such that an entire surface of a sample can be imaged quickly. In some embodiments, a sample is accessible by a user during imaging due to the sample being exposed.

Claims (46)

1. A microscopy system for imaging of samples, comprising:

an exposed transparent imaging window, the transparent imaging window providing a surface onto or over which a sample may be disposed in order to image the sample; and

a housing that at least partially encloses the microscopy system, wherein the housing comprises:

an upper working surface,

an imaging window support base that is recessed below the upper working surface, and

an imaging window support disposed on the imaging window support base,

wherein the transparent imaging window is disposed on the imaging window support,

wherein the imaging window is recessed below the upper working surface.

2. The microscopy system of claim 1 , wherein the sample is accessible to a user during imaging.

3. The microscopy system of claim 1 , wherein the imaging window support is attached to the imaging window, and the imaging window support and the imaging window are both sized and shaped to accommodate an optical interface of a sample dish.

4. The microscopy system of claim 1 , wherein the surface of the transparent imaging window is disposed within 1 cm of the upper working surface.

5. The microscopy system of claim 1 , wherein the imaging window support, the imaging window, or both the imaging window support and the imaging window accommodate an optical interface of a sample dish when the sample dish is mounted on the microscopy system.

6. The microscopy system of claim 5 , wherein the sample dish is mounted on the microscopy system and a distance between the optical interface and the upper working surface is no more than 5 mm.

7. The microscopy system of claim 5 , wherein a tool may rest simultaneously on the optical interface and the upper working surface in a substantially flat orientation.

8. The microscopy system of claim 1 , wherein the sample is laterally accessible to a user during imaging.

9. The microscopy system of claim 1 , wherein the transparent imaging window comprises sapphire and a c-axis of the sapphire is substantially parallel to an optical axis of the microscopy system.

10. The microscopy system of claim 1 , wherein the transparent imaging window has a thickness of no more than 900 μm.

11. The microscopy system of claim 1 , wherein the transparent imaging window has a Young's modulus of at least 100 GPa.

12. The microscopy system of claim 1 , wherein the transparent imaging window is at least as hard or harder than stainless steel.

13. The microscopy system of claim 1 , wherein the transparent imaging window comprises at least one of glass, quartz, sapphire, and plastic.

14. The microscopy system of claim 1 , wherein the transparent imaging window is free standing over a surface that is at least as large as a field of view for the microscopy system.

15. The microscopy system of claim 1 , wherein the microscopy system is a Class 1 laser product according to an ANSI standard, a Class 1 laser product according to an IEC standard, or a Class 1 laser product according to an ANSI standard and a Class 1 laser product according to an IEC standard.

16. The microscopy system of claim 1 , wherein the imaging window support base provides a watertight seal with the upper working surface and the imaging window support.

17. The microscopy system of claim 1 , wherein an optical interface of a sample dish is disposed in contact with only the transparent imaging window and the transparent imaging window comprises an anti-reflection coating disposed between the transparent imaging window and the optical interface.

18. The microscopy system of claim 1 , comprising a kinematic support structure, the kinematic support structure supporting the transparent imaging window such that the transparent imaging window can be adjusted relative to optics of the imaging system.

19. The microscopy system of claim 1 , comprising a sample dish comprising an optical interface, wherein the sample dish is mounted such that the sample dish contacts only the transparent imaging window and only by the optical interface.

20. The microscopy system of claim 5 , wherein the sample dish remains in a fixed position during imaging.

21. The microscopy system of claim 1 , wherein the transparent imaging window remains in a fixed position during imaging.

22. The microscopy system of claim 1 , wherein the imaging window support base is semi-rigid or flexible such that the imaging window support base can be deformed to allow for small movements of the imaging window support.

23. The microscopy system of claim 1 , wherein the sample is disposed on or over the transparent imaging window such that a large zone above and around the sample remains unobstructed during imaging.

24. The microscopy system of claim 1 , wherein the sample is disposed on or over the transparent imaging window and is not inserted into any cartridge, container, or holder.

25. The microscopy system of claim 1 , wherein the housing isolates any moving parts of the microscopy system used to image from users of the system.

26. The microscopy system of claim 1 , wherein the housing is constructed such that there is a large zone above and around the sample that is unobstructed during imaging of the sample.

27. The microscopy system of claim 1 , wherein the housing is constructed to provide lateral access to the sample during imaging of the sample.

28. The microscopy system of claim 1 , wherein the system is constructed such that the sample remains motionless during imaging of the sample.

The microscopy system of claim 1 , comprising imaging optics, wherein the imaging optics are disposed below the transparent imaging window such that illumination light is transmitted from the imaging optics through the imaging window to the sample and back-emitted light is transmitted from the sample through the imaging window to the imaging optics during imaging of the sample.

30. A microscopy system for imaging of samples, comprising:

an exposed transparent imaging window, the transparent imaging window providing a surface onto or over which a sample may be disposed in order to image the sample; and

a housing that at least partially encloses the microscopy system, wherein the housing comprises:

an upper working surface,

an imaging window support base that is recessed below the upper working surface, and

an imaging window support disposed on the imaging window support base,

wherein the transparent imaging window is disposed on the imaging window support,

wherein the imaging window is disposed at least partially above the imaging window support, and

wherein the surface of the imaging window is disposed more than 10 micrometers, and no more than 1 mm, above an upper surface of the imaging window support.

31. The microscopy system of claim 30 , wherein the imaging window support extends laterally beyond the imaging window.
