Abstract

The present disclosure provides treprostinil derivatives that can act as prodrugs of treprostinil. The treprostinil derivatives can be used to treat any conditions responsive to treatment with treprostinil, including pulmonary hypertension, such as pulmonary arterial hypertension.

Claims (43)

1. A method of promoting vasodilation, inhibiting platelet activation or aggregation, inhibiting thrombus formation, stimulating thrombolysis, inhibiting atherogenesis, inhibiting cell proliferation, promoting endothelial cell membrane remodeling, reducing inflammation, or providing cytoprotection, comprising administering to a subject in need of treatment a therapeutically effective amount of a compound of Formula (I):

wherein:

R 1 and R 2 independently are hydrogen,

wherein:

R 6 in each occurrence independently is hydrogen, R 3 , —C(═O)R 3 , —C(═O)OR 3 or —C(═O)NR 9 R 10 ;

R 3 in each occurrence independently is alkyl, haloalkyl, -alkylaryl, cycloalkyl, heterocyclyl, aryl or heteroaryl, each of which may optionally be substituted;

R 9 and R 10 in each occurrence independently are hydrogen, alkyl, -alkylaryl, cycloalkyl, heterocyclyl, aryl or heteroaryl; or

R 9 and R 10 and the nitrogen atom to which they are connected form a heterocyclic or heteroaryl ring;

j in each occurrence independently is an integer from 0 to 4; and

t in each occurrence independently is an integer from 1 to 9;

with the proviso that:

both R 1 and R 2 are not hydrogen; and

R 1 or R 2 is not

or a pharmaceutically acceptable salt, solvate, hydrate, clathrate, polymorph or stereoisomer thereof.

2. The method of claim 1 , wherein for the compound:

R 6 in each occurrence independently is hydrogen or R 3 ;

R 3 in each occurrence independently is C 1 -C 6 alkyl;

R 9 and R 10 in each occurrence independently are C 1 -C 6 alkyl, or R 9 and R 10 and the nitrogen atom to which they are connected form a 3- to 6-membered heterocyclic ring;

j in each occurrence independently is 0 or 1; and

t in each occurrence independently is 1 or 2.

3. The method of claim 1 , wherein for the compound R 1 and R 2 independently are selected from hydrogen,

with the proviso that both R 1 and R 2 are not hydrogen.

4. The method of claim 1 , wherein for the compound R 2 is hydrogen and —OR 1 is derivatized.

5. The method of claim 1 , wherein for the compound R 1 is hydrogen and —OR 2 is derivatized.

6. The method of claim 1 , wherein for the compound both —OR 1 and —OR 2 are derivatized, optionally with the same group.

7. The method of claim 1 , wherein the compound is administered orally.

8. The method of claim 1 , wherein the compound is administered parenterally.

9. The method of claim 1 , wherein the compound is administered subcutaneously or intravenously.

10. The method of claim 1 , wherein the compound is administered topically.

11. The method of claim 10 , wherein the compound is administered transdermally or by oral inhalation.

12. A method of promoting vasodilation, inhibiting platelet activation or aggregation, inhibiting thrombus formation, stimulating thrombolysis, inhibiting atherogenesis, inhibiting cell proliferation, promoting endothelial cell membrane remodeling, reducing inflammation, or providing cytoprotection, comprising administering to a subject in need of treatment a therapeutically effective amount of a compound of Formula (II):

wherein:

ring A is cyclopropyl, cyclobutyl, cyclopentyl or cyclohexyl; and

u is an integer from 0 to 9;

or a pharmaceutically acceptable salt, solvate, hydrate, clathrate, polymorph or stereoisomer thereof.

13. The method of claim 12 , wherein for the compound ring A is cyclopropyl and u is an integer from 0 to 9 or from 0 to 5.

14. The method of claim 13 , wherein the compound is

or a pharmaceutically acceptable salt, solvate, hydrate, clathrate, polymorph or stereoisomer thereof.

15. The method of claim 12 , wherein the compound is administered orally.

16. The method of claim 12 , wherein the compound is administered parenterally.

17. The method of claim 16 , wherein the compound is administered subcutaneously or intravenously.

18. The method of claim 12 , wherein the compound is administered topically.

19. The method of claim 18 , wherein the compound is administered transdermally or by oral inhalation.
