Abstract

Self-assembled gel compositions including a gelator, e.g., an enzyme-cleavable gelator, e.g., having a molecular weight of 2500 or less, are described. The self-assembled gel compositions can encapsulate one or more agents. Methods of making the self-assembled gel compositions, and methods of drug delivery using the self-assembled gel compositions are also described.

Claims (19)

1. A self-assembled xerogel comprising:

(i) one or more enzyme-cleavable amphiphilic gelators having a molecular weight of 2,500 or less selected from the group consisting of triglycerol monoalkanoates, ascorbyl alkanoates, sorbitan alkanoates, sucrose alkanoates, and combinations thereof, and

(ii) one or more agents,

wherein the one or more enzyme-cleavable amphiphilic gelators self-assemble in solution into lamellae, the lamellae forming nanostructures encapsulating the one or more agents in solution, wherein the nanostructures are dried to form the self-assembled xerogel.

2. The self-assembled xerogel of claim 1 , wherein the nanostructures are in the form of fibers and/or sheets.

The self-assembled xerogel of claim 1 , wherein the one or more enzyme-cleavable amphiphilic gelators comprise hydrophobic portions located in the inner regions of the lamellae, and the one or more enzyme-cleavable amphiphilic gelators comprise hydrophilic portions located at the outer surfaces of the lamellae.

4. The self-assembled xerogel of claim 1 , wherein the one or more enzyme-cleavable amphiphilic gelators comprise an ascorbyl alkanoate, a sucrose alkanoate, or a combination thereof.

5. The self-assembled xerogel of claim 1 , wherein the one or more enzyme-cleavable amphiphilic gelators comprise a triglycerol monoalkanoate, a sorbitan alkanoate, or a combination thereof.

The self-assembled xerogel of claim 5 , wherein the triglycerol monoalkanoate comprises a triglycerol monostearate, a triglycerol monopalmitate, a triglycerol monodecanoate, a triglycerol monolaurate, a triglycerol monocaprylate, a triglycerol monomyristate, a triglycerol monooleate, or a combination thereof.

7. The self-assembled xerogel of claim 4 , wherein the ascorbyl alkanoate comprises an ascorbyl palmitate, an ascorbyl decanoate, an ascorbyl laurate, an ascorbyl caprylate, an ascorbyl myristate, an ascorbyl oleate, or a combination thereof.

8. The self-assembled xerogel of claim 1 , wherein the one or more agents comprise an immune-suppressant, a steroid, an anti-inflammatory agent, or a chemotherapeutic.

9. The self-assembled xerogel of claim 8 , wherein the one or more agents comprise an anti-inflammatory agent.

10. The self-assembled xerogel of claim 8 , wherein the one or more agents comprise triamcinolone acetonide, dexamethasone, ethambutol, indomethacin, camptothecin, paclitaxel, temozolomide, carmustine, a PARP-inhibitor, or a combination thereof.

11. The self-assembled xerogel of claim 8 , wherein the one or more enzyme-cleavable amphiphilic gelators comprise a triglycerol monoalkanoate and the one or more agents comprise an immune-suppressor or an anti-inflammatory agent.

12. The self-assembled xerogel of claim 11 , wherein the triglycerol monoalkanoate comprises a triglycerol monostearate.

13. The self-assembled xerogel of claim 12 , wherein the anti-inflammatory agent comprises triamcinolone acetonide.

14. The self-assembled xerogel of claim 8 , wherein the one or more enzyme-cleavable amphiphilic gelators comprise an ascorbyl alkanoate and the one or more agents comprise a steroid.

15. The self-assembled xerogel of claim 14 , wherein the ascorbyl alkanoate comprises ascorbyl palmitate.

16. The self-assembled xerogel of claim 15 , wherein the steroid comprises dexamethasone.
