Claims (46)

1. A polypeptide comprising a first immunoglobulin (Ig)-binding domain, wherein the first Ig-binding domain comprises:

(1) the amino acid sequence that is at least 90% identical to SEQ ID NO: 1, wherein the amino acid that corresponds to S44 of SEQ ID NO: 1 is hydrophobic;

(2) the amino acid sequence that is at least 90% identical to SEQ ID NO: 2, wherein the amino acid that corresponds to S43 of SEQ ID NO: 2 is hydrophobic;

(3) the amino acid sequence that is at least 90% identical to SEQ ID NO: 3, wherein the amino acid that corresponds to S41 of SEQ ID NO: 3 is hydrophobic;

(4) the amino acid sequence that is at least 90% identical to SEQ ID NO: 4, wherein the amino acid that corresponds to S41 of SEQ ID NO: 4 is hydrophobic;

(5) the amino acid sequence that is at least 90% identical to SEQ ID NO: 5, wherein the amino acid that corresponds to S41 of SEQ ID NO: 5 is hydrophobic; or

(6) the amino acid sequence that is at least 90% identical to SEQ ID NO: 6, wherein the amino acid that corresponds to S41 of SEQ ID NO: 6 is hydrophobic.

2. The polypeptide of claim 1 , wherein the amino acid that corresponds to N55 of SEQ ID NO: 1, N54 of SEQ ID NO: 2, N52 of SEQ ID NO: 3, N52 of SEQ ID NO: 4, N52 of SEQ ID NO: 5 or N52 of SEQ ID NO: 6 is hydrophobic.

3. The polypeptide of claim 1 , wherein the first Ig-binding domain comprises one of the following amino acid sequences:

(1) the amino acid sequence that is identical to SEQ ID NO: 1, except that both of the amino acids at position 44 and position 55 of SEQ ID NO: 1 are hydrophobic;

(2) the amino acid sequence that is identical to SEQ ID NO: 2, except that both of the amino acids at position 43 and position 54 of SEQ ID NO: 2 are hydrophobic;

(3) the amino acid sequence that is identical to SEQ ID NO: 3, except that both of the amino acids at position 41 and position 52 of SEQ ID NO: 3 are hydrophobic;

(4) the amino acid sequence that is identical to SEQ ID NO: 4, except that both of the amino acids at position 41 and position 52 of SEQ ID NO: 4 are hydrophobic;

(5) the amino acid sequence that is identical to SEQ ID NO: 5, except that both of the amino acids at position 41 and position 52 of SEQ ID NO: 5 are hydrophobic; and

(6) the amino acid sequence that is identical to SEQ ID NO: 6, except that both of the amino acids at position 41 and position 52 of SEQ ID NO: 6 are hydrophobic.

4. The polypeptide of claim 1 , wherein the amino acid that corresponds to S44 of SEQ ID NO: 1, S43 of SEQ ID NO: 2, S41 of SEQ ID NO: 3, S41 of SEQ ID NO: 4, S41 of SEQ ID NO: 5 or S41 of SEQ ID NO: 6 is Ala, Val, Ile, Leu, Met, Phe, Try, or Trp.

5. The polypeptide of claim 1 , wherein the amino acid that corresponds to S44 of SEQ ID NO: 1, S43 of SEQ ID NO: 2, S41 of SEQ ID NO: 3, S41 of SEQ ID NO: 4, S41 of SEQ ID NO: 5 or S41 of SEQ ID NO: 6 is Ala or Val.

6. The polypeptide of claim 1 , wherein the amino acid that corresponds to N55 of SEQ ID NO: 1, N54 of SEQ ID NO: 2, N52 of SEQ ID NO: 3, N52 of SEQ ID NO: 4, N52 of SEQ ID NO: 5 or N52 of SEQ ID NO: 6 is Ala or Val.

The polypeptide of claim 1 , wherein the amino acid that corresponds to S44 of SEQ ID NO: 1, S43 of SEQ ID NO: 2, S41 of SEQ ID NO: 3, S41 of SEQ ID NO: 4, S41 of SEQ ID NO: 5 or S41 of SEQ ID NO: 6 is Ala and the amino acid that corresponds to N55 of SEQ ID NO: 1, N54 of SEQ ID NO: 2, N52 of SEQ ID NO: 3, N52 of SEQ ID NO: 4, N52 of SEQ ID NO: 5 or N52 of SEQ ID NO: 6 is Val.

8. The polypeptide of claim 1 , wherein the amino acid sequence comprises a sequence that is at least 90% identical to the sequence of SEQ ID NO: 11, 30, or 32.

9. The polypeptide of claim 1 , wherein the polypeptide further comprises a second Ig-binding domain,

wherein the second Ig-binding domain comprises

(1) the amino acid sequence that is at least 90% identical to SEQ ID NO: 1, wherein the amino acid that corresponds to S44 of SEQ ID NO: 1 is hydrophobic;

(2) the amino acid sequence that is at least 90% identical to SEQ ID NO: 2, wherein the amino acid that corresponds to S43 of SEQ ID NO: 2 is hydrophobic;

(3) the amino acid sequence that is at least 90% identical to SEQ ID NO: 3, wherein the amino acid that corresponds to S41 of SEQ ID NO: 3 is hydrophobic;

(4) the amino acid sequence that is at least 90% identical to SEQ ID NO: 4, wherein the amino acid that corresponds to S41 of SEQ ID NO: 4 is hydrophobic;

(5) the amino acid sequence that is at least 90% identical to SEQ ID NO: 5, wherein the amino acid that corresponds to S41 of SEQ ID NO: 5 is hydrophobic; or

(6) the amino acid sequence that is at least 90% identical to SEQ ID NO: 6, wherein the amino acid that corresponds to S41 of SEQ ID NO: 6 is hydrophobic.

10. The polypeptide of claim 9 , wherein the sequence of the first Ig-binding domain and the second Ig-binding domain are identical.

11. The polypeptide of claim 9 , wherein the sequence of the first Ig-binding domain and the second Ig-binding domain are different.

12. The polypeptide of claim 9 , wherein the first Ig-binding domain and the second Ig-binding domain are separated by a spacer domain.

13. The polypeptide of claim 1 , wherein the polypeptide further comprises a second Ig-binding domain, a third Ig-binding domain, a fourth Ig-binding domain, and a fifth Ig-binding domain,

wherein each Ig-binding domain comprises an amino acid sequence that is at least 90% identical to SEQ ID NO: 6, and both of the amino acids that correspond to S41 and N52 of SEQ ID NO: 6 in each Ig-binding domain are hydrophobic.

14. A vector comprising a polynucleotide encoding the polypeptide of claim 1 .

15. A chromatography ligand comprising the polypeptide of claim 1 .

16. A polypeptide comprising an immunoglobulin (Ig)-binding domain, wherein the Ig-binding domain comprises:

(1) the amino acid sequence that is at least 90% identical to SEQ ID NO: 1, wherein the amino acid that corresponds to N55 of SEQ ID NO: 1 is Val;

(2) the amino acid sequence that is at least 90% identical to SEQ ID NO: 2, wherein the amino acid that corresponds to N54 of SEQ ID NO: 2 is Val;

(3) the amino acid sequence that is at least 90% identical to SEQ ID NO: 3, wherein the amino acid that corresponds to N52 of SEQ ID NO: 3 is Val;

(4) the amino acid sequence that is at least 90% identical to SEQ ID NO: 4, wherein the amino acid that corresponds to N52 of SEQ ID NO: 4 is Val;

(5) the amino acid sequence that is at least 90% identical to SEQ ID NO: 5, wherein the amino acid that corresponds to N52 of SEQ ID NO: 5 is Val; or

(6) the amino acid sequence that is at least 90% identical to SEQ ID NO: 6, wherein the amino acid that corresponds to N52 of SEQ ID NO: 6 is Val,

wherein the amino acid that corresponds to S44 of SEQ ID NO: 1, S43 of SEQ ID NO: 2, S41 of SEQ ID NO: 3, S41 of SEQ ID NO: 4, S41 of SEQ ID NO: 5 or S41 of SEQ ID NO: 6 is hydrophobic.

17. The polypeptide of claim 16 , wherein the amino acid that corresponds to S44 of SEQ ID NO: 1, S43 of SEQ ID NO: 2, S41 of SEQ ID NO: 3, S41 of SEQ ID NO: 4, S41 of SEQ ID NO: 5 or S41 of SEQ ID NO: 6 is Ala, Val, Ile, Leu, Met, Phe, Try, or Trp.

18. A polypeptide comprising an immunoglobulin (Ig)-binding domain, wherein the Ig-binding domain comprises SEQ ID NO: 6 with a mutation at S41, wherein S41 is substituted with a hydrophobic amino acid residue.

19. The polypeptide of claim 18 , wherein the Ig-binding domain further comprises a mutation at N52, wherein N52 is substituted with a hydrophobic amino acid residue.
