Viral vector stabilization
Combining viral vector with surfactant preserves vector infectivity, and surfactant provided an unexpected benefit by protecting viral vector from damage due to transient elevated temperature.
1. A method of preserving the infectivity of a recombinant adenovirus, comprising: mixing the recombinant adenovirus comprising a transgene and having infectivity with a surfactant to form an admixture; removing substantially all water from the admixture, and storing the admixture at or about 4° C. or room temperature for at least about one week, wherein after the storing step the adenovirus retains a higher level of infectivity relative to control, and wherein the surfactant has Structure I:
2. The method of claim 1 , wherein the adenovirus comprises attenuated adenovirus.
3. The method of claim 1 , wherein the transgene encodes interferon.
4. The method of claim 1 , wherein the transgene encodes interferon alpha-2b.
5. The method of claim 1 , wherein the admixture is stored at about 4° C. or room temperature for at least about three months.
6. The method of claim 1 , wherein the admixture is stored at about 4° C. or room temperature for at least about twelve months.
7. The method of claim 1 , wherein the adenovirus is replication-deficient and cannot replicate in normal human cells.
8. The method of claim 1 , wherein the adenovirus is a type 5 adenovirus.
