Abstract

The present invention relates to novel humanized anti-IL-4 and IL-13 antibodies and fragments thereof and novel bispecific antibodies and fragments thereof that specifically bind to IL-4 and IL-13. The invention also includes uses of the antibodies to treat or prevent IL-4 and/or IL-13 mediated diseases or disorders, including allergic asthma and dermatitis.

Claims (12)

1. An isolated humanized antibody or antibody fragment thereof that specifically binds IL-13, wherein the antibody or antibody fragment thereof comprises a light chain variable domain comprising CDRs having the amino acid sequences RASESVDSYGQSYMH (SEQ ID NO: 8), LASNLES (SEQ ID NO: 9), and QQNAEDSRT (SEQ ID NO: 10) and a heavy chain variable domain comprising CDRs having the amino acid sequences GFSLTDSSIN (SEQ ID NO: 11), DGRID (SEQ ID NO: 12), and DGYFPYAMDF (SEQ ID NO: 13).

The antibody or antibody fragment thereof of claim 1 , wherein the light chain variable domain comprises an amino acid sequence at least 95% identical to the amino acid sequence of SEQ ID NO:1 and the heavy chain variable domain comprises an amino acid sequence at least 95% identical to the amino acid sequence of SEQ ID NO:2.

3. The antibody or antibody fragment thereof of claim 1 , wherein the light chain variable domain comprises the amino acid sequence of SEQ ID NO:1 and a heavy chain domain comprises the amino acid sequence of SEQ ID NO:2.

The antibody or antibody fragment thereof of claim 1 , wherein the antibody or antibody fragment is an antibody comprising a heavy chain and a light chain, wherein the heavy chain comprises CH1, CH2 and CH3 domains of a constant region and the light chain comprises a CL domain of a constant region.

5. The antibody or antibody fragment thereof of claim 4 , wherein the antibody is an IgG4 antibody.

6. A pharmaceutical composition comprising the antibody or antibody fragment of claim 1 and a pharmaceutically acceptable carrier.

7. The antibody or antibody fragment thereof of claim 1 , wherein the antibody or antibody fragment is conjugated to a second component.

8. The antibody or antibody fragment thereof of claim 7 , wherein the second component is a cell-binding agent, a cytotoxic agent, a radioisotope, an enzyme substrate, a fluorophore, a chromophore, an imaging agent or a metal ion.

9. A kit comprising the antibody or antibody fragment thereof of claim 1 .

10. The kit of claim 9 which further comprises instructions for use.

11. An article of manufacture containing the antibody fragment thereof of claim 1 .

12. The article of manufacture of claim 11 , wherein the container is an intravenous solution bag or a vial having a stopper pierceable by a hypodermic injection needle.
