Engineered enzymes
The present disclosure provides engineered RNA-guided enzymes for editing live cells.
1. An engineered nucleic acid-guided nuclease with a lowered cutting activity relative to the nucleic acid-guided nuclease having the sequence of SEQ ID No. 1, wherein the engineered nucleic acid-guided nuclease has a sequence comprising any of SEQ ID Nos. 8, 9, 10 or 15.
2. The engineered nucleic acid-guided nuclease of claim 1 comprising SEQ ID No. 8.
3. The engineered nucleic acid-guided nuclease of claim 1 comprising SEQ ID No. 9.
4. The engineered nucleic acid-guided nuclease of claim 1 comprising SEQ ID No. 10.
5. The engineered nucleic acid-guided nuclease of claim 1 comprising SEQ ID No. 15.
6. An enzyme cocktail comprising an engineered nucleic acid-guided nuclease of claim 1 .
7. An enzyme cocktail comprising an engineered nucleic acid-guided nuclease of claim 1 and SEQ ID No. 2.
8. An enzyme cocktail comprising an engineered nucleic acid-guided nuclease of claim 1 and SEQ ID No. 3.
9. An enzyme cocktail comprising an engineered nucleic acid-guided nuclease of claim 1 and SEQ ID No. 11.
10. An enzyme cocktail comprising an engineered nucleic acid-guided nuclease of claim 1 and SEQ ID No. 12.
