Abstract

The present invention relates to IL-17 Receptor A (IL-17RA or IL-17R) antigen binding proteins, such as antibodies, polynucleotide sequences encoding said antigen binding proteins, and compositions and methods for diagnosing and treating diseases mediated by IL-17 Receptor A activation by one or more IL-17 ligands. The present invention relates to the identification of neutralizing determinants on IL-17 Receptor A (IL-17RA or IL-17R) and antibodies that bind thereto. Aspects of the invention also include antibodies that compete for binding with the IL-17RA neutralizing antibodies described herein.

Claims (7)

1. A method of treating scleroderma in a human patient in need thereof, which method comprises administering to the patient an effective amount of human monoclonal antibody, or an antigen-binding fragment thereof, that specifically binds the human IL-17 receptor A, wherein the human monoclonal antibody comprises:

(a) a heavy chain CDR1 comprising the amino acid sequence of SEQ ID NO: 146, a heavy chain CDR2 comprising the amino acid sequence of SEQ ID NO: 147, a heavy chain CDR3 comprising the amino acid sequence of SEQ ID NO: 148, and a light chain CDR1 comprising the amino acid sequence of SEQ ID NO: 224, a light chain CDR2 comprising the amino acid sequence of SEQ ID NO: 225, and a light chain CDR3 comprising the amino acid sequence of SEQ ID NO: 226; or

(b) a heavy chain variable domain comprising the amino acid sequence of SEQ ID NO: 14 and a light chain variable domain comprising the amino acid sequence of SEQ ID NO: 40,

whereby the human monoclonal antibody specifically binds to human IL-17 receptor A and inhibits the binding of IL-17A to IL-17 receptor A, and the scleroderma is treated in the human patient.

The method of claim 1 , wherein the human monoclonal antibody or antigen-binding fragment thereof is selected from a single chain antibody, a diabody, a triabody, a tetrabody, a Fab fragment, a F(ab′)2 fragment, an IgD antibody, an IgE antibody, an IgM antibody, an IgG1 antibody, an IgG2 antibody, an IgG3 antibody; and an IgG4 antibody.

3. The method of claim 1 , wherein the human monoclonal antibody is administered to the human patient in the form of a composition comprising a pharmaceutically acceptable carrier.

4. The method of claim 3 , wherein the composition is administered to the patient subcutaneously, intramuscularly, or intravenously.
