Abstract

Provided are compositions, including isolated, synthetic or recombinant peptides for: expanding regulatory T cells (Treg) populations; for treating or ameliorating a vascular inflammation, and Kawasaki disease (KD) or a pediatric acute vasculitis of the coronary arteries, including vascular coronary abnormalities, and acute or chronic vascular inflammatory abnormalities, and method for making and using them. Provided are immunotherapies for promoting expansion of natural, Treg to establish, or re-establish, vascular homeostasis; or, for ameliorating: a disease or condition associated with an autoimmune disease or condition; as immune-mediated vascular disorder; a disease or condition treated with intravenous immunoglobulin (IVIG) therapy; a vascular coronary abnormality; an acute or a chronic vasculitis; an autoimmune vasculitis; Kawasaki disease (KD) or a pediatric acute vasculitis of the coronary arteries; atherosclerosis; preventing miscarriage in autoimmune women; rheumatoid arthritis or Juvenile Idiopathic Arthritis; a neoplastic hematological disorder, or a leukemia.

Claims (67)

1. A method for the treatment, prevention or amelioration of rheumatoid arthritis or juvenile idiopathic arthritis, comprising: administering to an individual in need thereof an effective amount of a combination of isolated, synthetic or recombinant peptides or polypeptides comprising:

thereby: or treating, preventing or ameliorating the rheumatoid arthritis or juvenile idiopathic arthritis.

2. The method of claim 1 , wherein the combination of isolated, synthetic or recombinant peptides or polypeptides is administered:

via an enteral or a parenteral route, or orally, subcutaneously, or systemically or topically; or

as a controlled release formulation, or formulated in a tablet, a pill, a gel, a patch, in an implant or in a spray, as an inhaled powder.

3. The method of claim 1 , wherein the combination of isolated, synthetic or recombinant peptides or polypeptides is administered:

parenterally by one bolus injection, or by two, three or four or more bolus injections, or by gradual perfusion over time;

at intervals of 1 week, 2 weeks, 4 weeks (or one month), 6 weeks, 8 weeks or one year; or

at a daily dose of peptide in a range of about 10 nanograms to 10 milligrams, or about 1 microgram to 10 milligrams.

The method of claim 1 , wherein the combination of isolated, synthetic or recombinant peptides or polypeptides is administered as a prophylactic or preventative measure to an individual having a personal history of, or to an individual having a detected genotype or phenotype indicating a predisposition to rheumatoid arthritis or juvenile idiopathic arthritis.

5. The method of claim 1 , wherein administration of the combination of isolated, synthetic or recombinant peptides or polypeptides to the individual generates natural regulatory T cells (nTregs) that suppress pro-inflammatory T cells and a pro-inflammatory T cell response.

6. The method of claim 5 , wherein administration of the combination of isolated, synthetic or recombinant peptides or polypeptides to the individual generates natural regulatory T cells (nTregs) suppress pro-inflammatory T cells and pro-inflammatory T cell responses.

7. The method of claim 1 , wherein the individual is a human.

The method of claim 1 , wherein any one of the isolated, synthetic or recombinant peptide or polypeptide of the combination of isolated, synthetic or recombinant peptides or polypeptides has at least one conservative amino acid substitution and retains its property of generating or retains its ability to generate natural regulatory T cells (nTregs) that can suppress pro-inflammatory T cells and pro-inflammatory T cell responses when administered to an individual.

9. The method of claim 8 , wherein the at least one conservative amino acid substitution comprises:

substituting an amino acid with another amino acid of like characteristics;

a replacement of an aliphatic amino acid with another aliphatic amino acid;

a replacement of a Serine with a Threonine or vice versa;

a replacement of an acidic residue with another acidic residue;

a replacement of a residue bearing an amide group with another residue bearing an amide group; an exchange of a basic residue with another basic residue; or

a replacement of an aromatic residue with another aromatic residue.

10. The method of claim 1 , wherein any one of the isolated, synthetic or recombinant peptides or polypeptides of the combination of isolated, synthetic or recombinant peptides or polypeptides is formulated as a pharmaceutical composition or a formulation.

The method of claim 1 , wherein any one of the isolated, synthetic or recombinant peptides or polypeptides of the combination of isolated, synthetic or recombinant peptides or polypeptides is formulated in a sterile solution for injection, or as a powder or a lyophilized or a freeze-dried composition.

12. The method of claim 1 , wherein any one of the polypeptide or peptides or polypeptides of the combination of isolated, synthetic or recombinant peptides or polypeptides is formulated in or as a liposome or a nanoparticle.

13. The method of claim 1 , wherein any one of the polypeptide or peptides or polypeptides of the combination of isolated, synthetic or recombinant peptides or polypeptides is formulated in or as a vaccine.

14. The method of claim 1 , wherein the method is for the treatment, prevention or amelioration of rheumatoid arthritis.

15. The method of claim 1 , wherein the method is for the treatment, prevention or amelioration of juvenile idiopathic arthritis.

16. The method of claim 1 , wherein the combination of isolated, synthetic or recombinant peptides or polypeptides consists of:

17. The method of claim 1 , wherein the combination of isolated, synthetic or recombinant peptides or polypeptides is administered as a vaccine with an adjuvant.

18. The method of claim 9 , wherein the peptide or polypeptide, or at least one amino acid in the peptide or polypeptide, comprises a peptidomimetic or a non-natural amino acid residue.

19. The method of claim 10 , wherein the pharmaceutical composition or formulation comprises a pharmaceutically acceptable excipient.

20. The method of claim 1 , wherein the combination of isolated, synthetic or recombinant peptides or polypeptides is administered via a parenteral route comprising a subcutaneous, an intravenous (IV), an intradermal, an intramuscular, an intraperitoneal, an intranasal, a transdermal or a buccal route;

intradermally as a sterile formulation.
