Abstract

A system for acquiring data regarding a wound in tissue comprises at least one excitation light source configured to directly illuminate a wound with excitation light; a spectral filtering mechanism configured to permit passage of optical signals responsive to illumination of the wound and having at least one wavelength corresponding to bacterial, fungal, and/or other microorganism autofluorescence and/or bacterial, fungal, viral, and/or microbial fluorescence, the spectral filtering mechanism including a plurality of selectable filters respectively corresponding to different discrete spectral bandwidths; an optical sensor configured to detect the spectrally filtered signals; and a processor configured to receive the detected, filtered signals and to identify a fluorescent signature of bacteria in the wound based at least in part on the detected, filtered signals and to output data regarding the bacterial, fungal, viral, and/or microbial fluorescent signature.

Claims (33)

1. A handheld, portable imaging system for acquiring data regarding a wound in tissue, comprising:

at least one excitation light source configured to illuminate a wound with excitation light and comprising at least one wavelength or wavelength band causing at least one biomarker in the illuminated wound to fluoresce;

a spectral filtering mechanism configured to permit passage of optical signals responsive to illumination of the wound and having a wavelength corresponding to bacterial autofluorescence and/or bacterial fluorescence, the spectral filtering mechanism including a plurality of selectable filters respectively corresponding to different discrete spectral bandwidths;

a mobile communication device having an image sensor to detect the spectrally filtered signals, wherein the mobile communication device is a cellular telephone, a smartphone, a laptop computer, a tablet computer, or a personal digital assistant,

wherein the plurality of selectable filters is movable relative to the image sensor so as to position a selected filter of the plurality of selectable filters between the wound and the image sensor to detect the spectrally filtered signals corresponding to the discrete spectral bandwidths of the selected filter;

a housing, separate from the mobile communication device, to support the at least one excitation light source and the spectral filtering mechanism relative to the mobile communication device during imaging and configured to receive at least a portion of the mobile communication device therein; wherein the spectral filtering mechanism includes a triple band-pass filter; and

a processor configured to receive the detected, filtered signals, to spatially and/or temporally co-register one or more of endogenous fluorescence data, exogenous fluorescence data, absorbance data, and reflectance data contained in the detected, filtered signals, and to identify a fluorescent signature of bacteria in the wound based at least in part on the data contained in the detected, filtered signals, and to output data regarding the bacterial fluorescent signature.

The system according to claim 1 , further comprising a memory storing fluorescence, absorbance, and/or reflectance spectral data of biological, tissue, cellular, and molecular components and/or biomarkers and non-biological materials for comparison with fluorescence, absorbance, and/or reflectance detected in and/or around the wound.

3. The system according to claim 2 , wherein the memory is configured to store the fluorescence, absorbance, and/or reflectance spectral data of biological, tissue, cellular, and molecular components and/or biomarkers and non-biological materials in a look-up table.

The system according to claim 1 , wherein the spectral filtering mechanism is further configured to permit passage of optical signals responsive to illumination of the wound and having a wavelength corresponding to tissue autofluorescence and wherein the output data comprises at least one fluorescent representation of bacteria and tissue components present in the wound.

5. The system according to claim 4 , wherein the processor is configured to analyze the at least one fluorescent representation in order to recognize, classify, and/or quantify various components of the wound, based on received signals or the output data.

6. The system according to claim 1 , wherein the image sensor is configured to detect the spectrally filtered signals a plurality of times at predetermined intervals.

7. The system according to claim 1 , further comprising a wireless data transfer port configured to transmit the data regarding the bacterial fluorescent signature to an external device.

8. The system according to claim 7 , wherein the data transfer port is configured to communicate via a Bluetooth protocol.

9. The system according to claim 1 , wherein the at least one excitation light source is configured to emit excitation light having a wavelength of about 400 nm to about 450 nm.

10. The system according to claim 1 , wherein the spectral filtering mechanism is configured to permit passage of optical signals having wavelengths within one or more of the following ranges: 405 nm±25 nm, 546 nm±25 nm, 600 nm±25 nm, about 490 nm to about 550 nm, and about 590 nm to about 650 nm.

11. The system according to claim 1 , further comprising a white-light source for white-light imaging.

The system according to claim 1 , wherein the at least one excitation light source includes a first excitation light source configured to emit excitation light having a first wavelength or wavelength band and a second excitation light source configured to emit excitation light having a second wavelength or wavelength band, the first wavelength or wavelength band being different than the second wavelength or wavelength band.

13. The system according to claim 1 , wherein the spectral filtering mechanism further comprises one or more polarization filters.

14. The system according to claim 1 , wherein the at least one excitation light source comprises at least one polarization filter.

The system according to claim 1 , wherein the at least one excitation light source is a plurality of excitation light sources, and respective ones of the plurality of excitation light sources are configured to emit light in an ultraviolet, visible, far-red, near infrared, and/or infrared range.

16. The system according to claim 1 , wherein the processor is a processor of the mobile communication device.

17. The system according to claim 1 , wherein the plurality of selectable filters respectively corresponding to different discrete spectral bandwidths includes a triple bandpass filter configured to separate narrow bandwidths of blue, green and red reflected light components from the wound.

The system according to claim 17 , wherein the processor is further configured to receive the filtered, reflected light components, and to output an image of the presence of blood and/or microvascular networks in the wound and surround tissues based at least in part on the data contained in the filtered reflected light components.

19. The system according to claim 17 , wherein the processor is further configured to receive the filtered, reflected light components and to output an image indicating the presence of blood containing both oxygenated and deoxygenated hemoglobin.

20. A handheld, portable system for acquiring data regarding a wound in tissue, comprising:

at least one excitation light source configured to illuminate a wound with excitation light and comprising at least one wavelength or wavelength band causing at least one biomarker in the illuminated wound to fluoresce;

a spectral filtering mechanism configured to permit passage of optical signals responsive to illumination of the wound and having a wavelength corresponding to bacterial autofluorescence and/or bacterial fluorescence, the spectral filtering mechanism including a plurality of selectable filters respectively corresponding to different discrete spectral bandwidths;

a mobile communication device having an image sensor to detect the spectrally filtered signals, wherein the mobile communication device is a cellular telephone, a smartphone, a laptop computer, a tablet computer, or a personal digital assistant,

wherein the plurality of selectable filters is movable relative to the image sensor so as to position a selected filter of the plurality of selectable filters between the wound and the image sensor to detect the spectrally filtered signals corresponding to the discrete spectral bandwidths of the selected filter;

a housing, separate from the mobile communication device, to support the at least one excitation light source relative to the mobile communication device during imaging and configured to receive at least a portion of the mobile communication device therein; and

a processor configured to receive the detected, filtered signals, to spatially and/or temporally co-register one or more of endogenous fluorescence data, exogenous fluorescence data, absorbance data, and reflectance data contained in the detected, filtered signals, and to identify a fluorescent signature of bacteria in the wound based at least in part on the data contained in the detected, filtered signals, and to output data regarding the bacterial fluorescent signature, wherein

the processor is further configured to correlate bacterial autofluorescence data and bacterial fluorescence data contained in the detected, spectrally-filtered signals to provide an indication of wound infection.
