Formulation of tocilizumab and method for treating COVID-19 by inhalation
The present invention is directed to formulations containing the anti-IL-6 therapeutic monoclonal antibody tocilizumab and pharmaceutically acceptable excipients that can be administered using a soft mist inhaler or a nebulizer.
1. A pharmaceutical formulation comprising an aqueous solution comprising:
tocilizumab in an amount ranging from about 5 mg/mL to about 30 mg/mL,
histidine in a concentration ranging from about 5 mM to about 25 mM,
arginine in an amount ranging from about 100 mM to about 350 mM,
methionine in an amount ranging from about 10 mM to about 50 mM, and
polysorbate 80 in an amount ranging from about 0.005% to about 3% (w/v),
wherein the pH of the pharmaceutical formulation ranges from about 5 to about 7, and
wherein the formulation is contained within a device for administration by nebulization.
2. A method for administering the pharmaceutical formulation of claim 1 comprising nebulizing a defined amount of the pharmaceutical formulation with an inhaler by using pressure to force the pharmaceutical formulation through a nozzle to form an inhalable aerosol.
3. The method of claim 2 , wherein the defined amount of the pharmaceutical formulation is less than about 70 microliters.
4. The method of claim 3 , wherein the average particle size of the aerosol is less than about 15 microns.
5. A method of treating COVID-19 in a patient comprising administering the pharmaceutical formulation of claim 1 to the patient by inhalation.
