Nucleic acid-polypeptide compositions and methods of inducing exon skipping
Disclosed herein are molecules and pharmaceutical compositions that induce an insertion, deletion, duplication, or alteration in an incorrectly spliced mRNA transcript to induce exon skipping or exon inclusion. Also described herein include methods for treating a disease or disorder that comprises a molecule or a pharmaceutical composition that induces an insertion, deletion, duplication, or alteration in an incorrectly spliced mRNA transcript to induce exon skipping or exon inclusion.
1. A method of inducing exon skipping in a targeted pre-mRNA transcript of DMD gene, comprising:
a) contacting a muscle cell with a phosphorodiamidate morpholino oligonucleotide (PMO) conjugate, wherein the PMO conjugate comprises an anti-transferrin receptor antibody or antigen binding fragment thereof, and a PMO targeting an acceptor splice site, a donor splice site, or an exonic splice enhancer element of the targeted pre-mRNA transcript of the DMD gene; wherein the PMO induces exon skipping in the targeted pre-mRNA transcript, and wherein the PMO conjugate is preferentially delivered into the muscle cell;
b) hybridizing the PMO to the targeted pre-mRNA transcript to induce exon skipping in the targeted pre-mRNA transcript; and
c) translating a mRNA transcript produced from the targeted pre-mRNA transcript processed in step b) in the muscle cell to generate a truncated DMD protein.
2. The method of claim 1 , wherein the PMO induces skipping of exon 8 of the DMD gene.
3. The method of claim 1 , wherein the PMO induces skipping of exon 23 of the DMD gene.
4. The method of claim 1 , wherein the PMO induces skipping of exon 35 of the DMD gene.
5. The method of claim 1 , wherein the PMO induces skipping of exon 43 of the DMD gene.
6. The method of claim 1 , wherein the PMO induces skipping of exon 44 of the DMD gene.
7. The method of claim 1 , wherein the PMO induces skipping of exon 45 of the DMD gene.
8. The method of claim 1 , wherein the PMO induces skipping of exon 53 of the DMD gene.
9. The method of claim 1 , wherein the PMO induces skipping of exon 50 of the DMD gene.
10. The method of claim 1 , wherein the PMO induces skipping of exon 51 of the DMD gene.
11. The method of claim 1 , wherein the PMO induces skipping of exon 52 of the DMD gene.
12. The method of claim 1 , wherein the PMO induces skipping of exon 55 of the DMD gene.
13. The method of claim 1 , wherein the antibody or antigen binding fragment thereof comprises a humanized antibody or antigen binding fragment thereof, chimeric antibody or antigen binding fragment thereof, monoclonal antibody or antigen binding fragment thereof, monovalent Fab′, divalent Fab2, single chain variable fragment (scFv), diabody, minibody, nanobody, single-domain antibody (sdAb), or camelid antibody or antigen binding fragment thereof.
14. The method of claim 1 , wherein the PMO comprises at least from about 10 to about 30 nucleotides in length.
15. The method of claim 1 , wherein the PMO comprises at least 90%, 95%, 99%, or 100% sequence identity to a sequence selected from SEQ ID NOs: 225-227, 252-263, 268-272, 352-427, 768-827, and 939-972.
16. The method of claim 1 , wherein the PMO comprises at least 90%, 95%, 99%, or 100% sequence identity to a sequence selected from SEQ ID NOs: 352-427 and 768-827.
17. The method of claim 1 , wherein the PMO is conjugated to the antibody or antigen binding fragment thereof via a linker.
18. The method of claim 17 , wherein the linker is a cleavable linker.
19. The method of claim 17 , wherein the linker is a non-cleavable linker.
20. The method of claim 17 , wherein the linker is selected from the group consisting of a heterobifunctional linker, a homobifunctional linker, a maleimide group, a dipeptide moiety, a benzoic acid group or derivatives thereof, a C 1 -C 6 alkyl group, and a combination thereof.
21. The method of claim 1 , wherein the PMO conjugate has a PMO to antibody ratio of about 1:1, 2:1, 3:1, or 4:1.
22. The method of claim 1 , wherein the method is an in vivo method.
