OLIGOSACCHARIDES COMPRISING AN AMINOOXY GROUP AND CONJUGATES THEREOF
The invention provides methods for the synthesis of oligosaccharides comprising an aminooxy group. The invention further provides oligosaccharides comprising an aminooxy group, methods for coupling oligosaccharides comprising an aminooxy group to glycoproteins, and oligosaccharide-protein conjugates. Also provided are methods of treating a lysosomal storage disorder in a mammal by administration of an oligosaccharide-protein conjugate.

56 . A method of treating fucosidosis in a patient, comprising administering to the patient (a) an effective amount of an oligosaccharide-acid α-L-fucosidase conjugate, comprising (1) acid α-L-fucosidase, (2) an oligosaccharide, and (3) an oxime group connecting the acid α-L-fucosidase and the oligosaccharide, and (b) an effective amount of an antipyretic, antihistamine, and/or immunosuppressant.

A method of inducing immune tolerance towards an oligosaccharide-protein conjugate in a patient having mucopolysaccharidosis I, comprising administering to the patient an effective amount of cyclosporine A and azathioprine, wherein the oligosaccharide-protein conjugate comprises: (1) α-L-iduronidase, (2) an oligosaccharide, and (3) an oxime group connecting the α-L-iduronidase and the oligosaccharide.
