Abstract

This disclosure relates to deuterated D-serine, pharmaceutically acceptable salts thereof, analogs and prodrugs thereof, pharmaceutical compositions thereof, and methods of use.

Claims (42)

1 . A method of treating a disease or condition selected from Parkinson's disease psychosis (PDP), autism spectrum disorder (ASD), a disorder or symptom associated with a GRIN mutation, dementia, behavioral and psychological symptoms of dementia (BPSD), psychosis, non-motor symptoms of Parkinson's disease (PD), motor symptoms of PD, a sleep disorder and vision impairment, the method comprising administering to a subject in need thereof an effective amount of a compound of Formula I or a pharmaceutical composition comprising a compound of Formula I:

wherein

R 1 is —OH, —OD, —O—C 1-4 alkyl, or an amino acid residue;

R 2 is H, D, —C 1-4 alkyl, —C(O)—C 1-6 alkyl, or —C(O)—C 1-6 hydroxyalkyl;

R 3 is H, D, or an amino acid residue;

R 4 is H or D; and

each of Y 1 , Y 2a and Y 2b is independently H or D, provided that at least one of Y 1 , Y 2a and Y 2b is D; wherein each position designated specifically as deuterium has at least 90% incorporation of deuterium; and wherein the compound of Formula I is at least about 90% stereomerically pure;

or a pharmaceutically acceptable salt thereof;

and a pharmaceutically acceptable carrier.

2 . The method of claim 1 , wherein R 1 or R 3 is a D-D-serine residue.

3 . The method of claim 1 , wherein the compound is a compound of Formula II:

wherein each of Y 1 , Y 2a and Y 2b is independently H or D, provided that at least one of Y 1 , Y 2a and Y 2b is D;

or a pharmaceutically acceptable salt thereof.

4 . The method of claim 1 , wherein Y 1 is D.

5 . The method of claim 1 , wherein Y 2a and Y 2b are each H.

6 . The method of claim 1 , wherein Y 2a and Y 2b are each D.

7 . The method of claim 1 , wherein the compound is selected from Compound 100 and Compound 103:

8 . The method of claim 1 , wherein the compound is Compound 100:

10 . The method of claim 1 , wherein any atom not designated as deuterium is present at its natural isotopic abundance.

12 . The method of claim 1 , wherein the pharmaceutical composition is suitable for oral administration.

13 . The method of claim 1 , wherein the pharmaceutical composition comprises 0.1 g to 60 g of the compound of Formula I.

14 . The method of claim 1 , wherein the pharmaceutical composition further comprises a second agent.

16 . The method of claim 1 , wherein the disease or condition is an autism spectrum disorder (ASD) selected from Asperger's syndrome, autistic disorder, childhood disintegrative disorder and pervasive developmental disorder (PDD or atypical autism).

17 . The method of claim 1 , wherein the disease or condition is a disorder or symptom associated with a GRIN mutation.

18 . The method of claim 17 , wherein the disorder or symptom is associated with a mutation in GRIN1, GRIN2A, GRIN2B, GRIN2C, GRIN2D, GRIN3A or GRIN3B.

20 . The method of claim 1 , wherein the disease or condition is dementia associated with a neurodegenerative disease, infectious disease or vascular disease.

The method of claim 20 , wherein the dementia is associated with Parkinson's disease, Alzheimer's disease, Huntington's disease, Pick's disease, a prion disease such as Creutzfeldt-Jakob Disease, Lewy body disease, amyotrophic lateral sclerosis (ALS), multiple sclerosis, frontotemporal lobar degeneration, corticobasal degeneration, HIV, neuroborreliosis, stroke or chronic oxygen deprivation to the brain (vascular dementia).

28 . The method of claim 1 , wherein the disease or condition is the non-motor symptoms of Parkinson's disease (PD) selected from depression, pain, anxiety, sleep disorders, impulse control disorders, fatigue, apathy, dysphoria, problems with urination, gastrointestinal problems, orthostatic hypotension, sexual problems, excessive perspiration, loss of sense of smell, speech problems, swallowing problems, vision changes and weight changes.

29 . The method of claim 1 , wherein the disease or condition is the motor symptoms of PD selected from rigidity, bradykinesia, resting tremor and postural instability.

30 . The method of claim 1 , wherein the disease or condition is a sleep disorder associated with depression, bipolar depression, schizophrenia, anxiety or Alzheimer's disease.
