Abstract

Methods of administering immunoconjugates that bind to CD37 (e.g., IMGN529) in combination with antibodies that bind to CD20 are provided. The methods comprise administering an anti-CD37 immunoconjugate (e.g., IMGN529) and an anti-CD20 antibody to a person in need thereof, for example, a cancer patient.

Claims (12)

1. A method for treating a human patient having a B-cell cancer comprising administering to said patient in need thereof (i) an immunoconjugate that binds to CD37 comprising a DM1 maytansinoid, a linker, and an antibody that binds to CD37 comprising the variable heavy chain sequence set forth in SEQ ID NO: 12 and the variable light chain sequence set forth in SEQ ID NO: 15; and (ii) rituximab, wherein said immunoconjugate that binds to CD37 is administered at a dose of 0.7 mg/kg once every three weeks and wherein the rituximab is administered at a dose of about 375 mg/m 2 on week 1, day 1 of a three week schedule.

2. The method of claim 1 , wherein the immunoconjugate that binds to CD37 and the rituximab produce a synergistic effect, with a Synergy Score of at least 4 in vitro.

3. The method of claim 1 , wherein the immunoconjugate that binds to CD37 and the rituximab have a combination index value of less than 0.5.

4. The method of claim 1 , wherein the cancer is leukemia or lymphoma.

The method of claim 1 , wherein the cancer is selected from the group consisting of NHL, precursor B-cell lymphoblastic leukemia/lymphoma, B-cell chronic lymphocytic leukemia (CLL)/small lymphocytic lymphoma (SLL), B-cell prolymphocytic leukemia, lymphoplasmacytic lymphoma, mantle cell lymphoma (MCL), follicular lymphoma (FL), cutaneous follicle center lymphoma, marginal zone B-cell lymphoma, hairy cell leukemia, diffuse large B-cell lymphoma (DLBCL), Burkitt's lymphoma, plasmacytoma, plasma cell myeloma, post-transplant lymphoproliferative disorder, Waldenstrom's macroglobulinemia, acute lymphoblastic leukemia (ALL), and anaplastic large-cell lymphoma (ALCL).

6. The method of claim 5 , wherein the cancer is NHL.

7. The method of claim 6 , wherein the NHL is relapsed or refractory NHL.

8. The method of claim 1 , wherein the cancer overexpresses MYC and/or BCL2.

9. The method of claim 1 , wherein the immunoconjugate that binds to CD37 and the rituximab are administered intravenously.

10. The method of claim 1 further comprising administering a growth factor to the patient.

11. The method of claim 1 , wherein the human patient has relapsed diffuse large B-cell lymphoma (DLBCL).

12. The method of claim 1 , wherein the antibody that binds to CD37 comprises the heavy chain sequence set forth in SEQ ID NO: 18 and the light chain sequence set forth in SEQ ID NO: 21.
