Abstract

The present disclosure described herein provides compositions with a membrane-penetrating properties and methods for allowing translocation across a membrane without disruption.

Claims (25)

1. A method of delivering a therapeutic comprising: administering a composition to a subject, the composition comprising: a polypeptide comprising at least one sequence of ABX″C, wherein A is selected from a hydrophobic amino acid or an amide containing backbone, with a nucleic acid side chain; B and C may be the same or different, and are independently selected from arginine, asparagine, glutamine, lysine, and analogs thereof; X is an amide containing backbone, with a nucleic acid side chain; and n is an integer from 1 to 4; and at least one heterologous moiety, wherein the heterologous moiety is the therapeutic, and wherein the composition increases intracellular delivery of the therapeutic as compared to the therapeutic alone.

2. The method of claim 1 , wherein:

the composition is targeted to a specific cell, or a specific tissue; or

(ii) the amide containing backbone is aminoethyl-glycine.

3. The method of claim 1 , wherein the composition crosses a blood-brain-barrier, a placental membrane, or a blood-testis barrier.

4. The method of claim 1 , wherein the composition is administered systemically.

5. The method of claim 1 , wherein the composition has differential pharmacokinetics or pharmacodynamics (PK/PD) as compared to the therapeutic alone.

6. The method of claim 1 , wherein the composition has reduced toxicokinetics as compared to the therapeutic alone.

7. The method of claim 1 , wherein the administration is non-parenteral and the therapeutic is a parenteral therapeutic.

8. The method of claim 1 , wherein the composition increases efficacy and/or decreases toxicity of the therapeutic as compared to the therapeutic alone.

9. The method of claim 1 , wherein the hydrophobic amino acid is selected from alanine, valine, isoleucine, leucine, methionine, phenylalanine, tyrosine, tryptophan, and analogs thereof.

10. The method of claim 1 , wherein B is selected from arginine or glutamine.

11. The method of claim 1 , wherein C is arginine.

12. The method of claim 1 , wherein n is 2.

13. The method of claim 1 , wherein the heterologous moiety is selected from the group consisting of a small molecule, a peptide, a peptide nucleic acid (PNA), siRNA, DNA, modified RNA, and RNA.

The method of claim 1 , wherein the heterologous moiety possesses at least one effector activity selected from the group consisting of modulates a biological activity, binds a regulatory protein, modulates enzymatic activity, modulates substrate binding, modulates receptor activation, modulates protein stability/degradation, and modulates transcript stability/degradation.

15. The method of claim 1 , wherein the heterologous moiety:

i) possesses at least one targeted function selected from the group consisting of modulates a function, modulates a molecule, and is localized to a specific location;

ii) is or comprises a tag or label;

iii) is selected from the group consisting of an epigenetic modifying agent, epigenetic enzyme, a bicyclic peptide, a transcription factor, a DNA or protein modification enzyme, a DNA-intercalating agent, an efflux pump inhibitor, a nuclear receptor activator or inhibitor, a proteasome inhibitor, a competitive inhibitor for an enzyme, a protein synthesis inhibitor, a nuclease, a protein fragment or domain, a tag or marker, an antigen, an antibody or antibody fragment, a ligand or a receptor, a synthetic or analog peptide from a naturally-bioactive peptide, an anti-microbial peptide, a pore-forming peptide, a targeting or cytotoxic peptide, a degradation or self-destruction peptide, a CRISPR component system or component thereof, DNA, RNA, artificial nucleic acids, a nanoparticle, an oligonucleotide aptamer, a peptide aptamer, and an agent with poor pharmacokinetics or pharmacodynamics (PK/PD); or

iv) is linked, via a linker or directly, to the polypeptide on an N terminus, a C terminus, both termini, or one or more amino acids of the polypeptide.

16. The method of claim 1 , wherein the polypeptide comprises at least two ABX″C sequences.

17. The method of claim 1 , wherein the polypeptide has a size in the range of about 5 to about 50 amino acid units in length.

18. The method of claim 1 , wherein the nucleic acid side chain is independently selected from the group consisting of a purine side chain, a pyrimidine side chain, and a nucleic acid analog side chain.

19. The method of claim 1 , wherein the nucleic acid side chain hybridizes to the heterologous moiety, wherein the heterologous moiety comprises a nucleic acid side chain.
