Abstract

The present invention relates to amino acid sequences that can bind to serum albumin. In particular, the present invention relates to immunoglobulin single variable domains, and in particular heavy-chain immunoglobulin single variable domains, that can bind to serum albumin. The invention also relates to proteins, polypeptides and other constructs, compounds, molecules or chemical entities that comprise at least one of the immunoglobulin single variable domains binding to serum albumin that are described herein.

Claims (20)

1 . An immunoglobulin single variable domain (ISVD) that is capable of binding to serum albumin and has the structure FR1-CDR1-FR2-CDR2-FR3-CDR3-FR4 in which FR1 to FR4 refer to framework regions and CDR1 to CDR3 refer to complementarity determining regions, wherein; CDR1 has the amino acid sequence of the CDR1 of an ISVD comprising the amino acid sequence of SEQ ID NO: 18; CDR2 has the amino acid sequence of the CDR2 of an ISVD comprising the amino acid sequence of SEQ ID NO: 18; and CDR3 has the amino acid sequence of the CDR3 of an ISVD comprising the amino acid sequence of SEQ ID NO: 18, wherein the CDRs are numbered according to the same antibody numbering system.

2 . The ISVD of claim 1 , wherein the CDRs are numbered according to Kabat.

3 . The ISVD of claim 1 , wherein the CDRs are numbered according to Abm.

4 . The ISVD of claim 1 , wherein the CDRs are numbered according to Chothia.

5 . The ISVD of claim 1 , wherein the CDRs are numbered according to Contact.

6 . The ISVD according to claim 1 , that has a degree of sequence identity with the sequence of SEQ ID NO: 18 of at least 85%, at least 90%, or at least 95%, wherein the CDRs are not taken into account for determining the degree of sequence identity or the amino acid differences.

7 . A polypeptide that comprises at least one ISVD according to claim 1 .

8 . The ISVD according to claim 1 , wherein the CDRs are numbered according to IMGT.

9 . The ISVD according to claim 1 , that has no more than 7, no more than 5, no more than 3, no more than 2, or no more than 1 amino acid differences with the sequence of SEQ ID NO: 18, wherein the CDRs are not taken into account for determining the degree of sequence identity or the amino acid differences.

10 . The ISVD according to claim 1 , that has a degree of sequence identity with the sequence of SEQ ID NO: 19 of at least 85%, at least 90%, or at least 95%, wherein the CDRs are not taken into account for determining the degree of sequence identity or the amino acid differences.

11 . The ISVD according to claim 1 , that has no more than 7, no more than 5, no more than 3, no more than 2, or no more than 1 amino acid differences with the sequence of SEQ ID NO: 19, wherein the CDRs are not taken into account for determining the degree of sequence identity or the amino acid differences.

12 . The ISVD according to claim 1 , that is a VHH, a humanized VHH or a camelized VH, optionally wherein the camelized VH is a camelized human VH.

13 . The ISVD according to claim 1 , that is a VHH and that contains, compared to the sequence of SEQ ID NO:18, one or more humanizing substitutions.

14 . The polypeptide according to claim 7 , that further comprises at least one therapeutic moiety or entity.

15 . The polypeptide according to claim 7 , wherein the polypeptide comprises, at its N-terminal end, an ISVD having a D at position 1.

16 . A pharmaceutical composition comprising a polypeptide according to claim 7 .

17 . A nucleic acid that encodes an ISVD according to claim 1 or a polypeptide comprising the same.

18 . An expression vector comprising the nucleic acid of claim 17 .

19 . A host or host cell that comprises the expression vector of claim 18 .

20 . A method for preparing a polypeptide comprising cultivating or maintaining a host cell according to claim 19 under conditions such that said host cell produces or expresses the polypeptide, and optionally further comprising isolating the polypeptide.
