Abstract

The present disclosure provides systems and methods for making a hydrogel comprising a cell, cell nucleus, or one or more components derived from a cell or cell nucleus. A method for making a hydrogel may comprise providing a cell or cell nucleus, a first polymer, wherein the first polymer comprises a plurality of first crosslink precursors, each of the plurality of first crosslink precursors comprising an azide group; providing a second polymer, wherein the second polymer comprises a plurality of second crosslink precursors, each of the plurality of second crosslink precursors comprising an alkyne group; and crosslinking the first polymer and the second polymer via a reaction between a first section of the first crosslink precursors and a second section of the second crosslink precursors, thereby providing the hydrogel comprising the cell or cell nucleus.

Claims (21)

1. A composition comprising:

(a) cell or cell nucleus; and

(b) a gel of two or more crosslinked polymers, wherein the two or more crosslinked polymers comprise a copper-chelating group and are configured to form crosslinks distributed throughout the gel and encapsulate the cell or cell nucleus.

2. The composition of claim 1 , wherein the crosslinks comprise a picolyl moiety.

3. The composition of claim 1 , wherein the copper-chelating group is a picolyl group.

4. The composition of claim 3 , wherein the picolyl group is an azido-picolyl group.

5. The composition of claim 1 , wherein the composition comprises a copper concentration from 0.3 mM or less.

The composition of claim 1 , wherein a polymer of the two or more crosslinked polymers comprises at least one member selected from the group consisting of a polyolefin, an olefin copolymer, an acrylic, a vinyl polymer, a polyester, a polyamide, a polyimide, a formaldehyde resin, a polyurethane, an ether polymer, a cellulosic, a thermoplastic elastomer, and a thermoplastic polyurethane.

7. The composition of claim 1 , wherein the gel is a hydrogel.

8. The composition of claim 1 , wherein a polymer of the two or more crosslinked polymers is a polyacrylamide.

9. The composition of claim 1 , further comprising at least one reagent attached by a triazole group to at least one of the two or more crosslinked polymers.

10. The composition of claim 9 , wherein the at least one reagent is an oligonucleotide.

11. The composition of claim 10 , wherein the oligonucleotide comprises a poly-T sequence.

12. The composition of claim 1 , wherein the crosslinks comprise a labile bond.

13. The composition of claim 12 , wherein the labile bond is selected from a disulfide bond, a carbamate bond, a peptide bond, or a combination thereof.

A method of forming a gel comprising: combining in a partition under copper-catalyzed click chemistry reaction conditions (i) a cell or cell nucleus, and (ii) two or more polymers configured to form crosslinks distributed throughout the gel and encapsulate the cell or cell nucleus, wherein at least one polymer comprises a copper-chelating group.

15. The method of claim 14 , wherein the copper-chelating group is a picolyl moiety.

16. The method of claim 14 , wherein the copper-catalyzed click chemistry reaction conditions comprise a copper concentration of about 0.3 mM or less.

17. The method of claim 14 , wherein the crosslinks formed comprise a labile bond selected from a disulfide bond, a carbamate bond, a peptide bond, or a combination thereof.

18. The method of claim 14 , wherein the partition is selected from a well and a discrete droplet in an emulsion.

19. The method of claim 14 , wherein a polymer of said two or more polymers comprises at least one member selected from the group consisting of a polyolefin, an olefin copolymer, an acrylic, a vinyl polymer, a polyester, a polyamide, a polyimide, a formaldehyde resin, a polyurethane, an ether polymer, a cellulosic, a thermoplastic elastomer, and a thermoplastic polyurethane.
