Abstract

Anti-TIGIT antibodies and antigen binding fragments thereof that inhibit TIGIT-mediated signalling are provided, together with combinations comprising said antibodies or antigen binding fragments thereof and methods for their use.

Claims (15)

20 . An isolated polynucleotide or combination of isolated polynucleotides encoding an antibody or antigen binding fragment which binds to human TIGIT, wherein the antibody or antigen binding fragment comprises a heavy chain variable domain (VH) comprising a HCDR1, a HCDR2, and a HCDR3, a light chain variable domain (VL) comprising a LCDR1, a LCDR2, and LCDR3, wherein:

the HCDR1 comprises SEQ ID NO: 16, the HCDR2 comprises SEQ ID NO: 17, and the HCDR3 comprises SEQ ID NO: 18; and

the LCDR1 comprises SEQ ID NO: 61, the LCDR2 comprises SEQ ID NO: 62, and the LCDR3 comprises SEQ ID NO: 63.

21 . An isolated polynucleotide encoding a VH domain and/or a VL domain of an antibody or antigen binding fragment which binds to human TIGIT, wherein the isolated polynucleotide comprises a sequence at least 90% identical to SEQ ID NO: 251 and/or a sequence at least 90% identical to SEQ ID NO: 252.

22 . The isolated polynucleotide or combination of isolated polynucleotides of claim 20 , wherein the isolated polynucleotide or combination of isolated polynucleotides comprises SEQ ID NO: 251 and SEQ ID NO: 252.

23 . An expression vector comprising the polynucleotide or combination of polynucleotides of claim 22 operably linked to regulatory sequences which permit expression of the antibody or antigen binding fragment in a host cell or cell-free expression system.

24 . A host cell or cell-free expression system containing the expression vector of claim 23 .

25 . A method of producing a recombinant antibody or antigen binding fragment thereof which comprises culturing the host cell or cell free expression system of claim 24 under conditions which permit expression of the antibody or antigen binding fragment and recovering the expressed antibody or antigen binding fragment.

53 . The isolated polynucleotide or combination of isolated polynucleotides of claim 20 , wherein the VH comprises SEQ ID NO: 221 or an amino acid sequence at least 95% identical thereto and the VH comprises SEQ ID NO: 222 or an amino acid sequence at least 95% identical thereto.

54 . The isolated polynucleotide of claim 21 , wherein the isolated polynucleotide comprises a sequence according to SEQ ID NO: 251 and/or a sequence according to SEQ ID NO: 252.

55 . The isolated polynucleotide of claim 21 , wherein the isolated polynucleotide comprises a sequence according to SEQ ID NO: 251 and a sequence according to SEQ ID NO: 252.

56 . The isolated polynucleotide of claim 55 , wherein the sequences are contiguous.

57 . The isolated polynucleotide of claim 55 , wherein the sequences are not contiguous.
