Method for Evaluating Sample Containing Cell Support-Derived Components
Provided is a method for evaluating a sample, the method including using a mixture containing a sample including at least one member selected from the group consisting of cell support-derived components, a cell suspension containing cells, an evaluation sample obtained from a cell suspension, a sample containing a liquid and microcarriers for use in cell culture, and a sample containing a liquid obtained following treatment of microcarriers, together with at least one substance selected from the group consisting of an aromatic compound having at least one functional group selected from the group consisting of a hydroxyl group, an amino group, a nitro group and a carbonyl group, and a fluorescent dye.
1 . A method for evaluating a sample, the method comprising: evaluating a sample using a mixture containing
a sample comprising at least one member selected from the group consisting of cell support-derived components, a cell suspension containing cells, an evaluation sample obtained from a cell suspension, a sample containing a liquid and microcarriers for use in cell culture, and a sample containing a liquid obtained following treatment of microcarriers, and
at least one substance selected from the group consisting of an aromatic compound having at least one functional group selected from the group consisting of a hydroxyl group, an amino group, a nitro group and a carbonyl group, and a fluorescent dye.

The method according to claim 1 , wherein the method comprises measuring cell support-derived components using a mixture containing polysaccharide cell support-derived components, an acid, and an aromatic compound having at least one functional group selected from the group consisting of a hydroxyl group, an amino group, a nitro group and a carbonyl group.
3 . The method according to claim 1 , wherein the method is a method for evaluating a cell suspension containing cells that are cultured in the presence of a dissolvable cell support and collected following dissolution of the cell support, and
the method comprises evaluating the cell suspension using a mixture containing an evaluation sample obtained from the cell suspension, an acid, and an aromatic compound having at least one functional group selected from the group consisting of a hydroxyl group, an amino group, a nitro group and a carbonyl group.
4 . The method according to claim 2 , wherein the method comprises measuring cell support-derived components, and
the method comprises heating a mixture containing polysaccharide cell support-derived components, an acid, and an aromatic compound having at least one functional group selected from the group consisting of a hydroxyl group, an amino group, a nitro group and a carbonyl group, and
subjecting a reaction product in the mixture following heating to fluorescence analysis or absorption analysis.
5 . The method according to claim 2 , wherein the method comprises measuring cell support-derived components,
the method comprises heating a mixture containing polysaccharide cell support-derived components, an acid, and an aromatic compound having at least one functional group selected from the group consisting of a hydroxyl group, an amino group, a nitro group and a carbonyl group, and
quantifying a reaction product in a mixture following heating, and
within the mixture, a mass of the aromatic compound is at least 200 times a mass of the polysaccharide cell support-derived components.
6 . The method according to claim 5 , wherein fluorescence analysis or absorption analysis is used for quantifying the reaction product.
7 . The method according to claim 4 , wherein a concentration of the aromatic compound in the mixture is from 1 mg/mL to 100 mg/mL.
8 . The method according to claim 3 , wherein the method is a method for evaluating a cell suspension containing cells that are cultured in presence of a dissolvable cell support and collected following dissolution of the cell support, and
the method comprises providing an evaluation sample obtained from the cell suspension,
preparing a mixture containing the evaluation sample, an acid, and an aromatic compound having at least one functional group selected from the group consisting of a hydroxyl group, an amino group, a nitro group and a carbonyl group,
heating the mixture, and
subjecting a reaction product in the mixture following heating to fluorescence analysis or absorption analysis.
9 . The method according to claim 3 , wherein the method is a method for evaluating a cell suspension containing cells that are cultured in presence of a dissolvable cell support and collected following dissolution of the cell support,
the method comprises providing an evaluation sample obtained from the cell suspension,
preparing a mixture containing the evaluation sample, an acid, and an aromatic compound having at least one functional group selected from the group consisting of a hydroxyl group, an amino group, a nitro group and a carbonyl group,
heating the mixture, and
quantifying a reaction product in the mixture following heating, and
a mass of the aromatic compound is at least 200 times a mass of saccharides produced by hydrolysis of the cell support.
10 . The method according to claim 9 , wherein fluorescence analysis or absorption analysis is used for quantifying the reaction product.
11 . The method according to claim 8 , wherein a concentration of the aromatic compound in the mixture is from 1 mg/mL to 100 mg/mL.
12 . The method according to claim 1 , wherein the method comprises evaluating a cell suspension, and
the method comprises adding a fluorescent dye that is capable of staining a polymer having a hydrophobic portion to a cell suspension containing cells,
irradiating the cell suspension containing the added fluorescent dye with an excitation light and performing fluorescence analysis, and
evaluating the cell suspension based on the fluorescence analysis.
13 . The method according to claim 1 , wherein the method comprises evaluating microcarriers, and
the method comprises providing a sample which either contains a liquid and microcarriers for use in cell culture, or contains a liquid obtained following treatment of microcarriers,
adding a fluorescent dye that is capable of staining a polymer having a hydrophobic portion to the sample and performing fluorescence analysis, and
evaluating the microcarriers based on the fluorescence analysis.
14 . The method according to claim 12 , wherein the fluorescent dye comprises at least one dye selected from the group consisting of porphyrin dyes, phthalocyanine dyes, poly(phenylene vinylene) dyes, pyrene dyes, xanthene dyes, coumarin dyes and DCM dyes.
15 . A cell suspension, comprising: cells, wherein a number of polymers having a hydrophobic portion and a size of 1 μm or smaller is 10 or fewer per 1×10 4 cells.

A method of measuring cell support-derived components in a sample, comprising detecting the cell support-derived components in the sample using a mixture containing polysaccharide cell support-derived components, an acid, and an aromatic compound having at least one functional group selected from the group consisting of a hydroxyl group, an amino group, a nitro group and a carbonyl group, or detecting the cell support-derived components in the sample using a fluorescent dye that is capable of staining a polymer having a hydrophobic portion to the sample.
