Abstract

Disclosed is an advancement in provoked chemical cleavage. Thereby the invention provides the use of a diene as a chemically cleavable group attached to a Construct, and the use of a dienophile to provoke the release of the Construct by allowing the diene to react with a dienophile capable of undergoing an inverse electron demand Diels Alder reaction with the diene. The invention includes a kit for releasing a Construct C A bound to a Trigger T R , the kit having a tetrazine and a dienophile, wherein the Trigger is the tetrazine. The invention also includes the use of the formation of a pyridazine by reacting a tetrazine having a Construct C A bound thereto and a dienophile, as a chemical tool for the release, in a chemical, biological or physiological environment, of the Construct.

Claims (58)

1. A tetrazine according to formula (1), or a salt thereof; wherein Formula (1) is:

A T , B T , C T , D T each independently are selected from the group consisting of CR 2 2 , C═O, C═CR 3 2 , C—NR 6 , NR 4 , O, S, S(═O), or S(═O) 2 , provided that no sets consisting of adjacent atoms are present selected from the group consisting of —O—O—,—S—N(R 4 )—,—O—S—,—O—S(═O)—,—O—S(═O) 2 —, and —S—S—, and wherein one R 2 or R 4 group from B T and one R 2 or R 4 group from A T or C T can together be a bond, so as to form a double bond, and wherein two or more groups selected from R 2 , R 3 , R 4 , R 5 , R 6 can together form a C 6 -C 12 aryl, or a 3- to 8-membered heterocycle, a 3- to 8-membered annelated hetero(alkyl), a 3- to 8-membered spiro carbocycle or a 3- to 8-membered spiro heterocycle; X C is O, S, CR 5 2 or NR 6 ; L C is a self-immolative linker with f being 0 or 1; C A is Construct A; Y C1 is O, S or a secondary amine or a tertiary amine, wherein these moieties are part of L C ; Y C2 is O, S or a secondary amine or a tertiary amine, wherein these moieties are part of C A , wherein f is 0 and g is 1, or wherein fis 1 and g is an integer from 1 to 9; wherein b, c, d and e are independently 0 or 1, provided that if b is 0 then c and d are 0, and if c is 0 then dis 0, and provided that if d is 1 then b and c are 1, and that if c is 1 then b is 1, and provided that if c is 0 then e is 1; wherein when f is 1 then Y C1 is referred to as Y C , and when f is 0, then Y C2 is referred to as Y C ;

wherein when b, e, and g are 1, c, d, and f are 0, Y C is O or a secondary anime, and A T and B T together form —CH 2 —CH 2 -, then X C is S, CR 5 2 or NR 6 ;

wherein each R 1 , R 2 , R 3 and R 5 is independently selected from the group consisting of H, (hetero)alkyl, aryl, heterocycle, carbocycle, F, Cl, Br, I, CF 3 , CF 2 —R′, NO 2 , N 3 , OR′, SR′, CN, C(═O)R′, S(═O)R′, S(═O) 2 R″, S(═O) 2 OR′, OS(═O) 2 R″, PO 3 R′ 2 , Si—R″ 3 , Si—O—R″ 3 , B(OR′) 2 , S(═O) 2 NR′ 2 , NR'S(═O) 2 R″, C(═O)NR′ 2 , C(═S)NR′ 2 , C(═NR′)NR′ 2 , NR′ 2 ,

NR′C(═O)R′, NR′C(═S)R′, NR′C(═O)NR′ 2 , NR′C(═S)NR′ 2 wherein each R′ is independently selected from the group consisting of H, (hetero)alkyl, aryl, carbocycle, heterocycle, and each R″ is independently selected from the group consisting of (hetero)alkyl, aryl, carbocycle, heterocycle, wherein all (hetero)alkyl, aryl, carbocycle, heterocycle moieties comprised in R 1 , R 2 , R 3 and R 5 can be unsubstituted or substituted, and wherein two R′ or R″ groups can together form a ring;

each R 4 and R 6 is independently selected from the group consisting of H, (hetero)alkyl, aryl, heterocycle, carbocycle, OR′, C(═O)R′, C(═O)NR′ 2 , C(═S)NR′ 2 , C(═NR′)NR′ 2 , NR′ 2 , such that no sets consisting of adjacent atoms are present selected from the group consisting of —N—N—,—N—O—,—N—S—,—N—Si-and-N—P—, and wherein each R′ is independently selected from the group consisting of (hetero)alkyl, aryl, carbocycle, heterocycle, wherein all (hetero)alkyl, aryl, carbocycle, heterocycle moieties comprised in R 4 and R 6 can be unsubstituted or substituted, and

wherein two R′ groups can together form a ring;

wherein in all of the above embodiments, one or more of R 1 , R 2 , R 3 , R 4 , R 5 , R 6 and the self-immolative linker LC, may be bound to an S P , C B , and/or S P -C B ;

wherein S P is a spacer;

wherein C B is Construct B;

wherein Construct A is selected from the group consisting of small molecules, organic molecules, metal coordination compounds, inorganic molecules, organometallic molecules, biomolecules, drugs, polymers, resins, particles, gels, surfaces, cells, biological tissues, pathogens, solid supports, and combinations thereof; and

wherein each Construct B is independently selected from the group consisting of small molecules, organic molecules, metal coordination compounds, inorganic molecules, organometallic molecules, biomolecules, drugs, polymers, resins, particles, gels, surfaces, cells, biological tissues, pathogens, solid supports, a radioisotope, and combinations thereof.

2. A tetrazine according to claim 1 , or a salt thereof; wherein Y T is R 1 , A T is CHR 2 , B T is CR 2 2 , X C is NR 6 , Y C is O or S attached to an aromatic carbon atom of L C or CA, satisfying at least one of the following requirements:

a) c is 0, b and e is 1, R 6 is H;

b) c is 0, b and e is 1, R 6 is (hetero)alkyl, aryl, heterocycle, or carbocycle;

c) b is 0, e is 1, R 6 is H, (hetero)alkyl, aryl, heterocycle, or carbocycle; and

d) b is 0, e is 1, R 6 is C 1 -C 3 alkyl.

3. A tetrazine according to claim 1 , or a salt thereof; wherein Y T is R 1 , X C is O, Y C is secondary or tertiary amine, b is 0, e is 1, satisfying at least one of the following requirements:

a) Y C is a secondary amine, A T is CHR 2 with R 2 being H, (hetero)alkyl, aryl, heterocycle, or carbocycle;

b) Y C is a secondary amine, A T is CHR 2 with R 2 being C 1 -C 3 alkyl;

c) Y C is a tertiary amine, A T is CHR 2 with R 2 being H, (hetero)alkyl, aryl, heterocycle, or carbocycle; and

d) Y C is a tertiary amine, A T is CHR 2 with R 2 being C 1 -C 3 alkyl.

4. A tetrazine according to claim 1 , or a salt, solvate, or hydrate thereof; wherein the X T satisfies one of the following structures, wherein Y C is O or S, wherein the O or S may be attached to an aromatic carbon atom of L C or C A

wherein U, V, W, Z are each independently chosen from the group consisting of N and CR 7 ; X is independently chosen from the group consisting of O, S and NR 6 ; E is independently chosen from the group consisting of O, S, NR 6 and CR 7 2 ; with R 7 , R 8 and R 9 each independently being selected from the group defined in claim 1 for R 1 , R 2 , R 3 , R 5 , wherein two R 6 , R 7 , R 8 and/or R 9 can together form a C 6 -C 12 aryl, or a 3- to 8-membered heterocycle, a 3- to 8-membered annelated hetero(alkyl), a 3- to 8-membered spiro carbocycle or a 3- to 8-membered spiro heterocycle; and wherein one or more of R 6 , R 7 , R 8 , R 9 , and L C may be bound to a Spacer S P , C B , and/or S P -C B .

5. A tetrazine according to claim 1 , or a salt thereof; wherein the X T satisfies one of the following structures:

wherein Y C is secondary or tertiary amine;

wherein U, V, W, Z are each independently chosen from the group consisting of N and CR 7 ; X is independently chosen from the group consisting of O, S and NR 6 ; E is independently chosen from the group consisting of O, S, NR 6 and CR 7 2 ; with R 7 , R 8 and R 9 each independently being selected from the group defined in claim 1 for R 1 , R 2 , R 3 , R 5 , wherein two R 6 , R 7 , R 8 and/or R 9 can together form a C 6 -C 12 aryl, or a 3- to 8-membered heterocycle, a 3- to 8-membered annelated hetero(alkyl), a 3- to 8-membered spiro carbocycle or a 3- to 8-membered spiro heterocycle; and wherein one or more of R 6 , R 7 , R 8 , R 9 , and L C may be bound to a Spacer S P , C B , and/or S P -C B .

6. A tetrazine according to claim 1 , or a salt thereof; wherein R 1 is (hetero)alkyl, aryl, heterocycle or carbocycle.

A tetrazine according to claim 1 , or a salt thereof; wherein one C A is bound to more than one T R or L C -T R moieties, wherein the T R and/or L C moieties may be bound to one or more S P , C B and/or S P -C B moieties, wherein C A is D D , C B is T T , M M or a Binding Partner for M M ; wherein D D is a drug, T T is a targeting agent, and M M is a masking moiety.

A tetrazine according to claim 1 , or a salt thereof; wherein C A and C B are each independently selected from the group consisting of carbohydrates, peptides, peptoids, lipids, proteins, enzymes, oligonucleotides, DNA, RNA, PNA, LNA, aptamers, hormones, toxins, steroids, cytokines, antibodies, antibody fragments, diabodies, triabodies, antibody fusions, antibody fragment fusions, and combinations thereof.

9. A tetrazine according to claim 1 , or a salt thereof; wherein either or both of the Constructs C A and C B is a biomolecule.

10. A tetrazine according to claim 1 , or a salt thereof; wherein the Construct C A or C B is a polymer, resin, particle, solid support, gel, or surface.

11. A tetrazine according to claim 1 , or a salt thereof; wherein C A is or is comprised in a binding to a polymer, resin, particle, solid support, gel, surface, and wherein C B comprises a radioisotope.

12. A tetrazine according to claim 1 , or a salt thereof; wherein the Construct A comprises one or more chemical reactants, the reactivity of which is masked by being linked to the diene.

13. A tetrazine according to claim 1 , or a salt thereof; wherein g is an integer from 1 to 3.

14. A tetrazine according to claim 1 , or a salt thereof; wherein g is 1.

15. A tetrazine according to claim 1 , or a salt thereof; wherein one or more of R 1 , R 2 , R 3 , R 4 , R 5 , R 6 and the self-immolative linker L C , is bound to an S P , C B , and/or S P -C B .

A Construct-Trigger conjugate or a salt thereof; wherein the Construct-Trigger conjugate comprises a Construct A linked, directly or indirectly, to a tetrazine in such a way that the Construct A is released following an inverse-electron demand Diels-Alder reaction of the tetrazine with a dienophile.

A tetrazine according to claim 1 , or a salt thereof; wherein one or more R a,b,c moieties is bound to one or more of the group consisting of Targeting agent T T , Spacer S P , chelator, albumin-binding moiety, polymersome, micelle, particle, resin, gel, surface, bead, membrane translocation moiety, radioisotope, and fluoresecent moiety.

18. A tetrazine according to claim 1 , or a salt thereof; wherein Construct A is a drug.

19. A tetrazine according to claim 1 , or a salt thereof; wherein Construct A is a protein, a toxin, a chelator, monomethyl auristatin E, or doxorubicin.

20. A tetrazine according to claim 5 , or a salt thereof; wherein X T is:

wherein R 8 is methyl; R 9 is hydrogen; Y C is a tertiary amine; and Y T is phenyl, alkyl, or X T ;

wherein the phenyl is optionally bound to a C B , and/or S P -C B , wherein S P is (CH 2 ) r C(O)NR 4 (CH 2 ) r , (CH 2 CH 2 O) r , (CH 2 CH 2 O) r CH 2 , (CH 2 )C(O)NR 4 (CH 2 CH 2 O) r , (CH 2 ) r C(O)NR 4 (CH 2 CH 2 O) r CH 2 , (CH 2 CH 2 O) r C(O)NR 4 (CH 2 CH 2 O) r , (CH 2 CH 2 O) r , C(O)NR 4 (CH 2 CH 2 O) r CH 2 , (CH 2 CH 2 O)C(O)NR 4 CH 2 , or alkenylene; wherein r is independently an integer from 1-10, and R 4 is H or unsubstituted C 1 -C 3 alkyl; and C B is an antibody.

21. A tetrazine according to claim 1 , or a salt thereof; wherein A T is-CH 2 —; b, c, d, and e are 0; Y C is an oxygen atom attached to an aromatic carbon atom of L C or C A ; and Y T is alkyl, substituted phenyl, or unsubstituted phenyl.

22. A tetrazine according to claim 20 , or a salt thereof; wherein Y C is —NR C —, and wherein R C is methyl or isopropyl.

23. A tetrazine according to claim 22 , or a salt thereof; wherein f is 0, g is 1, and Y C is Y C2 .

24. A tetrazine according to claim 23 , or a salt thereof, wherein C A is monomethyl auristatin E.

25. A tetrazine according to claim 24 , or a salt thereof; wherein Y T is X T .

26. A tetrazine according to claim 24 , or a salt thereof, wherein Y T is methyl or isopropyl.

27. A tetrazine according to claim 20 , or a salt thereof; wherein C B is CC49, AVP0450, or AVP0458.

28. A tetrazine according to claim 22 , or a salt thereof, wherein C B is CC49, AVP0450, or AVP0458.

29. A tetrazine according to claim 23 , or a salt thereof, wherein C B is CC49, AVP0450, or AVP0458.

30. A tetrazine according to claim 24 , or a salt thereof; wherein C B is CC49, AVP0450, or AVP0458.

31. A tetrazine according to claim 25 , or a salt thereof; wherein C B is CC49, AVP0450, or AVP0458.
