Abstract

A parallel assembly of chromatography column modules connected in a rigid housing the assembly having one common assembly inlet and one common assembly outlet each column module comprising a bed space filled with chromatography medium and each column module comprises integrated fluid conduits which when the column module is connected with other column modules in the rigid housing are adapted to connect the bed space of the column module with the assembly inlet and the assembly outlet wherein the total length and/or volume of the fluid conduit from the assembly inlet to one bed space together with the length and/or volume of the fluid conduit from the same bed space to the assembly outlet is substantially the same for all bed spaces and modules installed in the parallel assembly.

Claims (27)

1. A parallel assembly of chromatography column modules, the assembly having one common assembly inlet flow path and one common assembly outlet flow path, each chromatography column module comprising:

a bed space filled with chromatography medium; and

one or more integrated fluid conduits which, when the chromatography column module is connected with other chromatography column modules, are adapted to connect the bed space of the chromatography column module with the assembly inlet flow path and the assembly outlet flow path,

wherein each of the integrated fluid conduits comprise an integrated fluid inlet conduit forming a single inlet aperture to the bed space within a bottom surface of the bed space, and an integrated fluid outlet conduit forming a single outlet aperture from the bed space within a top surface of the bed space, and

wherein the total length and/or volume of the fluid conduit from the assembly inlet to one bed space together with the length and/or volume of the fluid conduit from the same bed space to the assembly outlet is substantially the same for all bed spaces and modules installed in the parallel assembly.

2. The parallel assembly according to claim 1 , wherein said chromatography column modules are stacked together inside a rigid housing.

3. The parallel assembly according to claim 1 , wherein a compression force is applied to the chromatography column modules to achieve and/or secure fluid tight connections within the fluid conduits of the assembly.

The parallel assembly according to claim 1 , further comprising two fluid distribution modules one arranged at each side of the stack of column modules, each fluid distribution module having one distribution module fluid connection representing the assembly inlet or assembly outlet of the parallel assembly and a connection face towards the stack of column modules, the connection face having at least two connection face fluid connections for connecting to the inlet or outlet side of at least two stacked chromatography column modules.

5. The parallel assembly according to claim 1 , wherein the chromatography column modules can be composed of at least two serially connected chromatography column submodules.

6. A chromatography column module adapted to be used in a parallel assembly according to claim 1 .

7. The chromatography column module according to claim 6 , characterised in that it is disposable.

8. The chromatography column module according to claim 6 , characterised in that end and a tube of the chromatography column modules together define the bed space and the end pieces are facing each other when stacking said chromatography column modules and are prepared by moulding techniques.

9. The chromatography column module according to claim 6 , wherein a tube of chromatography column module is manufactured with integrated fluid conduits.

10. The chromatography column module according to claim 6 , wherein sealing means are provided to fluid connections of the chromatography column modules.

11. The chromatography column module according to claim 6 , wherein it is sterilized by gamma radiation.

The chromatography column module according to claim 6 , characterised in that the bed space is prefilled with dry chromatography gel in an amount such that when liquid is added to the bed space the dry gel will swell to a volume occupied in a non confined space that is at least 2% larger than the volume of the confined bed space.

13. The chromatography column module according to claim 6 , characterised in that it comprises a sensor.

14. The chromatography column module according to claim 6 , characterised in that it comprises an adjustable flow restrictor.

15. The chromatography column module according to claim 6 , characterised in that aseptic films are provided to fluid connections between the modules.

16. The chromatography column module according to claim 6 , wherein the chromatography column module is composed of at least two serially connected chromatography column submodules.

A rigid housing adapted to hold any number of chromatography column modules according to claim 6 , in a parallel connection mode, wherein said rigid housing comprises two stiff end plates between which the chromatography column modules should be provided and said end plates are adapted to be forced towards each other in order to provide fluid tight connections between fluid connections of the chromatography column modules.

18. A method for connecting chromatography column modules according to claim 6 , said method comprising the steps of:

providing the desired number of chromatography column modules in a stack with fluid connections facing each other; and

forcing the chromatography column modules towards each other such that fluid tight connections are provided there between.

The method according to claim 18 , further comprising providing chromatography column modules comprising dry chromatography gel and, after said fluid tight connections are provided, adding liquid to the column modules such that the dry gel will swell to a volume occupied in a non confined space that is at least 2% larger than the volume of the confined bed space.

20. The method according to claim 18 , further comprising calibrating flow resistance through the different chromatography column modules by separately measuring flow resistance through each module and separately adjusting flow restrictors provided in each module.

21. The method according to claim 18 , further comprising removing aseptic films provided over the fluid connections on the chromatography column modules.
