Abstract

Processes and compositions are described for preparing new, colloidally stable, coated nanomagnetic particles useful for both in-vitro and in-vivo biomedical applications, including cell targeting and capturing cells, microorganisms, and cellular organelles or entities such as exosomes. These nanomagnetic particles can also be used as imaging contrast agents due to their small size and high magnetic moment. The nanomagnetic particles include a series of sequentially added, stabilizing surface coatings rendered onto nano-sized magnetic crystal clusters (e.g., magnetite particles) to impart colloidal stability in complex biological samples with minimal leaching of the coating materials, high binding capacity, and low non-specific binding. Another benefit of this invention is the ability to utilize both external and internal magnetic field-generating separation devices to effect separation of the magnetic nanoparticles.

Claims (27)

1 . A method of using biomolecule/particle complexes, comprising:

(a) contacting a biological sample known or suspected to contain a biomolecule of interest with targeted nanomagnetic particles, wherein each targeted nanomagnetic particle comprises:

(i) a magnetic core particle;

(ii) a glass layer encapsulating the magnetic core particle;

(iii) a protein and polymer composite layer bound to the glass layer, wherein the composite layer comprises albumin and dextran; and

(iv) a targeting moiety that comprises one member of a bioaffinity ligand pair covalently bound to the protein and polymer composite layer, wherein the one member of the bioaffinity ligand pair specifically binds to the biomolecule of interest to form at least one biomolecule/particle complex; and

(b) separating the biomolecule/particle complex from the biological sample using a magnetic field.

2 . The method according to claim 1 , wherein using a magnetic field comprises using a magnetic column separation or a high gradient magnetic separation (HGMS) method to apply a magnetic field.

3 . A method of separating a target biomolecule species from a reaction mixture, the method comprising:

(a) in a reaction mixture, contacting a biological sample known or suspected to contain a biomolecule of interest, with targeted nanomagnetic particles, wherein each targeted nanomagnetic particle comprises:

(i) a magnetic core particle;

(ii) a glass layer encapsulating the magnetic core particle;

(iii) a protein and polymer composite layer bound to the glass layer, wherein the composite layer comprises albumin and dextran; and

(iv) a targeting moiety that comprises one member of a bioaffinity ligand pair covalently bound to the protein/polymer composite layer, wherein the one member of the bioaffinity ligand pair specifically binds to the biomolecule of interest to form at least one biomolecule/particle complex; and

(b) eluting the biomolecule/particle complexes from the reaction mixture using a magnetic field.

4 . A method of separating a plurality of target biomolecule species from a biological sample, the method comprising:

(a) in a reaction mixture, contacting a biological sample known or suspected to contain a first and a second biomolecule species of interest with targeted nanomagnetic particles and targeted buoyant microparticles wherein each targeted nanomagnetic particle comprises:

(i) a magnetic core particle;

(ii) a glass layer encapsulating the magnetic core particle;

(iii) a protein and polymer composite layer bound to the glass layer, wherein the composite layer comprises albumin and dextran; and

(iv) a targeting moiety that comprises one member of a bioaffinity ligand pair covalently bound to the protein/polymer composite layer, that targets the first biomolecule species of interest to form at least one first target biomolecule/particle complex, wherein the targeted buoyant microparticles target the second biomolecule species of interest to form at least one second target biomolecule/particle complex; and wherein the targeted buoyant microparticles comprise a bioaffinity ligand that targets the second biomolecule species of interest to form at least one second biomolecule/particle complex;

(b) separating the first biomolecule/particle complexes from the reaction mixture using a magnetic field; and

(c) eluting the second target biomolecule/particle complexes from the reaction mixture.

5 . The method according to claim 4 , wherein the targeted nanomagnetic particles have a diameter ranging from about 5 nm to about 500 nm, preferably from about 30 nm to about 300 nm.

6 . The method according to claim 4 , wherein the magnetic core particles of the targeted nanomagnetic particles comprise magnetite (Fe 3 O 4 ) crystals, optionally wherein the magnetite crystals have a diameter ranging from about 5 nm to about 300 nm.

7 . A kit comprising a composition comprising targeted nanomagnetic particles and targeted buoyant microparticles according to claim 4 , in a container and optionally including instructions for use of the composition.

8 . The method of claim 3 , wherein using the magnetic field includes changing or removing the magnetic field.
