Abstract

Beta-hydroxybutyric acid compositions for oral delivery are substantially free of beta-hydroxybutyrate salts and are effective in rapidly raising blood ketone levels without causing acute acidosis or gastrointestinal (GI) distress when consumed in sufficiently dilute form and/or as a gel or suspension. By excluding beta-hydroxybutyrate salts (e.g., less than 1% by weight) containing alkali or alkaline earth metal ions, beta-hydroxybutyric acid solutions, gels, or suspensions can deliver exogenous ketone bodies without significantly altering electrolyte balance. Although aqueous beta-hydroxybutyric acid solutions are moderately acidic with a pH of about 3.5 to 4, when diluted with sufficient water, the water acts as a pseudo buffering agent that offsets otherwise harsh acidic effects when consumed orally. Gels and suspensions can also ameliorate acidic effects by partially encapsulating the beta-hydroxybutyric acid. Beta-hydroxybutyric acid can be enriched with the R- or the S-enantiomer, a racemic mixture, pure R-beta-hydroxybutyric acid, or pure S-beta-hydroxybutyric acid.

Claims (39)

1 . A solid beta-hydroxybutyrate composition for oral delivery to increase blood ketone level in a subject, comprising:

beta-hydroxybutyric acid; and

a beta-hydroxybutyrate salt;

wherein the composition contains a higher concentration of the beta-hydroxybutyric acid than the beta-hydroxybutyrate salt by combined weight of the beta-hydroxybutyric acid and the beta-hydroxybutyrate salt.

2 . The solid beta-hydroxybutyrate composition of claim 1 , wherein the composition is a powder.

3 . The solid beta-hydroxybutyrate composition of claim 1 , wherein the composition is provided in a dosage form that provides a premeasured quantity of the solid beta-hydroxybutyrate composition.

4 . The solid beta-hydroxybutyrate composition of claim 3 , wherein the dosage form comprises a packet or pouch.

5 . The solid beta-hydroxybutyrate composition of claim 3 , wherein the dosage form comprises a tablet or capsule.

6 . The solid beta-hydroxybutyrate composition of claim 3 , wherein the dosage form comprises an effervescent tablet.

7 . The solid beta-hydroxybutyrate composition of claim 3 , wherein the dosage form provides a premeasured quantity of about 0.5 gram to about 25 grams of the solid beta-hydroxybutyrate composition.

8 . The solid beta-hydroxybutyrate composition of claim 3 , wherein the dosage form provides a premeasured quantity of about 0.75 gram to about 20 grams of the solid beta-hydroxybutyrate composition.

9 . The solid beta-hydroxybutyrate composition of claim 3 , wherein the dosage form provides a premeasured quantity of about 1 gram to about 15 grams of the solid beta-hydroxybutyrate composition.

The solid beta-hydroxybutyrate composition of claim 1 , further comprising at least one additive selected from the group consisting of flavorant, sweetener, stabilizer, vitamin, mineral, stimulant, nootropic, edible acid, vasodilator, muscle-promoting compound, silica, effervescing agent, cannabinoid, 1,3-butanediol, beta-hydroxybutyrate ester, gelling agent, and amino acid.

The solid beta-hydroxybutyrate composition of claim 1 , wherein the beta-hydroxybutyric acid is a non-racemic mixture enriched with R-beta-hydroxybutyric acid relative to S-beta-hydroxybutyric acid so as to contain a greater quantity of enantiomeric equivalents of R-beta-hydroxybutyric acid than S-beta-hydroxybutyric acid.

The solid beta-hydroxybutyrate composition of claim 1 , wherein the beta-hydroxybutyric acid is a non-racemic mixture enriched with S-beta-hydroxybutyric acid relative to R-beta-hydroxybutyric acid so as to contain a greater quantity of enantiomeric equivalents of S-beta-hydroxybutyric acid than R-beta-hydroxybutyric acid.

13 . The solid beta-hydroxybutyrate composition of claim 1 , wherein the beta-hydroxybutyric acid is a racemic mixture of R-beta-hydroxybutyric acid and S-beta-hydroxybutyric acid.

14 . The solid beta-hydroxybutyrate composition of claim 1 , wherein the beta-hydroxybutyric acid is enantiomerically pure S-beta-hydroxybutyric acid.

15 . The solid beta-hydroxybutyrate composition of claim 1 , wherein the beta-hydroxybutyric acid is enantiomerically pure R-beta-hydroxybutyric acid.

16 . A kit for administering beta-hydroxybutyrate by oral delivery to increase blood ketone level in a subject to a subject, the kit comprising:

the solid beta-hydroxybutyrate composition of claim 1 ;

a container in which the solid beta-hydroxybutyrate composition is placed, wherein the container is selected from the group consisting of carton, box, can, jar, bag, pouch, bottle, jug, and keg; and

one or more dosage forms or measuring devices in or associated with the container and configured to provide a premeasured quantity of the solid beta-hydroxybutyrate composition, wherein the one or more dosage forms or measuring devices are selected from the group consisting of a plurality of packets, a plurality of pouches, a plurality of tablets, a plurality of effervescent tablets, a plurality of capsules, a cup, a scoop, a spatula, and a spoon.

17 . A solid beta-hydroxybutyrate composition for use in making a beverage or food for oral delivery to increase blood ketone level in a subject, comprising:

beta-hydroxybutyric acid;

a beta-hydroxybutyrate salt; and

at least one additive selected from the group consisting of flavorant, sweetener, stabilizer, vitamin, mineral, stimulant, nootropic, edible acid, vasodilator, muscle-promoting compound, silica, effervescing agent, cannabinoid, 1,3-butanediol, beta-hydroxybutyrate ester, gelling agent, and amino acid,

wherein the composition contains a higher concentration of the beta-hydroxybutyric acid than the beta-hydroxybutyrate salt by combined weight of the beta-hydroxybutyric acid and the beta-hydroxybutyrate salt.

18 . A kit for administering beta-hydroxybutyrate by oral delivery to increase blood ketone level in a subject to a subject, the kit comprising:

the solid beta-hydroxybutyrate composition of claim 17 ;

a container in which the solid beta-hydroxybutyrate composition is placed, wherein the container is selected from the group consisting of carton, box, can, jar, bag, pouch, bottle, jug, and keg; and

one or more dosage forms or measuring devices in or associated with the container and configured to provide a premeasured quantity of the solid beta-hydroxybutyrate composition, wherein the one or more dosage forms or measuring devices are selected from the group consisting of a plurality of packets, a plurality of pouches, a plurality of tablets, a plurality of effervescent tablets, a plurality of capsules, a cup, a scoop, a spatula, and a spoon.

19 . A solid beta-hydroxybutyrate composition for use in making a beverage or food for oral delivery to increase blood ketone level in a subject, comprising:

beta-hydroxybutyric acid;

a beta-hydroxybutyrate salt; and

a dosage form comprising or containing therein a unit dose of about 0.5 gram to about 25 grams by combined weight of the beta-hydroxybutyric acid and the beta-hydroxybutyrate salt, wherein the dosages form is selected from a packet, a pouch, a tablet, a capsule, and an effervescent tablet,

wherein the composition contains a higher concentration of the beta-hydroxybutyric acid than the beta-hydroxybutyrate salt by combined weight of the beta-hydroxybutyric acid and the beta-hydroxybutyrate salt.

20 . A kit for administering beta-hydroxybutyrate by oral delivery to increase blood ketone level in a subject to a subject, the kit comprising:

the solid beta-hydroxybutyrate composition of claim 19 ; and

a container containing therein multiple unit doses of the solid beta-hydroxybutyrate composition, wherein each unit dose is provided by a respective dosage form.
